This thesis investigates the ultrafast loss and recovery of ferromagnetic (FM) and antiferromagnetic (AFM) order in exchange-coupled metallic FM/AFM and FM/AFM/FM heterostructures using time-resolved resonant x-ray magnetic circular (R-XMCD) and linear dichroism (R-XMLD) measurements in reflectivity geometry. The work examines how an AFM layer and its spin structure influence the ultrafast dynamics of adjacent FM layers, as well as the intrinsic dynamics of the AFM itself following ultrashort optical excitation. Tailored Co/Mn/Co trilayers and Mn2Au/Fe bilayers exhibiting specific magnetic couplings were grown using molecular beam epitaxy (MBE) and characterized structurally and magnetically using Auger electron spectroscopy (AES), low-energy electron diffraction (LEED), and magneto-optical Kerr effect (MOKE) measurements. The experimental findings of this thesis are organized in three main studies. The first investigates the ultrafast magnetization dynamics of two FM Co layers interlayer-exchange-coupled through an AFM Mn spacer layer. An accelerated demagnetization is observed in case of a collinear Mn spin structure, attributed to an enhanced optically induced intersite spin transfer (OISTR) between Co and Mn, with results qualitatively reproduced by ab initio time-dependent density-functional theory (TD-DFT) calculations. The second study focuses on the epitaxial growth of AFM Mn2Au on gold-capped Nb(001) and Ta(001) substrates. Layer-by-layer growth on the gold-capped Nb(001) substrate is observed. Magnetic coupling to an adjacent FM Fe layer reveals distinct coupling and non-coupling areas on the sample, resulting in a partial exchange-bias (EB) shift after field cooling. These regions are locally identified and quantitatively analyzed via Kerr microscopy, revealing dimensions of tens of µm. The final study examines the ultrafast loss and recovery of AFM and FM order in a strongly magnetically coupled Mn2Au/permalloy (Py) bilayer following ultrashort laser excitation. A remarkably fast recovery of AFM order, within 2-3 ps, is observed in the Mn2Au layer, occurring 1-2 orders of magnitude faster than the FM order recovery in the adjacent Py layer. So far, the observed AFM dynamics could not be fully reproduced by an atomistic spin model coupled to a two-temperature model for a simple Mn2Au layer. Absorption calculations for both pump and probe pulses reveal that only surface-near Mn2Au layers are probed, where the excitation is 2-3 times more intense than in bulk layers. These findings demonstrate that AFMs enable ultrafast spin dynamics, highlighting their potential in future THz spintronic devices.
View lessProtein-Biomarker stellen die Eckpfeiler gegenwärtiger und zukünftiger personalisierter Onkologie dar. Ihre akkurate quantitative Messung ist jedoch mit methodischen Hürden verbunden. Ausgehend von der bisherigen, quantitativ limitierten Standardtechnik Immunhistochemie wird gezeigt, dass enzymatische Deglykosylierung diese methodeninhärenten Probleme nicht mildern kann. Am Beispiel des Zielproteins eines Antikörper-Wirkstoff-Konjugats wird die quantitative Messung von Proteo- und Glykoformen eines Biomarkers durchgeführt und die Relevanz der Tumorspezifität hervorgehoben. Zur Verbesserung proteomischer Analysen klinischer Proben werden mehrere präanalytische Optimierungen vorgestellt, um auch niedrig-abundante Membranproteine als onkologische Schlüsselpopulation besser mittels LC-MS/MS messen zu können. Für die Stabilisation der Normalisierung und damit der Quantifizierung tumorspezifischer Unterschiede wird ein robuster Normalisierungsalgorithmus vorgeschlagen. Diese sowie weitere methodische Verbesserungen werden genutzt, um anhand einer Harnblasenkarzinom-Kohorte den prognostischen und prädiktiven Wert proteomischer Analysen zu zeigen. Dabei wird der phänotypisch-funktionelle Wert des Proteoms herausgestellt und die quantitative Tumorspezifität von Effektoren und (potenziellen) therapeutischen Zielen als grundlegende Ressource bestimmt.
View lessMyanmar and the Eastern Himalayan Syntaxis (EHS) represent a complex tectonic transition zone shaped by the ongoing Indo-Asian continental collision and subduction. Unlike the northward subduction of the Indian Plate beneath the Tibetan Plateau, the lithosphere subducts eastward under Myanmar, driving diverse structural evolution and deformation. This process is evidenced by intermediate-depth seismicity, the Wuntho-Popa volcanic arc, and the prominent Sagaing strike-slip fault. Despite its tectonic significance, the subsurface structure of Myanmar, particularly its northern part, remains poorly understood due to a historical lack of high-resolution geophysical data. In this study, we utilize a three-year dataset from the Myanmar Subduction-to-Collision imaging Array (MySCOLAR), a 30-station broadband network deployed by the GFZ German Research Centre for Geosciences in collaboration with the Department of Meteorology and Hydrology of Myanmar. To extend our study area, we integrated data from additional temporary and permanent networks in the surrounding areas, including the Shan Plateau and the Tengchong volcanic field. By applying surface wave tomography and receiver function analysis, we provide new constraints on the crustal and upper mantle in Northern Myanmar with shear-wave velocity (VS ) structure and lithospheric interfaces. Our VS model reveals a prominent, widespread low-velocity zone (LVZ) extending from ∼60 to 90 km depth, characterized by velocities VS < 4.0 km/s. This pronounced anomaly has a lateral extent of ∼400 km, stretching from beneath the Sagaing Fault eastward across the Shan Plateau. The LVZ is overlain by a high-velocity layer (VS ≈ 4.4 km/s) situated within the Wuntho-Popa Arc, which indicates a shallow Moho in this region. Beneath the Indo-Burma Ranges, we identify two distinct eastward-dipping positive interfaces, situated at 30–60 km depth; this layer thins to less than 10 km beneath the Tengchong Volcanic Field and extends westward toward the Red River Fault zone in the northern part of the profile. At 30–50 km depth, a low-velocity anomaly (VS ≤ 4.0 km/s) is observed beneath the Indo-Burman Ranges (IBR) and the western Central Myanmar Basin (CMB). This feature primarily sits atop the subducting slab, which itself is marked by high-velocity anomalies associated with intermediate-depth seismicity. We consider that the Indian sediment and crust have subducted down to the Burma mantle with the Indian mantle slab. Additionally, at shallow depths (10–20 km), distinct low-velocity zones (∼2.8 km/s) represent thick sedimentary sequences in the Bengal, CMB, and Assam basins. The sedimentary cover in the Bengal and CMB reaches thicknesses of ∼20 km, while the Assam basin remains thinner than ∼15 km. Our receiver function analysis indicates that the Moho depth across Northern Myanmar, encompassing the eastern back-arc basin and the Shan Plateau, ranges between 35 and 40 km but rises to 20 km under at depths of approximately 25 km and 40 km. The deeper interface extends to 80 km beneath the Central Myanmar Basin (CMB), while the shallower interface reaches 50 km depth and is overlain by a prominent negative velocity gradient at ∼15 km. Integrating these results with our VS model, we interpret these features as evidence that Bengal Basin sediments are being scraped off and accreted into a thickening wedge beneath the IBR, extending to roughly 94°E. Simultaneously, a significant volume of these sediments is subducted alongside the Indian crust to depths of 50–90 km beneath the Wuntho-Popa Arc, likely contributing to the observed volcanic magmatism. We further propose that a portion of this subducted sediment may have detached and laterally injected into the upper mantle beneath the Shan Plateau. This process could account for the extremely low-velocity zone (LVZ) between 60 and 90 km, where sediment-derived fluids or melts have thinned the overlying lithosphere and fueled magmatism in the Tengchong Volcanic Field.
