Hintergrund und Zielsetzung: Ein wesentlicher Bestandteil des Glioblastoms (GBM) sind sogenannte gliom-assoziierte Makrophagen und Mikrogliazellen (GAMs). Da diese die Progression und Invasivität des GBMs fördern, besitzen GAMs eine zentrale Rolle im Verständnis der Tumorbiologie. Eine der Schlüsselfunktionen von Mikroglia und Makrophagen ist die Phagozytose, deren Rolle im humanen GBM jedoch weitestgehend unklar ist. Ziel dieser Arbeit war die Etablierung eines Phagozytose-Assays unter Benutzung humaner organotypischer GBM-Schnittkulturen, anhand derer eine systematische Analyse dieser zentralen Funktion von Mikroglia und Markrophagen im Gliomkontext erfolgen sollte. Zudem wurde der Einfluss der in der Literatur beschriebenen phagozytose-relevanten Nukleotide ATP, UDP (Experiment 1) sowie des klinisch eingesetzten Chemotherapeutikums Temozolomid (Experiment 2) auf die Phagozytose von GAMs getestet. Methoden: Nach Anfertigung der Schnittkulturen wurden diese zusammen mit Polystyren-Partikeln als Phagozytose-Marker inkubiert. Die Auswertung erfolgte nach immunhistochemischer Anfärbung, konfokalmikroskopischer Aufnahme und 3D-Rekonstruktion der GAMs. Im Anschluss erfolgte eine computergestützte standardisierte quantitative Auswertung. Ergebnisse: In den humanen, organotypischen GBM-Schnittkulturen wurde die Phagozytose, bedingt durch eine geringe Eindringtiefe der Polystyren-Partikel, randständig untersucht. Dabei hatte weder 100μM ATP noch 100μM UDP einen signifikanten Einfluss auf die Phagozytose von GAMs. Überraschenderweise inhibierte Cytochalasin D ebenfalls die Phagozytose-Rate nicht signifikant. In Experiment 2 wurde gezeigt, dass Temozolomid in einer Konzentration bis 200μM die Phagozytose-Rate von GAMs nicht signifikant beeinflusste. In beiden Experimenten fiel eine ausgeprägte intertumorale Heterogenität der Phagozytose-Aktivität auf. Schlussfolgerungen: Die Ergebnisse zeigen, dass sowohl 100μM ATP und UDP als auch Temozolomid (bis zu einer Konzentration von 200μM) die Phagozytose von GAMs nicht relevant beeinflussen. Aufgrund einer potenziell enukleierenden Wirkung von Cytochalasin D auf GAMs, könnte möglicherweise die Kühlung mit Eis eine alternative Negativkontrolle für Phagozytose-Experimente an GAMs darstellen (Experiment 1 mit modifizierter Negativkontrolle). Für künftige Phagozytose-Experimente dürfte eine vorherige molekulargenetische Stratifizierung in GBM-Subtypen kombiniert mit einer höheren Tumorfallzahl und der vorherigen Bestimmung des intratumoralen Entnahmeorts wichtige Erkenntnisse zu tumorindividuellem Phagozytose-Verhalten von GAMs liefern.
Glioma-associated microglia and macrophages (GAMs) constitute an important cell population of glioblastoma multiforme (GBM). Promoting the progression and invasion of GBMs, GAMs play a pivotal role in tumour biology. Though phagocytosis represents one of the key functions in microglia and macrophages, the exact role of it in human glioblastoma remains almost unknown. This study pursued to establish a phagocytosis assay using human organotypic GBM slice cultures. Based on this experimental platform two experiments were performed: The effect on phagocytosis was analysed for a) UDP and ATP, two nucleotides which were reported previously in literature as relevant in the process of phagocytosis (experiment 1), and b) the chemotherapeutical Temozolomide, part of the current standard of care (experiment 2). Methods: Following to the preparation of organotypic GBM-slice cultures, slices were incubated with polystyrene particles as markers for phagocytosis. GAMs were stained immuno-histochemically, recorded by confocal microscopy and reconstructed digitally in 3D. Analysis was performed in a standardized way with an imaging software. Results: Phagocytosis was analysed in both experiments close to the surface, due to a low penetration depth of polystyrene particles into the organotypic slice cultures. Neither 100μM ATP nor 100μM UDP influenced phagocytosis of GAMs significantly. In the second experiment it was shown that temozolomide (up to a concentration of 200μM) had no significant effect on phagocytosis of GAMs. In both experiments intertumoral heterogeneity was remarkable. Conclusion: The results suggest that 100μM ATP as well as UDP and Temozolomid (up to 200μM) do not influence phagocytosis of GAMs substantially. Surprisingly, cytochalasin D did also not inhibit phagocytosis significantly. Ice cooled medium could serve as effective negative control, as cytochalasin D potentially enucleated many GAMs. Previous selection of intratumoral region, genetic determination of GBM subtypes and a higher n number of tumours could provide interesting more differential insights of GAM phagocytosis dependent on the individual tumour probe.
