Der starke Anstieg dezidierter Forschungsprojekte belegt: Die Rolle des
Immunsystems in der körpereigenen Tumorabwehr hat in den letzten Jahren
zunehmend an wissenschaftlicher Bedeutung gewonnen. So konnte z. B. gezeigt
werden, dass Tumorzellen durch die Zellen des Immunsystems erkannt und
anschließend eliminiert werden. Bei dieser Abwehrreaktion übernehmen
insbesondere Zytokine eine äußerst wichtige regulierende Funktion. Bereits
heute ist eines dieser Zytokine, das Interferon-alpha2 (IFN-alpha2) zur
adjuvanten Therapie des malignen Melanoms zugelassen. Doch die genaue
Wirkungsweise des Therapeutikums ist bislang noch weitestgehend unbekannt. Im
Rahmen dieser Arbeit sollten die Wirkungsmechanismen von IFN-alpha2 bei der
antitumoralen Immunantwort zelltypgenau untersucht werden. Hierfür wurden
murine Tumorzellen des malignen Melanoms in verschiedene transgene Mauslinien
transplantiert und mit syngenen Wildtyp-Kontrollen verglichen. Während der
Untersuchungen von Stroma und T Zellen konnte die essentielle Bedeutung des
IFN-alpha2 bei der funktionierenden Tumorabwehr bestätigt werden. Die
Resultate zeigen, dass lokal produziertes IFN-alpha2 die Blutgefäßdichte im
Tumorstroma und damit die Versorgung der Tumorzellen verringert. Dies
resultiert in einem verminderten Tumorwachstum. Zudem exprimiert das Stroma
unter Einfluss von IFN-alpha2 spezielle Proteine, die als Chemokine bekannt
sind. Diese wiederum rekrutieren Immunzellen, welche aktiv an der
Tumorabwehrreaktion beteiligt sind. Ein völlig neuer Aspekt zeigt sich in dem
Befund, dass IFN-alpha2 T Zellen tatsächlich auf direktem Weg kontrolliert und
von dieser Kontrolle die gesamte Melanomabwehr abhängt. Die vertiefte
Erkenntnis über die zelltypgenaue Wirkungsweise des IFN-alpha2 kann die
Entwicklung effizienter Therapien für Tumorerkrankungen unterstützen. So
könnten neue therapeutische Ansätze die Interaktion von T Zellen und Stroma
stärker berücksichtigen.

In the last few years the role of the immune system in the body s own tumour
defence has been increasingly appreciated. Many studies have shown that the
immune system can recognise and eliminate tumour cells, with cytokines
secreted playing a central role. Indeed, one of these cytokines interferon-
alpha2, is used therapeutically as adjuvant treatment against malignant
melanoma. However, despite its efficacy, its anti-tumour mechanism of action
remains largely unclear. The aim of the work in this thesis is to investigate
how IFN alpha2 affects the anti-tumour immune response, specifically through
its interaction with various cell types. In order to study this, mouse
malignant melanoma cells were transplanted into transgenic mice lacking the
Interferon type I receptor, using wild mice as controls. The results show that
locally produced IFN alpha2 reduces the blood vessel density in the tumour
stroma, and thus the blood supply of tumour cells, resulting in reduced tumour
growth. Additionally, under the influence of IFN alpha2 the stroma produces
chemokines, which play a role in recruiting immune cells to the tumour.
Furthermore, IFN alpha2 also directly controls T cell activity and the entire
anti-melanoma defence is dependent on this action. The new knowledge gained
from these studies about the role of IFN alpha2 on the anti-tumour immune
response will aid in the development of more efficient therapies for human
cancers, by focusing on the interaction between T cells and the surrounding
tissues.