View lessThe central nervous system (CNS) regulates many critical processes, including cognition, emotions, and consciousness. As part of the CNS, the brain processes and integrates information, manages thinking and memory formation, and coordinates vital homeostatic functions such as breathing and heart rate. Therefore, the brain should be tightly protected to maintain a healthy life. Together with physical protection, which is mainly provided by the skull, multi-layered selective cellular barriers, including the blood-cerebrospinal fluid (B-CSF) and blood-brain barriers, coordinate brain homeostasis by modulating CSF dynamics and preventing the entry of blood-borne harmful substances into the brain, respectively. When these barriers are dysregulated, pathological conditions may develop. For instance, imbalanced production and absorption of CSF might lead to hydrocephalus, which might be lethal if not treated. Disruptions of the BBB integrity are observed in many neurological pathological conditions such as stroke, multiple sclerosis, and Alzheimer’s disease. Therefore, identifying new modulatory players of these barriers will enhance our understanding to develop targeted therapies for the related pathological conditions. In this study, first, the role of Angiomotin (AMOT) in brain endothelial cell-cell barrier integrity is described. Second, the contribution of the pathological AMOT variant to isolated X-linked congenital hydrocephalus is molecularly characterized. AMOT is a scaffolding protein that interacts with many proteins, including apical polarity and junction-associated proteins, and participates in the modulation of the cell-cell barrier integrity. In particular, its long isoform, AMOT130, plays a role in maintaining junctional integrity in endothelial cells. In this present study, the subcellular localization of endogenous AMOT and overexpressed AMOT130 was investigated. It was found that endogenous AMOT is localized in the cytoplasm, cell membrane, and nucleus, whereas overexpressed AMOT130 is mainly localized at the apical side in brain microvascular endothelial cells. Moreover, elevated AMOT130 levels via overexpression were shown to disrupt cell-cell barrier integrity in brain endothelial cells. During the second part of the thesis, a newly identified likely pathogenic AMOT130 variant, AMOT130∆N, was characterized. It was found that due to the N-terminal 91 amino acid truncation compared to wild-type AMOT130, the stability of AMOT130∆N is increased. The mechanistic explanation for the elevated AMOT130∆N protein level is the loss of the tankyrase binding domain, which is required for RNF146-mediated degradation of AMOT130, and altered N-degron degradation signaling. Subcellular localization analysis revealed that AMOT130∆N is mainly apically localized and enriched in the plasma membrane fraction. Functional assays to analyze cell-cell barrier integrity indicated that epithelial and brain microvascular endothelial cells expressing AMOT130∆N exhibit decreased barrier integrity and increased cell-cell permeability. Taken together, these results highlight that fine-tuned regulation of AMOT130 turnover is essential for maintaining cell-cell barrier integrity in epithelial and endothelial cells. Otherwise, as we demonstrated with the AMOT130∆N variant, elevated AMOT130 protein levels can lead to pathological conditions such as congenital hydrocephalus.

View lessThis thesis investigates chemo-mechanical systems, with a focus on fluid, solid, and multiphase partial differential equation (PDE) models. Such systems arise in a wide range of natural phenomena, including geological processes that motivate several of the research questions addressed in this work. These models are inherently complex, as they typically involve the coupling of conservative and dissipative mechanisms, multiple temporal and spatial scales, and a dependence on the choice of variables and coordinate systems. To address these challenges, this thesis adopts the variational framework of GENERIC, an acronym for General Equations for Non-Equilibrium Reversible-Irreversible Coupling. Within this setting, we develop a systematic approach to transformations of variables and coordinates that preserves thermodynamic consistency while making explicit the role of energetic and dissipative structures. This perspective enables a structural interpretation of PDE models and provides a coherent methodology to relate different formulations. The main contributions of this work are as follows. First, we establish a systematic transformation theory for GENERIC formulations, distinguishing between invertible and non-invertible mappings, including changes of variables, coordinate transformations, reductions, and extensions. This strategy clarifies how energy functionals and geometric operators behave under transformations between state spaces. In particular, the Lagrangian- Eulerian correspondence is treated as a specific instance of a more general structural operation, leading to a consistent interpretation of key quantities such as the determinant of the deformation gradient as a volume density, and its relation to intensive and extensive quantities. Second, we apply this framework to reformulate and derive models in different variable settings. This includes the systematic comparison of Lagrangian and Eulerian descriptions, as well as the construction of thermodynamically consistent models for fluid-structure interaction problems, porosity waves, and rock dehydration processes. These results illustrate how seemingly distinct models can be interpreted within a common structural framework. Third, we demonstrate that the GENERIC structure provides practical advantages beyond modeling. In the context of porosity waves, it enables the design of energy-based discretization schemes that yield robust and stable numerical methods for nonlinear, coupled systems, such as the elastic-diffusive Eulerian model studied in this thesis. Lastly, for rock dehydration models, we examine physically motivated modeling assumptions and compare different analytical approaches to the well-posedness analysis, addressing the nonlinearities and degeneracies inherent in the system. Overall, this work offers a unified structural perspective on chemo-mechanical PDE systems, connecting modeling, thermodynamics, and simulations within a single coherent framework. The developed approach facilitates the comparison, transformation, and extension of models across different physical regimes and provides a foundation for future developments in both theoretical and applied settings.
