Optic atrophy 1 (OPA1) is a dynamin-related GTPase, which is imported into mitochondria. Mutations in OPA1 cause autosomal dominant optic atrophy (adOA) and lead to blindness. Haploinsufficiency was postulated as pathological mechanism for adOA, resulting in loss of retinal ganglion cells and degeneration of the optic nerve. OPA1 was extensively studied in the last five years, and it was implicated in fusion of mitochondria. Fusion and fission of mitochondria are processes constantly happening in cells, making mitochondria not static, but dynamic organelles. They ensure that an equilibrium is kept between short and long forms of mitochondria and allow exchange of their contents. Even with many excellent studies on OPA1 done in mammalian cells, the function of this protein is not yet clear. In order to study its physiological function in vivo and its role in the manifestation of autosomal dominant optic atrophy, the OPA1 gene was disrupted by homologous recombination. OPA1-deficient heterozygous (OPA1+/-) mice show reduced OPA1 protein levels, but are phenotypically indistinguishable from wild-type littermates. On the other hand, OPA1-deficient homozygous (OPA1-/-) embryos are reduced in size and die during gastrulation. Mitochondria in OPA1-/- embryos are completely fragmented, loose their DNA and many cells show hallmarks of apoptosis. Mouse embryonic fibroblasts (MEFs) derived from OPA1-/- embryos also contain completely fragmented mitochondria, which have lost their ability to undergo mitochondrial fusion. Their mitochondria are enlarged, with abnormal inner membrane morphology. They show complete respiration deficiency and mitochondrial membrane potential decrease. OPA1-/- fibroblasts are less sensitive to staurosporine-induced apoptosis and do not undergo cytochrome c release, opposite to what was seen in wild-type cells after the same treatment. Transfection of OPA-/- MEFs with wild-type OPA1 isoform 1 restored respiratory chain activity, mitochondrial membrane potential, tubular morphology and fusion of the mitochondrial network. However, the sensitivity to staurosporine-induced apoptosis was not restored. Results presented in this thesis show that functional mitochondrial networks are essential in early embryonic development and that OPA1 is required for cristae maintenance, activity of respiratory chain and tubulation of mitochondrial networks. Therefore, OPA1 is absolutely necessary for early embryonic development and survival. Besides, reconstitution experiments showed that one particular OPA1 isoform, out of eight isoforms known, has special and distinct functions in mitochondria.
Optische Atrophie 1 (OPA1) ist eine Dynamin-verwandte GTPase, die in die Mitochondrien importiert wird. Mutationen im OPA1 Gen führen zur Erblindung in Patienten mit autosomal dominant erblicher Optikusatrophie (adOA). Als Pathomechanismus für die adOA wurde Haploinsuffizienz postuliert, die zu einem Verlust an retinalen Ganglienzellen führt und in einer Degeneration des optischen Nervs resultiert. OPA1 wurde während der letzten fünf Jahre sehr intensiv studiert und die Ergebnisse lassen eine zentrale Rolle bei der Fusion von Mitochondrien erwarten. Fusion und Teilung von Mitochondrien sind Prozesse, die ständig in Zellen ablaufen, und Mitochondrien zu sehr dynamischen Organellen machen. Sie stellen sicher, dass ein Gleichgewicht zwischen kurzen und langen Mitochondrien in der Zelle herrscht, und dass einzelne Mitochondrien ihren Inhalt, wie z.B. mitochondriale DNA, untereinander austauschen können. Obwohl viele Untersuchungen zu OPA1 an Säugerzellen unternommen wurden, ist die Funktionsweise dieses Proteins noch nicht verstanden. Um seine physiologische Rolle in vivo zu studieren und seinen Einfluss bei der Manifestation der erblichen Optikusatrophie zu untersuchen, wurde die orthologe OPA1-Gensequenz in Mäusen durch homologe Rekombination zerstört. Heterozygote OPA1-defiziente Mäuse (OPA1+/-) weisen reduzierte OPA1 Proteinlevel auf, sind aber phänotypisch unauffällig im Vergleich zu Wildtyptieren. Im Gegensatz dazu zeigen homozygote OPA1-defiziente Mäuse (OPA1-/-) ein reduziertes Wachstum während der frühen Embryonalphase und sterben noch während der Gastrulation. Mitochondrien in OPA1-/- Embryonen sind vollständig fragmentiert und weisen einen Verlust an mitochondrialer DNA auf. Viele Zellen des Embronalgewebes demonstrieren typische Anzeichen des programmierten Zelltods. OPA1-/- Embryonalgewebe wurde zur Etablierung von Fibroblastenkulturen (MEF, mouse embryonic fibroblasts ) genutzt. Auch diese OPA1-/- Zellen enthalten vollständig fragmentierte Mitochondrien, die ihre Fähigkeit zur Fusionierung verloren haben. Die Mitochondrien sind vergrössert und zeigen eine veränderte Morphologie der inneren Mitochondrienmembran. Sie sind ausserdem respirationsdefizient und weisen ein verringertes mitochondriales Membranpotential auf. Schließlich reagieren OPA1-/--Fibroblasten im Gegensatz zu Wildtypzellen weniger sensitiv auf die Apoptose-induzierende Stimulation mit Staurosporin und setzen kein Cytochrom c frei. Die Transfektion der OPA1-/- Zellen mit der Wildtypform von OPA1-Isoform1 führt zur Wiederherstellung der Atmungskettenfunktion, des mitochondrialen Membranpotentials, einer tubulären Mitochondrienmorphologie und der Fusionskompetenz des mitochondrialen Netzwerks. Es ist zu folgern, dass OPA1 absolut notwendig ist für die frühe Embryonalentwicklung und das Überleben von Zellen bzw. Embryonen. Ausserdem konnte durch Rekonstitutionsexperimente für eine der acht bekannten Isoformen von OPA1 gezeigt werden, für welche mitochondrialen Funktionen sie von Bedeutung ist.
