RuvBL1 and its homolog RuvBL2 (RuvB-like) are evolutionarily highly conserved eukaryotic proteins belonging to the AAA+ family of ATPases (ATPase associated with diverse cellular activities). They play important roles in chromatin remodelling, in transcriptional regulation, in DNA repair and in the c-Myc and Wnt signalling pathways. Proteins involved in these pathways are often mutated in human cancers. Both RuvBL proteins form a complex and act together in diverse cellular processes, for example in chromatin remodelling within the INO80 and TIP60 complexes. RuvBL1 and RuvBL2 also have additional separate functions during transcriptional regulation and mitosis. In this thesis I present the three-dimensional structures of the selenomethionine variant of human RuvBL1 at 2.2 Å resolution and of the complex formed by RuvBL1 and RuvBL2 in which their respective domains II have been truncated. RuvBL1 assembled into a hexameric structure, whereas the RuvBL1/RuvBL2 complex formed a dodecamer consisting of two hexameric rings. Small-angle x-ray scattering (SAXS) experiments confirmed this structural organisation of RuvBL1 and of the RuvBL1/RuvBL2 complex in solution. Within the RuvBL1 hexamer and the dodecameric complex structure, an ADP molecule was bound to the monomers. RuvBL1 and RuvBL2 monomers contained three domains, of which the first and the third were involved in ATP binding and hydrolysis. Structural homology suggested that the second domain, which is unique among AAA+ proteins and not present in the bacterial homolog RuvB, was a novel DNA/RNA-binding domain. Nucleic acid-binding studies confirmed that RuvBL1 and also its purified domain II interacted with ssDNA, ssRNA and dsDNA, while RuvBL2 did not bind to various nucleic acid substrates. These results underpin that both proteins have different characteristics to fulfil their specialised functions. Other groups already localised the regions in RuvBL1 and RuvBL2 responsible for interaction with c-Myc, β-catenin and other proteins. The crystal structure of RuvBL1 revealed that these interacting regions belonged to domain II, which is ideally situated to contact other proteins, as it protrudes out of the hexameric ring. My results suggest that domain II is not only important for nucleic acid binding of RuvBL1, but also for the recruitment of cofactors interacting with RuvBL1 and RuvBL2. Although it has been shown that the ATPase activity of RuvBL1 and RuvBL2 is needed for several in vivo functions, I could only detect a marginal enzymatic activity with the purified individual proteins. The structure of the RuvBL1/ADP complex showed that hexamerisation blocked the nucleotide-binding pocket and that the ADP molecule was tightly bound to the monomer. A significantly higher ATPase activity was observed for the RuvBL1/RuvBL2 complex. Surprisingly, this activity was further stimulated after truncation of domain II, suggesting that conformational changes within the RuvBL1/RuvBL2 complex allowed a better exchange between ADP and ATP. Since RuvBL1 and RuvBL2 contain all the structural motifs characteristic of an ATP- driven helicase, this activity was tested with the purified proteins. I could clearly demonstrate that RuvBL1, RuvBL2 and their complex can only exert helicase activity when domain II is truncated, suggesting that cofactors drive conformational changes via domain II and allow in vivo enzymatic activity of these highly conserved proteins.