View lessDas Ziel dieser kumulativen Habilitationsschrift war es, zu einem besseren Verständnis der Pathogenese der Spondyloarthritis beizutragen – zunächst klinisch über eine Analyse der Assoziation zwischen Spondyloarthritis und der akuten anterioren Uveitis, des Weiteren pathophysiologisch über die Analyse expandierter T-Zell-Klone in entzündeten Gelenken HLA-B*27 positiver Patient:innen mit Spondyloarthritis und ferner über eine Analyse verschiedener Biomarker und deren Assoziation mit Therapieansprechen, Krankheitsaktivität sowie radiografischer spinaler Progression bei Patient:innen mit axialer Spondyloarthritis. In den dieser Habilitation zugrunde liegenden Publikationen konnten wir zeigen, dass eine – in den meisten Fällen noch undiagnostizierte – Spondyloarthritis bei über der Hälfte der Patient:innen mit akuter anteriorer Uveitis vorkommt; mit Psoriasis, männlichem Geschlecht, HLA-B*27-Positivität und erhöhten Entzündungsparametern assoziiert ist; und nicht ausreichend durch bereits existierende Überweisungsstrategien erkannt wird. Zudem identifizierten wir in Einzelzellanalysen zytotoxischer T-Zellen aus der Synovialflüssigkeit klonale expandierte Populationen mit charakteristischen T-Zell-Rezeptor-Motiven bei HLA-B*27 positiven Patient:innen mit Spondyloarthritis. Transkriptomanalysen zeigten, dass expandierte Klone einen Tissue-Resident-Memory-Phänotyp aufwiesen und Gene für T-Zell-Exhaustion und Apoptose hochregulierten. Dies weist auf eine (auto-)antigen-getriebene Immunantwort hin. Biomarker, die über die routinemäßig bestimmten Entzündungsparameter hinausgehen, können Hinweise auf pathogenetische Zusammenhänge geben – so waren Calprotectin und LBP als Biomarker einer (intestinalen) Entzündung sowie gestörten Darmbarriere erhöht bei Patient:innen mit axialer Spondyloarthritis und zeigten eine Assoziation zur Krankheitsaktivität. Ferner können Biomarker prognostischen Wert besitzen: Die Biomarker Calprotectin, Visfatin, Leptin und sCTX waren mit einer radiografischen spinalen Progression in den folgenden Jahren unter Therapie mit Tumor-Nekrose-Faktor-alpha-Inhibitoren assoziiert. Allerdings scheinen global betrachtet diese Biomarker nebst deren Kombinationen nur einen geringen Vorteil gegenüber etablierten klinischen Parametern hinsichtlich der Beurteilung des Risikos einer radiografischen Progression zu besitzen.
View lessOsteoporotische Wirbelkörperfrakturen stellen aufgrund ihrer hohen Prävalenz und der damit verbundenen Morbidität eine bedeutende Herausforderung in der Wirbelsäulenchirurgie dar. Ziel dieser Habilitationsschrift war die Evaluation diagnostischer Verfahren zur Beurteilung der lokalen Knochenqualität sowie die Untersuchung klinisch relevanter Einflussfaktoren auf die operative Behandlung mittels Kyphoplastie.
Zunächst wurden die Aussagekraft der Dual-Röntgen-Absorptiometrie (DXA) und der hochauflösenden Multidetektor-Computertomographie (HR-MDCT) anhand intraoperativer Wirbelkörperbiopsien mit mikrocomputertomographischer (µCT) Analyse als Referenzstandard untersucht. Es zeigte sich, dass weder die DXA bei Patient mit osteoporotischen Wirbelkörperfrakturen noch die HR-MDCT eine verlässliche Beurteilung der lokalen vertebralen Knochenqualität ermöglichen. Die Ergebnisse verdeutlichen den Bedarf an der Entwicklung und Validierung neuer diagnostischer Verfahren.
Der zweite Schwerpunkt der Arbeit lag auf der Optimierung der operativen Therapie mittels Kyphoplastie. Es konnte nachgewiesen werden, dass die Kyphoplastie nicht nur zu einer effektiven Schmerzreduktion führt, sondern auch das globale sagittale Profil der Wirbelsäule verbessert. Dieser Effekt korrelierte mit der lokalen Reposition des frakturierten Wirbelkörpers und war unabhängig von der postoperativen Schmerzreduktion. Darüber hinaus zeigte sich, dass das radiologische Repositionsergebnis maßgeblich vom Operationszeitpunkt abhängt: Eine operative Versorgung innerhalb der ersten sechs Wochen nach Fraktur ermöglichte eine signifikant bessere Wiederherstellung der Wirbelkörperhöhe als spätere Eingriffe. Schließlich konnte erstmals ein prädiktiver Zusammenhang zwischen der OF-Klassifikation und dem zu erwartenden Repositionsergebnis nach Kyphoplastie nachgewiesen werden.
Insgesamt tragen die vorliegenden Arbeiten zu einem verbesserten Verständnis der diagnostischen Limitationen bei osteoporotischen Wirbelkörperfrakturen sowie zu einer evidenzbasierten Optimierung der Indikationsstellung und operativen Therapie bei. Die Ergebnisse liefern wichtige Grundlagen für zukünftige diagnostische Entwicklungen und eine individualisierte Behandlung dieser Patienten.
View lessBiological systems exhibit remarkable diversity, yet many functions essential to life remain preserved across the genomes and proteomes of evolutionarily distant organisms. Meanwhile, the rapid expansion of modern biological datasets has transformed even the most fundamental computational tasks into significant challenges at scale. Queries such as set membership and cardinality, though conceptually straightforward, become increasingly difficult to resolve efficiently using conventional methods when applied to massive sequence collections. Conserved biological motifs are naturally represented using regular expressions; however, despite decades of theoretical development and mature software implementations, existing regular expression search approaches remain insufficient for efficiently interrogating large biological sequence databases. In this dissertation, we introduce several novel algorithms for regular expression search that narrow the effective search space by exploiting the recently proposed (Hierarchical) Interleaved Bloom Filter, a compact data structure designed for scalable set membership queries. We demonstrate that integrating our approach with classical search techniques leads to substantial runtime improvements over current state-of-the-art methods.
View lessZiel der Studie zum Methodenvergleich (Boelow et al. 2022) war eine Gegenüberstellung des FECPAKG2-Verfahrens als neue FEC-Methode zur koproskopischen Untersuchung auf gastrointestinale Pferdehelminthen mit den Standard-Methoden M-FLOTAC und der semi-quantitativen Sed/Flo. Es wurden Analysen zur Häufigkeit positiv erkannter Proben, Korrelation der Ergebnisse, Intensität der Eiausscheidung und der Praktikabilität der Anwendung hinsichtlich des Zeitaufwandes der drei Methoden miteinander verglichen. Ergebnis dieser Analysen war, dass die Sed/Flo für einen Nachweis auf das Vorhandensein von Parasiten im Vergleich dieser drei Verfahren am sensitivsten ist. Soll die diagnostische Methode zur Ermittlung behandlungsbedürftiger Pferde bei einer selektiven Entwurmung oder zur Untersuchung der Wirksamkeit eines Anthelminthikums eingesetzt werden, ist der Einsatz einer FEC-Methode wie das M-FLOTAC-Verfahren oder das FECPAKG2-Verfahren erforderlich. Die M-FLOTAC-Methode ist sensitiver als das FECPAKG2-Verfahren. Bei einem EpG von über 200 gibt es eine gute Übereinstimmung zwischen den beiden Methoden. Zu berücksichtigen ist, dass eine Untersuchung mittels FECPAKG2 für Anoplocephaliden und O. equi bisher nicht