In much of the brain, no neurons are born in the adulthood but two regions are exceptional to this rule: the dentate gyrus of the hippocampus (where neurons arrive from the subgranular zone) and the olfactory bulb (where neurons arrive from the subventricular zone). Even though several theories regarding the possible functional role of adult neurogenesis have been proposed, the properties of the precursor cells and their regulation in the dentate gyrus remain largely unknown. To get a better insight into this topic the following work has focused on the precursor cells of the adult dentate gyrus by growing them in vitro and studying their properties. In this thesis work I have focused on the following questions: 1\. Can precursor cells from the adult murine dentate gyrus be cultured ex vivo and maintained in a tissue culture environment for prolonged periods of time? 2\. Does the adult dentate gyrus harbor stem cells in the true sense of their definition? 3\. What are the characteristics of adult hippocampal precursor cells and how do they react to their environment? 4\. Do the precursor cells cultured ex vivo generate neurons that are similar in property to their in vivo counterparts? 5\. Does neural activity regulate neural precursor cells property in the dentate gyrus? If so, what is the mechanism that realizes this cross talk?
Im ausgewachsenen Gehirn entstehen keine Nervenzellen - mit Ausnahme zweier Regionen: des Gyrus dentatus' im Hippocampus (wo neu gebildete Nervenzellen aus der subgranulären Zone vorkommen) und des Bulbus olfactorius' (wo neu gebildete Nervenzellen aus der subgranulären Zone vorkommen). Obwohl es mehrere Theorien zur möglichen Funktion der adulten Neurogenese gibt, sind die Eigenschaften von Vorläuferzellen und ihre Regulation im Gyrus dentatus weitgehend unbekannt. Um diesen Aspekt der adulten Neurogenese besser zu verstehen, untersucht die vorliegende Arbeit die Eigenschaften von Vorläuferzellen aus dem adulten Gyrus dentatus der Maus ex vivo. Die vorgelegte Doktorarbeit konzentriert sich auf die folgenden Fragestellungen: 1\. Können Vorläuferzellen aus dem adulten Gyrus dentatus der Maus ex vivo in Zellkultur gebracht und über einen längeren Zeitraum in Gewebe-Zellkultur gehalten werden? 2\. Beinhaltet der adulte Gyrus dentatus Stammzellen? 3\. Welche Charakteristika weisen die Vorläuferzellen im adulten Hippocampus auf, und wie reagieren diese Zellen auf ihre Umgebung? 4\. Bringen ex vivo- kultivierte Vorläuferzellen aus dem adulten Gyrus dentatus Nervenzellen hervor, die charakteristische Eigenschaften von in vivo-Nervenzellen zeigen? 5\. Reguliert die neurale Aktivität die Eigenschaften der Vorläuferzellen im Gyrus dentatus? Wenn ja, welcher Mechanismus gewährleistet diese Interaktion?