RuvBL1 und RuvBL2 (RuvB-like) sind hochkonservierte eukaryotische Proteine, die zu der Familie der AAA+ ATPasen gehoeren (ATPases associated with diverse cellular activities). Zu ihren wichtigen Funktionen in der Zelle zaehlt die Regulation der Chromatinstruktur und der Transkription. Darueberhinaus sind RuvBL1 und RuvBL2 an der DNA Reparatur beteiligt und stellen regulatorische Komponenten der c-Myc und Wnt Signalwege dar. Viele Proteine, die bei diesen Signalwegen Funktionen uebernehmen, sind bei humaner Cancerogenese mutiert. Obwohl RuvBL1 und RuvBL2 einen Komplex bilden und so gemeinsam arbeiten, fuehren sie einige Aufgaben auch separat aus. In dieser Arbeit wurde die dreidimensionale Struktur von humanem RuvBL1 und von dem Komplex aus RuvBL1 und RuvBL2 geloest. RuvBL1 bildet eine hexamere Ringstruktur, waehrend der Komplex aus RuvBL1 und RuvBL2 ein Dodekamer darstellt, welches sich aus 2 hexameren Ringen zusammensetzt. SAXS-Experimente (small-angle x-ray scattering) bestaetigten, dass RuvBL1 und der RuvBL1/RuvBL2 Komplex in Loesung tatsaechlich Hexamere, beziehungsweise Dodekamere bilden und diese strukturelle Organisation kein Artefakt der Kristallisation war. Aus den Kristallstrukturen ist ersichtlich, dass in allen RuvBL1 und RuvBL2 Monomeren ein ADP Molekuel in der Nukleotid-Bindungstasche vorhanden ist. Die Proteine RuvBL1 und RuvBL2 bestehen aus 3 Domaenen, von denen die erste und die dritte fuer die ATP-Bindung und Hydrolyse wichtig sind. Die zweite Domaene, welche durch die Strukturaufklaerung von RuvBL1 das erste Mal identifiziert wurde, ist bislang einzigartig in der Familie der AAA+ ATPasen und kein Bestandteil des bakteriellen Homologs RuvB. Sie weist nicht nur strukturelle AEhnlichkeiten zu einigen DNA-Bindedomaenen anderer Proteine auf, sondern interagierte im Fall von RuvBL1 tatsaechlich mit einzel- und doppelstraengiger DNA und RNA. Im Gegensatz dazu interagierte RuvBL2 nicht mit Nukleinsaeuren. Diese Ergebnisse verdeutlichen, dass RuvBL1 und RuvBL2 unterschiedliche Eigenschaften haben, um ihre speziellen Funktionen auszuueben. Andere Arbeitsgruppen lokalisierten bereits die Aminosaeure-Abschnitte in RuvBL1 und RuvBL2, welche fuer die Interaktion mit c-Myc, β-catenin und weiteren Proteinen verantwortlich sind. Die Kristallstruktur von RuvBL1 zeigte, dass diese Interaktionsflaechen zu der Domaene II gehoeren. Diese ragt aus dem hexameren Ring heraus und ist somit ideal positioniert, um mit anderen Proteinen zu interagieren. Die hier dargestellten Ergebnisse belegen, dass Domaene II nicht nur an der DNA-Bindung von RuvBL1 beteiligt ist, sondern auch fuer die Interaktion von RuvBL1 und RuvBL2 mit Kofaktoren benoetigt wird. Obwohl bereits mehrfach in der Literatur gezeigt wurde, dass die ATPase Aktivitaet von RuvBL1 und RuvBL2 fuer verschiedene in vivo Funktionen essentiell ist, besitzen die einzelnen aufgereinigten Proteine nur eine sehr geringe in vitro Aktivitaet. Die Struktur des RuvBL1/ADP Komplexes macht deutlich, dass die Hexamerisierung der RuvBL1 Monomere die Nukleotid- Bindungstasche blockiert und dass die ADP Molekuele sehr stark an die einzelnen Monomere gebunden sind. Allerdings zeigt der Komplex aus RuvBL1 und RuvBL2 eine deutlich hoehere ATPase Aktivitaet als die einzelnen Proteine. Diese Aktivitaet wird ueberraschenderweise gesteigert, wenn die Domaenen II in RuvBL1 und RuvBL2 verkuerzt werden. Die Ergebnisse deuten darauf hin, dass Konformationsaenderungen innerhalb des RuvBL1/RuvBL2 Komplexes einen besseren Austausch zwischen ADP und ATP erlauben. Da RuvBL1 und RuvBL2 alle strukturellen Eigenschaften enthalten, die charakteristisch fuer ATP- abhaengige Helikasen sind, wurden die gereinigten Proteine auf Helikaseaktivitaet getestet. Es wurde gezeigt, dass RuvBL1, RuvBL2 und der Komplex aus beiden Proteinen nur dann Helikaseaktivitaet ausueben koennen, wenn ihre Domaene II verkuerzt ist. Dies ist ein Hinweis darauf, dass innerhalb der Zelle ueber die Domaene II Konformationsaenderungen hervorgerufen werden, die moeglicherweise durch Kofaktoren bewerkstelligt werden und eine enzymatische in vivo Aktivitaet dieser hochkonservierten Proteine erlauben.