ausreichend evaluiert worden ist. Unabhängig von den Ergebnissen der Studie ist zu berücksichtigen, dass die diagnostische Methode hinsichtlich des Untersuchungziels und der epidemiologischen Faktoren ausgewählt werden sollte. In der zweiten Studie (Boelow et al. 2023) wurden relevante Umwelt-, Wirts- und Management-spezifische Risikofaktoren ermittelt, die einen Einfluss auf die Prävalenz und die Häufigkeit auf das Auftreten von den am häufigsten beim Pferd auftretenden gastrointestinalen Helminthen, d.h. Strongyliden und Parascaris spp., haben (Relf et al. 2013; Kornaś et al. 2010). Relevante Faktoren wurden in Abhängigkeit von der jeweiligen Parasitenart/-gruppe berechnet. Jährlinge waren sigifikant häufiger positiv für Strongyliden und hatten eine höhere Eiausscheidungsintensität als adulte Pferde oder Fohlen. Es konnte eine erhöhte Eiausscheidung im Herbst im Vergleich zum Frühjahr gefunden werden. Es wurden keine Hinweise gefunden, die auf eine eingeschränkte Wirksamkeit von ML gegen Stongyliden auch in Kombination mit PY hinweisen würden. Die Pferde, die mit ML bzw. der Kombination mit ML und PY entwurmt wurden, zeigten auch bis zu 2 Monate nach der Entwurmung niedrigere Strongyliden-Prävalenzen und Eiausscheidungen. Hinsichtlich des Vorkommens von Parascaris spp. standen die wichtigsten Einflussfaktoren im Zusammenhang mit dem Alter, d.h. Fohlen waren signifikant häufiger betroffen. Bezüglich des Befalls mit Parascaris spp. korrelierte eine der Untersuchung vorhergehende Entwurmung mit einem Wirkstoff aus der Wirkstoffgruppe der ML, wie bei den Strongyliden, mit niedrigerer Prävalenz und geringeren Eizahlen. Trotz dieser Erkenntnisse besteht nach wie vor Forschungsbedarf hinsichtlich einer Weiterentwicklung der koproskopischen diagnostischen Verfahren, zum Beispiel im Hinblick auf die Speziesdifferenzierung von kleinen Strongyliden und einer Verbesserung der Genauigkeit und Präzision. Automatisierte Verfahren wie das OvaCyte® oder der VETSCAN IMAGYST® können dazu beitragen, innerhalb vergleichsweise kurzer Zeit viele Proben mit wenig personellem Aufwand zu untersuchen. Die Genauigkeit ist für die beim Pferd häufig vorkommenden Helminthen wie den Strongyliden und Parascaris spp. mit der der McMaster und M-FLOTAC Verfahren vergleichbar. Die Verwendung von künstlicher Intelligenz ermöglicht eine ständige Verbesserung durch maschinelles Lernen. Im Hinblick auf die Notwendigkeit einer möglichst genauen, effizienten Diagnostik zur Bekämpfung von Helminthen beim Pferd und dem Entgegenwirken von Anthelminthika-Resistenzen durch eine ungezielte Entwurmung, können diese automatisierten Verfahren einen wichtigen Beitrag leisten.
View lessHintergrund: Jugendliche, die sich nicht mit dem bei Geburt zugewiesenen Geschlecht identifizieren und mitunter die Selbstbezeichnung trans* verwenden, sind im Vergleich zu ihren cis-geschlechtlichen Peers häufiger von psychischen Problemen betroffen. Diagnostische Manuale und das medizinische Verständnis beziehen sich hierbei auf das Inkongruenzerleben zwischen Geschlechtsidentität, Zuweisungsgeschlecht und Körper, welches bei manchen gender-diversen Menschen Leidensdruck und Geschlechtsdysphorie verursachen kann. Weithin vernachlässigt wird allerdings, dass trans* Jugendliche verschiedenen Formen von Minderheitenstress wie Diskriminierung, Pathologisierung und Gewalt ausgesetzt sind. Auch Eltern trans* Jugendlicher sind mit Unsicherheiten, gesellschaftlichen Vorurteilen und eigenen emotionalen Herausforderungen konfrontiert. Es besteht jedoch ein Mangel an spezialisierten Versorgungsangeboten sowie Unwissen und Vorbehalte aufseiten des medizinisch-psychologischen Fachpersonals. Sorgeberechtigte werden außerdem zu wenig im Versorgungsprozess berücksichtigt, obwohl die elterliche Unterstützung als einer der wichtigsten protektiven Faktoren für die psychische Gesundheit trans* Jugendlicher belegt wurde. Als eine weitere Ressource und Gegengewicht zur Geschlechtsdysphorie und der Belastung wurde das Erleben von Geschlechtseuphorie identifiziert, wobei es an Studien mangelt, welche die gelebten Erfahrungen von trans* Jugendlichen widerspiegeln. Partizipative, community-basierte Forschungsansätze verfügen über das Potential, Lebensrealitäten von marginalisierten Personengruppen adäquat abzubilden und zeitgleich das Machtgefälle zwischen Forschenden und Beforschten abzubauen, finden jedoch weiterhin zu wenig Anwendung. Zielsetzung: Die vorliegende kumulative Dissertation widmet sich in vier Studien der Frage, welche Belastungen und Ressourcen trans* Jugendliche und ihre Eltern erleben. Dabei sollen sowohl interpersonelle als auch systemische Facetten und Perspektiven der Lebensrealitäten von trans* Jugendlichen und ihren Eltern beleuchtet werden. Methode: In den Studien 1 und 2 wurden quantitative Daten aus Erstvorstellungen der Inanspruchnahme-Population einer Spezialsprechstunde für trans* Kinder und Jugendliche der Charité-Universitätsmedizin Berlin analysiert. In Studie 1 wurde bei N=50 trans* Jugendlichen im Alter von 11-18 Jahren der Zusammenhang zwischen psychischer Symptombelastung, Peerbeziehungen und elterlichen Wahrnehmung der Symptome untersucht. In Studie 2 wurde zunächst die Körperunzufriedenheit von N=99 trans* Jugendlichen (TG) im Alter von 11 bis 18 Jahren mit einer Schulstichprobe (CG) von N=527 cis-geschlechtlichen Jugendlichen im Alter von 13 bis 16 Jahren verglichen. Innerhalb einer Sub-Stichprobe der TG (n=74) wurde anhand von korrelativen und Regressionsanalysen sowie Testungen auf Mittelwertsunterschiede Zusammenhänge zwischen Körperunzufriedenheit und Geschlechtsdysphorie mit schlechten Peer-Beziehungen und Transfeindlichkeit untersucht. In Studie 3 und 4 wurden qualitative Daten aus dem partizipativen Forschungsprojekt TRANS*PARENT analysiert, in welchem trans* Jugendliche sowie Expert*innen aus der Trans*Community in alle Forschungsstadien miteinbezogen wurden. In Studie 3 wurde in Fokusgruppen mit N=24 Eltern von trans* Jugendlichen untersucht, von welchen Belastungen die Eltern betroffen sind und ob sie selbst Minderheitenstress erleben. In Studie 4 wurde in Fokusgruppen mit N=30 trans* Jugendlichen im Alter von 11-18 Jahren exploriert, welchen Einfluss soziale Interaktionen und Normen auf ihr Erleben von Geschlechtsdysphorie und Geschlechtseuphorie haben. Ergebnisse: Es konnte in Studie 1 gezeigt werden, dass trans* Jugendliche häufiger von psychischen Problemen berichteten als ihre Peers und über die Hälfte der Stichprobe Suizidalität und Mobbingerfahrung angab. Je geringer die elterliche Wahrnehmung der Symptome und je schlechter die Peer-Beziehungen, desto höher waren die psychischen Probleme. Auch in Studie 2, 3 und 4 konnte gezeigt werden, dass ein nicht-affirmatives soziales Umfeld Einfluss auf das Wohlbefinden von trans* Jugendlichen hat: In Studie 2 zeigten diese höhere Körperunzufriedenheit als ihre cis-geschlechtlichen Peers, welche in Zusammenhang mit schlechten Peer-Beziehungen stand. Zudem wiesen trans* Jugendliche stärkere Geschlechtsdysphorie auf, wenn sie von schlechten Peer-Beziehungen und Transfeindlichkeit betroffen waren. Von ähnlichen