In this work a new type of core-multishell architecture via straight forward three-step synthesis was developed. As precursor, cheap, commercially available building blocks were used. These systems can act as universal supramoleculary aggregated hosts to encapsulate various types of guest molecules such as polar and nonpolar organic molecules or metal ions. The solubility of these polymers in a broad range of solvents permit the transport of guest molecules into polar or nonpolar environments independent of the guests polarity and solubility. This is in contrast to already existent systems which can either transport nonpolar molecules into aqueous environment, or transfer polar molecules into a hydrophobic media. Therefore, core-multishell architectures, with their universal transport behavior, can be considered as chemical chameleons . To understand the role of the individual domains (core, inner shell, outer shell) of these multishell architectures for the transport of guest molecules each individual part was varied. Encapsulation experiments revealed that modifications of every domain quantitatively influence the transport capacities of polymers. In contrast to the quantitative changes caused by modifications of core and outer shell, the presence of the inner, hydrophobic domain is essential for encapsulation abilities of core-multishell architectures. This influence was crucial for both polar and nonpolar guest molecules. Additionally, the influence of nonstructural factors, such as polymer concentration, guest molecule concentration, and time of encapsulation, on the transport capacity of nanotransporters has been established. To better understand the underlying principles of transport abilities of core-multishell architectures, the system was investigated by four independent techniques: surface tension measurement, dynamic light scattering (DLS), AFM, and CyroTEM. The measurements indicates that supramolecular aggregates are responsible for their transport ability. Localization of the encapsulated guest molecules in the aggregate structure was performed with the environment-sensitive dye nile red. Analysis of the UV/Vis absorbance spectra of encapsulated dye as a function of the time of the uptake process, revealed two different nanoenvironments around the guest molecules. This corresponds to the two different sites of the dye encapsulation: inter- and intramolecular cavities . In summary core-multishell architectures are new and very promising materials for host-guest chemistry. They can be used to transport and/or stabilize the different guest molecules such as drug, dyes, toxic compounds, metal nanoparticles and ions, and fluorescent markers and therefore can find a multiple applications in medicine, pharmaceutical and cosmetics industry, as well as catalysis.
Im Rahmen dieser Arbeit wurde eine effiziente dreistufige Synthese zur Herstellung neuer Kern-Schale-Architekturen mit einem hochverzweigten polymeren Kern, entwickelt. Als Vorläufer dieser Architekturen dienten kommerziell erhältliche Bausteine. Die sogenannten Multischalennanotransporter können nicht nur polare, unpolare und ionische Gastmoleküle einschließen, sondern sich auch an verschiedenste Polaritäten der Umgebung anpassen. Ihre gute Löslichkeit in einer Vielzahl organischer Lösungsmittel erlaubt den Transport eingeschlossener Gastmoleküle in sehr unterschiedliche Umgebungen, einschließlich unpolaren und polaren organischen Lösungsmitteln, so wie wässrigen Lösungsmitteln. Im Unterschied zu bereits existierenden micellaren Systemen können diese Systeme entweder unpolare Moleküle in wässrige Umgebung, oder polare Moleküle in eine hydrophobe Umgebung transportieren. Basierend auf diesen universellen Transporteigenschaften verhalten sie sich wie chemische Chamäleons . Um die Rolle jeder einzelnen Domäne (Kern, innere Schale, äußere Schale) zu verstehen, wurden diese im Rahmen dieser Arbeit unabhängig voneinander variiert. Durch geeignete Experimente wurde gezeigt, dass die Transportkapazität der Polymere durch die Änderung einzelner Domänen beeinflusst wird. Während die Größe der inneren und äußeren Schale nur einen geringen Einfluss auf die Transportkapazität haben, haben die Größe des polaren Kerns und die Kettenlänge der unpolaren inneren Schale einen signifikanten Einfluss auf den Transport hydrophiler und hydrophober Gastmoleküle. Dieser Einfluss wurde eindrucksvoll sowohl für unpolare, als auch für polare Moleküle nachgewiesen. Des Weiteren wurde der Einfluss nichtstruktureller Faktoren, wie die Konzentrationen des Polymers und des Gastmoleküls, die Einschlusszeit und die Transportkapazität untersucht. Um die Prinzipien dieses universellen Transportverhaltens besser zu verstehen, wurden die Aggregationseigenschaften der Nanotransporter mit unabhängigen experimentellen Methoden untersucht: Messung der Oberflächenspannung, dynamische Lichtstreuung (DLS), AFM, und CryoTEM. Diese neuen universellen Nanotransportsysteme können vielseitig für den Transport von Gastmolekülen wie Wirkstoffen, Farbstoffen, toxischen Verbindungen, Stabilisatoren, Metallnanopartikeln und Fluoreszenzmarkern eingesetzt werden und sind deshalb interessant für eine Vielzahl von biomedizinischen und technologischen Anwendungen.
