The phytosteroid ecdysterone is classified as an anabolic agent and has been included on the monitoring list of the World Anti-Doping Agency since 2020. Therefore, the consumption of food rich in ecdysterone, such as quinoa and spinach, is the focus of a lively debate. Thus, the urinary excretion of ecdysterone and its metabolites in humans was investigated following quinoa consumption alone and in combination with spinach. Eight participants (four male and four female) were included, and they ingested 368 ± 61 g cooked quinoa alone and in combination with 809 ± 115 g spinach after a washout. Post-administration urines were analyzed by LC-MS/MS. After intake of both preparations, ecdysterone and two metabolites were excreted in the urine. The maximum concentration of ecdysterone ranged from 0.44 to 5.5 µg/mL after quinoa and from 0.34 to 4.1 µg/mL after quinoa with spinach. The total urinary excreted amount as parent drug plus metabolites was 2.61 ± 1.1% following quinoa intake and 1.7 ± 0.9% in combination with spinach. Significant differences were found in the total urinary excreted amount of ecdysterone, 14-deoxy-ecdysterone, and 14-deoxy-poststerone. Only small portions of ecdysterone from quinoa and the combination with spinach were excreted in the urine, suggesting that both quinoa and spinach are poor sources of ecdysterone in terms of bioavailability.
Weniger anzeigenEarly human habitation of the arid to semiarid Central Iranian Plateau was strongly connected to the availability of water and associated natural hazards, such as flooding and drought events. In this geoarchaeological study, we focus on the occupation at the prehistoric site of Ajor Pazi within the formerly active fluvial environment of the Varamin Plain. Through radiocarbon and luminescence-dated sediment cross-sections, we apply multivariate statistics to sedimentological characteristics of bulk samples collected during a rescue excavation in 2018. Based on facies interpretations, we differentiate depositional processes and present their implications for the environs of Ajor Pazi. Our results show evidence of settlement activities between 6.4 and 5.6 ka cal B.P. (4.4–3.6 ka cal. B.C.E.). Phases of reduced geomorphodynamics can be distinguished when soil-forming processes take place. Our findings provide insights into the hitherto scarcely explored Transitional Chalcolithic II period during which the site of Ajor Pazi emerged and began to shape its environs.
Weniger anzeigenSo far, evaluation of training initiatives for dairy farm employees has been limited to the reaction and learning level. The effect of training on dairy farm employees' behavior has not been studied yet. The objective of this study was to determine the effectiveness of online training to change employee behavior toward best-practice routines. An interactive training course related to udder health was developed in 5 modules in Spanish and English: (1) collecting an aseptic milk sample, (2) administering intramammary treatment, (3) deciding on salable milk, (4) treating a cow at dry-off with antibiotics, and (5) administering teat sealants. Participants navigated the modules at their own pace or watched a narrated video. Employees were assessed performing 2 procedures related to dry cow treatment (i.e., treating a cow at dry-off with antibiotics, administering internal teat sealant) by using an objective structured clinical examination. If possible, all employees were scored performing the procedure on 2 cows before and within 2 to 3 wk after the training was applied.
Weniger anzeigenDigital data have become a valuable resource for autocratic governments seeking to influence societal behaviours. The rise of social credit systems in China has garnered a great deal of attention, with some even referring to them as ‘Orwellian’ surveillance systems. This study expands on previous research that has found surprisingly high levels of acceptance of social credit systems in China to the Southeast Asian region. Through an online opinion survey conducted in Thailand, Indonesia, Malaysia and the Philippines, we discovered that citizens in these countries exhibit higher acceptance rates of social credit systems than opposition rates, although lower than those observed in China. Moreover, we find that acceptance rates would decline significantly if the technologies supporting these systems originated from China. By introducing an external technology acceptance model, we provide an explanation for these findings based on citizens' attitudes towards their domestic situation and their perceptions of China's potential benefits to their countries. Interestingly, most of the ‘China Threat’ perceptions do not translate into opposition against Chinese social credit system technologies, except for military risks. Instead, citizens' negative views are primarily influenced by specific technology-related risks. These findings contribute to the existing literature on the acceptance of government-run social credit systems and public perception in the context of international relations.

Yet, there is little evidence on whether it might influence how parents describe their children’s personality. To fill this gap, we used latent multitrait-multimethod (CTCM-1) models to examine familial socioeconomic background as possible predictor of parental perceiver effects regarding their offspring’s personality by contrasting parental assessments against teacher-reports. Study 1 (N = 5,798) investigated reports on elementary school students’ Big Five and Study 2 (N = 3,771) focused on school-related personality facets. Socioeconomic status predicted the parental report in both studies. Participation in high-culture arts incrementally predicted parental report over and above socioeconomic status. Specifically, parents with higher participation in high-culture arts rated their children in a more positive light than class teachers. These background specific perceiver effects might reflect both varying personality judgments or actual differences in behavior.
Weniger anzeigenBenthic graptolites (Graptolithina) were surprisingly common and diverse in the Miaolingian (Cambrian), but have rarely been described in detail. Encrusting and erect growing colonies already evolved and can be differentiated in early Miaolingian faunas. The Rhabdopleuridae with their encrusting colonies provide few fossils, but members of the erect growing, bushy colonies of the Dithecodendridae are more common, at least as fragments indicating considerable fragmentation and transport. In the Wuliuan, the benthic graptolites reached a considerable diversity at the genus level with at least 6 genera appearing in this interval. The most common taxon is the encrusting genus Sphenoecium with its robust colonies, showing a worldwide distribution. Most taxa, however, are known from few records and their biostratigraphical and palaeogeographical distribution cannot be established yet. Erroneously, the widely distributed Tarnagraptus with its conical thecae has often been misidentified as the Ordovician Mastigograptus, but differs considerably in its tubarium construction and both might not be closely related.