Erfahrungen erzählten die trans* Jugendlichen in Studie 4; sie erlebten Geschlechtsdysphorie vor allem in Reaktion auf nicht-affirmative soziale Interaktionen und den Druck durch soziale Gendernormen, die teilweise Einfluss auf ihre Transitionsentscheidungen hatten. In Studie 3 berichteten auch die Eltern trans* Jugendlicher von Stressoren wie strukturellen Problemen (z.B. in Schule und Gesundheitssystem) und sozialer Ablehnung ihrer Kinder (z.B. Mobbing, Misgendern), aber auch eigenen Erfahrungen von Abweisung und Diskriminierung, welche zu emotionalen Belastungen wie Frustration, Traurigkeit und Ängsten der Eltern führten. Zeitgleich erlebten die trans* Jugendlichen in Studie 4 Geschlechtseuphorie, Authentizität und Stolz in affirmativen und supportiven sozialen Umfeldern. Diskussion: Die in dieser Dissertation vorgestellten Studien tragen dazu bei, das Verständnis für die Belastungen und Ressourcen von trans* Jugendlichen und ihren Eltern zu erweitern. Die Ergebnisse zeigen, dass trans* Jugendliche nicht durch die Geschlechtsinkongruenz per se, sondern vor allem durch Minderheitenstress wie nicht-affirmative bis transfeindliche soziale Interaktionen belastet sein können. Diese haben sowohl auf die psychische Gesundheit als auch das Erleben von Geschlechtsdysphorie Einfluss, weshalb in Anlehnung an andere Forscher*innen das Konzept einer „sozialen Geschlechtsdysphorie“ postuliert wird. Die Ergebnisse zeigen auch, dass die Eltern von trans* Jugendlichen bei der Versorgung mitgedacht werden müssen, um sie zu stärken, ihre Kinder zu unterstützen und eigene Belastungen zu reduzieren. Schließlich plädiert diese Dissertation dafür, durch strukturelle Interventionen und Aufklärung Minderheitenstress für trans* Jugendliche und ihre Eltern abzubauen und Geschlechtseuphorie und Stolz zu fördern. Hierfür ist es maßgeblich, sie als Expert*innen für ihre Lebensrealität wahrzunehmen, ihnen Gehör zu verschaffen und Mitgestaltung zu ermöglichen.
View lessStaphylococcal enterotoxins (SEs) are heat-stable, highly potent superantigens produced by Staphylococcus aureus. They play a central role in staphylococcal food poisoning and toxic shock syndrome. SEs have the capacity to trigger disease even in the absence of viable bacteria due to their stability during food processing and storage, thus posing a substantial risk to food safety and public health. The classical five SEs, SEA to SEE, have historically been the primary focus of food safety monitoring. However, there has been an increasing association of additional SEs, such as SEG, SEH, and SEI, with outbreaks. The accurate detection of SEs at the protein level is essential, given that genetic detection does not verify the expression of the toxins. However, the use of immunoassays for the detection of SEs has been hindered by the interference of protein A (SpA), which is a well-known virulence factor produced by S. aureus. As SpA able to bind specific parts of antibodies, they can cause falsepositive signals in immunoassays which complicate result interpretation.
The first part of this work addressed the challenge of broadening the scope and detecting multiple SEs simultaneously, as existing commercial assays are limited to SEA–SEE and often suffer from insufficient sensitivity or specificity. A novel multiplex suspension immunoassay (SIA) was developed to enable the simultaneous detection, differentiation, and quantification of SEA through SEI from minimal sample volumes using Luminex® technology. To achieve this objective, high-affinity monoclonal antibodies (mAbs) against SED, SEH and SEI were generated through immunisation and hybridoma technology, followed by rigorous screening for high specificity and minimal cross-reactivity. Those mAbs together with mAbs against SEA, SEB, SEC, SEE, and SEG available at RKI were each implemented in both individual sandwich enzyme-linked immunosorbent assays (ELISAs) and a multiplex SIA format. The multiplex SIA achieved detection limits in the low picogram per millilitre range for all eight SEs, thereby enabling sensitive detection across the entire targeted toxin spectrum, SEA to SEI.
In the process of assay validation, a comprehensive panel of 145 whole-genome sequenced strains of Staphylococcus spp., and Enterococcus faecalis was analysed. The multiplex SIA accurately detected and distinguished the presence of SEA to SEI in the liquid culture supernatants of these strains, achieving assay accuracies between 86.9% and 100%, depending on the SE type. Importantly, the multiplex SIA was capable of detecting one to four sequence variants for each SE, reflecting the natural sequence diversity of SEs and ensuring broad epidemiological coverage. The performance of the multiplex SIA was further confirmed using three exemplarily selected complex food matrices: milk, a processed meat product, and cheese. The multiplex SIA demonstrated high recovery rates and reproducible results, thus confirming its suitability for practical food safety testing. The implementation of the mAbs in the multiplex SIA represents the first occasion on which simultaneous detection of SEA through SEI can be performed with such high sensitivity and specificity from a single, minimal-volume sample.
The second part of this work focused on the development and optimisation of a highly sensitive sandwich ELISA specifically targeting SEB, a clinically and historically significant SE. In view of the fact that SEB has been subject as a biothreat agent, precise and low-level detection of SEB is critical for both civilian and biodefence applications. A previously established sandwich ELISA for SEB with a detection limit in the low picogram per millilitre range was modified to overcome interference caused by SpA. To achieve this, detection antibodies were engineered by adapting the IgG subclass to reduce SpA binding and by generating antibody Fragments through recombinant expression, which lack the Fc domain targeted by SpA. Concurrently, control capture antibodies were meticulously selected to guarantee the differentiation of specific SEB binding and cross-reactivity to SpA. The resulting optimised ELISA successfully abolished false-positive signals due to SpA in S. aureus culture supernatants, while maintaining excellent sensitivity with a limit of detection in the one-digit pg/mL range. The findings demonstrate a practical approach to resolving SpA-related interference in SE detection assays without the need for pre-incubation with blocking sera, which frequently compromises assay sensitivity.
The methodology of optimising the antibodies for detection of SEs in bacterial supernatants was utilised in the development of the new multiplex SIA to the overcome major limitations of existing methods by directly addressing SpA interference.
In summary, the present work provides advanced, reliable tools for the sensitive, specific, and simultaneous detection of eight SEs. These developments will enhance diagnostic Methods for food safety monitoring, enable precise outbreak investigation, and support effective public health interventions in response to staphylococcal food poisoning events.
View lessAuf Grund der großen wirtschaftlichen Bedeutung der Hypocalcämie der Milchkühe im peripartalen Zeitraum (Inzidenz der Gebärparese als der klinisch manifesten Form der Hypocalcämie 3 % bis 10 % der pluriparen Kühe, subklinische Hypocalcämie als eine gateway disease und damit wichtigen Risikofaktor für den postpartalen Krankheitskomplex 50 %) ist ein fest integriertes Prophylaxeprogramm gegen die peripartale Hypocalcämie obligater Bestandteil des modernen Herdenmanagements. Von den vier etablierten Prophylaxekonzepten (Calciumbolusgabe, calciumarme Trockenstehfütterung, Anionenration, intramuskuläre Injektion von 10 Mio IE Vitamin D3 2 bis 8 Tage ante partum) ist die 1977 in Deutschland eingeführte Hypervitamin-D-Prophylaxe in der internationalen Literatur umstritten. In Studien werden mit dieser Methode unterschiedliche, widersprüchliche Ergebnisse erreicht, die von positiven über keine bis zu negativen Wirkungen auf die Hypocalcämie, die Milchleistung und Erkrankungshäufigkeiten reichen. Ein zentraler Modulator der Prophylaxeeffektivität könnte die DCAB der Trockenstehration sein. Aufgabe dieser Untersuchung war es, im Rahmen einer Feldstudie die Wirksamkeit der Hypervitamin-D-Prophylaxe im Zusammenhang mit einer Trockenstehration mit moderat positiver DCAB zu prüfen, die weder eine azidogene noch eine alkalogene Stoffwechsellage fördert (DCAB +100 bis + 250 mequ/kg TS). Durch Gegenüberstellung der Ergebnisse mit aktuellen Studien mit vergleichbarem Design und gezielt eingestellter DCAB wird dem Einfluss der DCAB der Trockenstehration besondere Aufmerksamkeit geschenkt. In einem kommerziellen Milchviehbetrieb im Land Brandenburg mit 980 Holstein Friesian Milchkühen (Jahresmilchleistung 10500 kg, 4,02 % Fettgehalt, 3,34 % Eiweißgehalt, Lebenstagsleistung 15,0 kg, zweiphasige Trockenstehfütterung) wurden zufällig 322 Probanden ausgesucht. 155 Kühe wurden geplant 2 bis 8 Tage (real 0 bis 8 Tage) vor der Kalbung 10 Mio IE Vitamin D3 (Ursovit® D3, wässrig pro inj., Serumwerk Bernburg) injiziert, 167 Kühe dienten als Kontrolle. Die Vorbereiterration enthielt 6,4 NEL/kg TS, 147 g/kg TS Rohprotein, 5,2 g/kg TS Calcium, 4,4 g/kg TS Phosphor, 2,4 g/kg TS Magnesium. Die DCAB betrug +166 mequ/kg TS. Als Probanden wurden nur Kühe eingeschlossen, die zwischen 5.00 Uhr und 17.00 gekalbt hatten. Die erste venöse Blutprobenentnahme erfolgte am Folgetag um 7.00 Uhr (Tag 1) sowie an den zwei folgenden Tagen (Tag 2 und 3). In den Blutserumproben wurden die Konzentrationen an Calcium, Phosphor (anorganisches Phosphat) und Magnesium analysiert. Von den Probanden wurden Fruchtbarkeitskennzahlen, Parameter zum Geburtsverlauf und die Erkrankungen in den ersten 60 Laktationstagen dokumentiert. In der statistischen Auswertung wurde der Einfluss des Probentages, der Laktationszahl, der Jahreszeit, der Vitamin-D-Behandlung und des antepartalen Vitamin-D-Injektionszeitpunktes auf die Blutserumkonzentrationen an Calcium, Phosphor und Magnesium sowie des Einflusses der Vitamin-D-Injektion auf die Erkrankungshäufigkeiten in den ersten 60 Laktationstagen untersucht. 94,4 % der Probanden zeigten eine Normalgeburt, 1,2 % eine Zwillingsgeburt. Die Gebärpareseinzidenz betrug 5,9 %. Die Blutserumcalciumkonzentrationen (ȳ ± s) in der Gesamtstichprobe lagen an den Probentagen 1, 2, 3 bei 2,03 ± 0,28, 2,11 ± 0,26, 2,27 ± 0,22, die Phosphorkonzentrationen bei 1,28 ± 0,43, 1,40 ± 0,40, 1,63 ± 0,40, die Magnesiumkonzentrationen bei 0,96 ± 0,14, 0,94 ± 0,14, 0,87 ± 0,13. Die Laktationszahl beeinflusste hochsignifikant die Blutserumkonzentrationen an Calcium, Phosphor und Magnesium. Die Vitamin-D-Injektion hatte keinen Effekt auf die Blutserumcalciumkonzentration (mit Ausnahme der Kühe in der dritten Laktation an Probentag 2). Die Probanden mit einer Vitamin-D-Injektion zeigten signifikant höhere Blutphosphorkonzentrationen (Ausnahme Kühe ab 5. Laktation) und signifikant niedrigere Magnesiumkonzentrationen (Ausnahme Probanden in der 4. und ab 5. Laktation). Der Vitamin-D-Injektionszeitpunkt hatte keinen relevanten Einfluss auf die Blutserumkonzentrationen an Calcium, Phosphor und Magnesium. Die Häufigkeit von Erkrankungen in den ersten 60 Laktationstagen unterschied sich nicht zwischen der Vitamin-D-Gruppe und der Kontrollgruppe. In der vorliegenden Studie hatte die Hypervitamin-D-Injektion keinen relevanten Effekt auf die Blutserumcalciumkonzentrationen und auf die Erkrankungshäufigkeiten, was eine geringe Prophylaxewirkung anzeigt. Die Blutserumcalciumkonzentration an Tag 1 (2,03 mmol/l) spricht für eine geringe Bedeutung der subklinischen Hypocalcämie (per Definition < 2,0 mmol/l) in der Studienherde auf Grund eines bereits guten Herdenmanagements, weshalb Zusatzeffekte der Vitamin-Gabe gering ausfallen. Es wurden aber auch keine negativen Auswirkungen der Hypervitamin-D-Applikation festgestellt. Unter Berücksichtigung vergleichbar konzipierter Studien in der Literatur kann der DCAB der Trockenstehration eine entscheidende Rolle in der Ausprägung der Effekte der Hypervitamin-D-Injektion zugesprochen werden. Azidogene Trockenstehrationen (DCAB < 0 mequ/kg TS) unterstützen negative Effekte (Milchleistungsdepression, Anstieg postpartaler Erkrankungshäufigkeiten), bei neutralen Trockenstehrationen (DCAB +100 bis +250 mequ/kg TS) sind geringe Effekt auf die Hypocalcämie und Erkrankungshäufigkeiten zu erwarten und bei alkalogenen Trockenstehrationen (DCAB > +250 mequ/kg TS) können positive Effekte der Hypervitamin-D-Behandlung wirksam werden. Darüber hinaus wird die Effektivität der Vitamin-D-Prophylaxe immer von der Qualität des allgemeinen Herdenmanagements und insbesondere des Fütterungsmanagements modifiziert.
View lessDie Geländeprüfung in der Vielseitigkeit stellt eine kurzzeitige, hochintensive Belastung dar, bei der Aminosäuren als Substrate und Regulatoren des Muskelstoffwechsels sowie der Regeneration eine zentrale Rolle spielen. Die erste Publikation untersuchte longitudinale Veränderungen von Plasmaaminosäuren bei international startenden Vielseitigkeitspferden unter realen Wettkampfbedingungen. Es zeigten sich konsistente, zeitpunktabhängige Verschiebungen im Plasmaaminosäureprofil. Die Konzentrationen der Mehrzahl der Aminosäuren stiegen unmittelbar nach der Belastung an und normalisierten sich bis zum Folgemorgen weitgehend. Auffällig waren persistierend erhöhte Prolinkonzentrationen sowie gesunkene Glycinkonzentrationen am Folgetag. Die Metaboliten Ammoniak und Harnstoff blieben ebenfalls erhöht.
Die zweite Publikation untersuchte die saisonalen Veränderungen der Plasmaaminosäuren sowie deren Zusammenhänge mit der Leistung im Gelände bei den gleichen Pferden über eine 23-wöchige Turniersaison hinweg. Dabei zeigten Asparagin, Ornithin und Prolin signifikante Konzentrationschwankungen im Saisonverlauf. Zudem waren höhere Leucin-Basalwerte und höhere Histidinkonzentrationen nach der Belastung mit besseren Ergebnissen verbunden, während erhöhte Prolin- und Alaninkonzentrationen nach der Belastung mit schlechteren Fehlerpunktzahlen korrelierten. Leucin und Histidin könnten potenziell leistungslimitierende Aminosäuren darstellen, während Prolin und Alanin als sensitive Marker für Trainingszustand und Ermüdung geeignet sein könnten.
Die Aussagekraft der Studienergebnisse ist aufgrund ihres korrelativen Charakters und unberücksichtigter Einflussgrößen wie Fütterung, Transportstress sowie individueller Trainings- und Regenerationsstrategien eingeschränkt. Es sind kontrollierte Folgestudien erforderlich, die Störfaktoren systematisch berücksichtigen, Wiederholungsmessungen standardisieren und prüfen, ob Aminosäuren im Zusammenhang unter Berücksichtigung individueller Referenzbereiche und in Kombination mit Parametern wie der Herzfrequenz robuste Marker für Trainingszustand, Erholung und potenzielle Überlastung darstellen. Das Potenzial von Plasmaaminosäuren als wichtigem Bestandteil eines innovativen, multimodalen Ermüdungs- und Leistungsmonitorings für Vielseitigkeitspferde im Hochleistungssport wird in dieser Arbeit aufgezeigt.
View lessDie vorliegende Dissertation untersucht systematisch, ob sich merkmalsbasierte Distraktorinhibition – wie sie im Paradigma der Distraktorinduzierten Blindheit (DIB) beschrieben wird – auch über die visuelle Modalität hinaus im auditiven Kontext (Distraktorinduzierte Taubheit, DID) beobachten lässt und in welchem Ausmaß die Modalität des Hinweisreizes diesen Inhibitionsprozess zusätzlich beeinflusst. Hierzu wurden drei Studien durchgeführt: eine crossmodale DIB-Studie (Studie I), eine unimodale DID-Studie (Studie II) sowie eine crossmodale DID-Studie (Studie III), die behaviorale und elektrophysiologische Marker systematisch miteinander vergleichen.
Auf Verhaltensebene zeigt sich in allen Konfigurationen ein konsistenter, kumulativ anwachsenden Distraktoreffekt mit charakteristischem SOA-Muster: Mit steigender Anzahl zielreizgleicher Distraktoren sinkt die Wahrscheinlichkeit bewusster Zielreizdetektion, insbesondere bei simultaner Hinweisreiz-Zielreiz-Präsentation. Dieser Befund wird als Hinweis darauf gewertet, dass der zugrunde liegende Inhibitionsmechanismus prinzipiell durch visuelle wie auditive Distraktoren aktiviert werden kann.
Elektrophysiologisch zeigt sich hingegen ein stark differenziertes Bild. Für visuelle Distraktoren repliziert sich die frontale Negativität (visFN) als robuster Marker eines kumulativen, merkmalsbasierten Inhibitionsprozesses, der weitgehend unabhängig von der Modalität des Hinweisreizes sowie der konkreten visuellen Merkmalsdimension von Distraktoren und Zielreizen auftritt. Für die auditive Distraktorverarbeitung lässt sich hingegen kein strukturell analoges Korrelat identifizieren. Stattdessen zeigt sich hier ein komplexes Muster aus einer frühen fronto-zentralen Negativität (audFN), deren Ausprägung in Abhängigkeit von Distraktoranzahl und Hinweisreizmodalität variierte, sowie einer nachfolgenden, distraktor-getriebenen Modulation im centro-parietalen P3- Zeitfenster, die auf evaluative Weiterverarbeitung verweist. Es liegt nahe, dass insbesondere im frühen Zeitfenster mehrere Prozesse – darunter devianzbezogene Mechanismen, lokale Erwartungsprozesse sowie inhibitorische Kontrollprozesse – zusammenlaufen.
In der Gesamtschau sprechen die Befunde vorsichtig für eine supramodale Koordination inhibitorischer Kontrolle auf Ebene der Aufgabenanforderung, allerdings bei zugleich modalitätsspezifischer neuronaler Implementierung des zugrunde liegenden Mechanismus. Die Arbeit leistet damit einen Beitrag zur theoretischen Weiterentwicklung des DIB-/DID-Modells und zur Klärung der Frage, inwieweit vergleichbare Verhaltensphänomene in unterschiedlichen Sinnesmodalitäten auf gemeinsamen oder differenzierten neuronalen Mechanismen beruhen.
View lessThis dissertation develops and applies molecular simulation methods to investigate the time evolution of many-atom systems under diverse thermodynamic conditions, with emphasis on rare events such as chemical reactions. By integrating computational and data science approaches, it introduces automated tools that construct reaction networks and provide realistic, comprehensive descriptions beyond the reach of traditional methods.
Applications range from simple unimolecular reactions, such as formaldehyde isomerization, to more complex bimolecular processes like glycolaldehyde synthesis. Automated reaction network construction revealed new pathways and temperature dependencies that are challenging to capture with conventional approaches. To address mechanistic details, a trajectory-based molecular dynamics framework was developed, enabling bond-breaking and bond-forming processes to be analyzed without relying on high-dimensional potential energy surfaces.
Further contributions include improved methods for estimating molecular lifetimes, molecular dynamics–based approaches to infrared spectroscopy that naturally account for anharmonic effects, and a novel partial-pressure barostat for simulating multicomponent systems. Collectively, these methods enhance the predictive power, mechanistic insight, and experimental relevance of molecular simulations, advancing computational chemistry and many-body modeling.
View lessAccording to some scientists, we are currently living the 6th biodiversity mass extinction, marked by an unprecedented loss of biodiversity primarily driven by human activities. Habitat destruction, climate change, pollution, overexploitation, and the introduction of invasive species have accelerated species extinction rates to levels 10-100 times higher than the natural background rate. This biodiversity crisis represents one of the most critical environmental challenges of our time.
Among the species most severely affected are rhinoceroses, which face significant threats due to poaching and habitat loss. Rhino populations have declined drastically, with some species, such as the northern white rhino, now on the brink of extinction. Poaching for rhino horn, driven by traditional beliefs and status symbolism, continues to devastate these animals despite conservation efforts.
To combat such losses, assisted reproductive technologies (ART) have been increasingly incorporated into conservation programs, alongside the most traditional one, such as habitat protection and tailored breeding programs. Techniques such as in vitro fertilization (IVF), artificial insemination, and cryopreservation of genetic material offer hope for preserving genetic diversity and boosting populations of critically endangered species. These technologies, though still in development for many wildlife species, are a promising tool in the fight to preserve Earth’s biodiversity.
In this study, we present two different cases in which ART have been employed as conservational tools for two different species. in Chapter II, cryopreservation of matured oocytes was successfully applied to the critically endangered okapi (Okapia Johnstoni). This approach enabled the temporal storage of matured gametes while awaiting the availability of a suitable male counterpart. The cryopreserved oocytes were subsequently thawed and subjected to ICSI.
In Chapter III, an adaptation of the existing ART protocol for the murine species was utilized to produce blastocysts in the NRM (Hetecephalus glaber). This process began with the collection of cumulus-oocyte complexes (COCs) from sexually inactive individuals. Naked mole rats are particularly valuable for cancer and aging research due to their unique evolutionary traits, including exceptional longevity and cancer resistance.
View lessMelanoma is the cancer of melanocytes, pigment producing cells and an aggressive type of cancer due to its ability to migrate into distant organs at early stages. While cutaneous melanoma (CM) accounts for the majority of cases and often responds to targeted and immune therapies, uveal melanoma (UVM), a rarer form originating in the eye, remains largely unresponsive and is frequently managed through drastic measures such as enucleation. This disparity likely reflects fundamental molecular differences: UVM exhibits a low tumor mutational burden (TMB) compared to CM and distinct gene expression patterns, suggesting the possibility that epigenetic mechanisms may play a key role in its progression and treatment resistance. By integrating genomics, transcriptomics, and epigenomics data within a systems biology approach, we leveraged multi-omics research to reveal the pathways and regulatory networks, helping to explain the contrasting behaviors of CM and UVM and in the long run paving the way for more precise, effective therapies.
Our bioinformatics analyses on an advanced metastatic melanoma cohort confirmed established knowledge into the genetic landscape and subgroup-specific differences of melanoma. Our in-depth transcriptome analysis extended the current knowledge on gene expression landscapes. We uncovered co-expressed mechanisms embedded in distinct gene regulatory networks, suggesting alternative modes of co-regulation for different subtypes; one such example was on immune activation. We also profiled the epigenome of uveal melanocytes, which showed an unexpectedly high level of DNA methylation compared to healthy tissues including the skin melanocyte. The methylation difference observed in the tissue-of-origin level of the healthy samples is potentially a key player of the molecular divergence of UVM from CM. Differentially methylated regions (DMRs) between UVM and CM were enriched in enhancer elements. Enhancer elements are often characterized using ChIP-seq on histone mark combinations. As an alternative approach, we explored enhancer-associated RNAs (so called Enhancer RNA (eRNA)s), ncRNAs transcribed from active enhancers. We developed EnhancR, a command-line tool that systematically extracts and quantifies eRNA activity from deeply sequenced bulk RNA-seq data, then links eRNAs to their putative target genes. Using both patient cohort data and an in vitro cell line model, we examined the performance of EnhancR, identified its technical boundaries, and gained further insights into epigenetic regulation of UVM.
View lessThis dissertation examines how expectations, global price linkages, financial frictions, and institutional arrangements shape the transmission and conduct of monetary, fiscal, and financial-stability policies. Combining high-frequency event studies, Bayesian structural vector autoregressions, empirical policy counterfactuals, and New Keynesian models, its four essays provide complementary evidence from international and euro-area settings. First, the dissertation shows that credible inflation targeting insulates economies from U.S. monetary-policy spillovers: when long-term inflation expectations are weakly anchored, foreign tightening produces stronger inflationary pressures, more aggressive domestic interest-rate responses, and larger output losses, thereby restricting monetary-policy autonomy. Second, it demonstrates that ECB monetary tightening lowers global oil prices and euro-area consumer energy prices; because energy prices adjust rapidly, this channel accelerates disinflation and improves the inflation-output trade-off, while treating energy prices as exogenous leads to overstated interest-rate requirements and a less favorable assessment of mandate-optimal policy responses to energy-supply shocks. Third, a novel identification of Eurosystem collateral policy interventions reveals that collateral easing raises bank equity values, reduces financial uncertainty and bank default risk, and lowers sovereign borrowing costs—especially in peripheral countries—highlighting a distinct and uneven transmission mechanism operating through banking-sector fragmentation and sovereign-bank linkages. Finally, the dissertation finds that although both tax cuts and government spending increases stimulate output in a monetary union, government consumption and investment generate substantially larger and more persistent effects because their inflationary impact lowers real interest rates when common monetary policy does not respond to country-specific shocks. Overall, the findings show that macroeconomic policies cannot be evaluated independently of the environments in which they operate, as their effectiveness and associated policy trade-offs depend critically on credibility, endogenous global prices, financial-market structure, and monetary-union institutions.
View lessDescribing the electronic structure of molecules using computational methods is of central importance in many areas of chemical research and industry. To this day, the variational quantum Monte Carlo (VMC) algorithm, which utilizes the variational principle of quantum mechanics to optimize an ansatz towards the true electronic wave function, is one of the lesser employed of such computational methods. While it is generally applicable, and provides direct access to the wave function, its accuracy has been limited by insufficiently expressive ansatzes. This barrier was recently lifted with the introduction of deep-learning-based ansatzes, providing unprecedented flexibility and corresponding accuracy, at the cost of increased computational requirements. In this thesis, our implementation of the VMC algorithm with deep-learning ansatzes is described, along with a number of developments facilitating the computation of electronic excited states, extraction of various observables, and characterization of potential energy surfaces, at a reduced computational cost. The method is implemented as open-source software, employing a common framework to encompass most ansatzes present in the literature. Utilizing the direct wave function access granted by VMC, an approach is developed to extract one-electron reduced densities, that are free from Gaussian basis set effects. Significant advances in the methodology of computing electronic excited states via the ladder-of-states approach are made, including an improved form of the overlap penalty term with an automatically determined prefactor, and a new loss term with which the total spin of the solution can be selected. The efficiency of computing excited potential energy surfaces are enhanced by optimizing a single ansatz for a range of molecular configurations. To enable such transferable training in the presence of state crossings, a dynamic, geometry-wise ordering in the ladder of states is introduced. Geometry-optimization-based tasks are identified as natural fits for such transferable ansatzes, and a transferability-native approach for extracting the necessary information from VMC data is devised. Finally, transferable training is employed to simultaneously model different chemical species, resulting in a foundation model predicting bond breaking at a cost-accuracy ratio competitive with mature multi-reference methods. The work presented here contributes significantly to the development of a practically useful deep-learning-based VMC method. The described approaches have the potential to become tools for a wide community of computational chemists, as a source of accurate data, describing strong correlation in both ground and excited states.