Weniger anzeigenMit dem Individualized Classroom Assessment Scoring System (inCLASS) liegt ein Beobachtungsinstrument vor, das zum Ziel hat, Interaktionen in Kitas auf Kindebene erfassen und in ihrer Qualität bestimmen zu können. Der Beitrag geht den Fragen nach, inwieweit das inCLASS diesem Anspruch nach Maßgabe gängiger Gütekriterien gerecht wird und welche Herausforderungen und Limitationen damit verbunden sind. Die Ausführungen stützen sich auf eigene Forschungserfahrungen mit dem Instrument. Über das inCLASS hinaus werden Bezüge zu weiteren Qualitätsmessverfahren (CLASS und KES) hergestellt. Im Ergebnis zeigt das inCLASS einige Stärken und Schwächen. Stärken liegen unter anderem in der forschungsbasierten Ausrichtung auf die Individualebene von Kindern, was es ermöglicht, Verläufe der kindlichen Interaktionsqualität längsschnittlich nachzuzeichnen. Limitationen und Herausforderungen betreffen insbesondere die Faktorenstruktur und die Konfundierung der Erfassung von kindlichen Interaktionen und Kompetenzen.
Weniger anzeigenTaking the example of conspiracy-related communication online as one form of contentious politics, this study examines the data collection challenges for multidimensional comparative research across platforms, time, and cultural embeddings. It compares the architectures and features relevant to data collection, access regimes, and use cultures for a set of digital platforms and communication venues. Differentiating between actor- and content-based strategies, this study discusses the potentials and limitations of these approaches, considering differences in platforms, temporal dynamics, and cultural embeddings as well as several layers of equivalence. The discussion highlights crucial insights into designing data collection strategies in multidimensional comparative studies.
Weniger anzeigenChromatin is a dynamic network that regulates genome organization and gene expression. Different types of chromatin regulators are highly conserved among Archaeplastida, including unicellular algae, while some chromatin genes are only present in land plant genomes. Here, we review recent advances in understanding the function of conserved chromatin factors in basal land plants and algae. We focus on the role of Polycomb-group genes which mediate H3K27me3-based silencing and play a role in balancing gene dosage and regulating haploid-to-diploid transitions by tissue-specific repression of the transcription factors KNOX and BELL in many representatives of the green lineage. Moreover, H3K27me3 predominantly occupies repetitive elements which can lead to their silencing in a unicellular alga and basal land plants, while it covers mostly protein-coding genes in higher land plants. In addition, we discuss the role of nuclear matrix constituent proteins as putative functional lamin analogs that are highly conserved among land plants and might have an ancestral function in stress response regulation. In summary, our review highlights the importance of studying chromatin regulation in a wide range of organisms in the Archaeplastida.
Weniger anzeigenThe objective of this study was to correlate movement assessed by a pyroelectric infrared sensor system in preweaning dairy calves with lying and standing time assessed by a 3D accelerometer considering the temperature-humidity index (THI). A total of 35 dairy calves (1–7 d of age) were enrolled in the study and 20 calves were included in the final analyses. The lying and standing time of the calves was monitored with a 3D accelerometer (Hobo Pendant G Data Logger, Onset Computer Corporation, USA), which was used as the gold standard reference. The infrared sensor monitoring system (IMS; Calf Monitoring System, Futuro Farming GmbH, Germany) was fixed to the fence of the calf hutch within the calf's reach. Temperature-humidity was monitored with 2 validated THI sensors inside and on outside of each calf hutch. Additionally, one THI sensor was located near the calf hutches. The observation period lasted 14 consecutive days. The average standing time assessed by the 3D accelerometer was 13.4 ± 12.7 (mean ± standard deviation) min/h and the average lying time was 46.6 (±12.7) min/h. The median (25th percentile; 75th percentile) number of movements measured by the IMS was 360 (60; 919) movements per hour. Number of movements per hour measured by the IMS was compared with data obtained with a validated 3D accelerometer. The Pearson correlation coefficient between both standing and lying time and the number of movements was r = 0.85 and r = −0.85, respectively. The Pearson correlation coefficients were only slightly influenced by THI (low THI [<68]: r = 0.86; medium THI [68–72]: r = 0.85; high THI [>72]: r = 0.81). Our data show that the number of movements of dairy calves measured by IMS were highly correlated with the chosen gold standard reference method. High THI slightly affects the measurement accuracy of IMS.
Weniger anzeigenWhat instruments of migration control is the current Mexican administration employing? Do these differ from those of past administrations? Past administrations have applied policies to stem the entrance and stay of irregularized migrants; however, president Andres Manuel Lopez Obrador (2018-2024) stated that his administration would provide jobs to migrants and respect their human rights. For that, his government designed a New Migration Policy. Drawing on secondary qualitative data, policy analysis, reports, and statistical data, this paper examines five actions the ongoing Mexican administration has performed to manage irregularized migration, and evaluates if the strategies correspond with the principles of the New Migration Policy. Findings show that the Mexican government has used new and long-standing bordering practices to contain irregularized migration, contrasting with the discourse of respect to migrants’ human rights. The actions performed have gone from “permissive” to repressive as the pressure from the US and the influx of migrants increased.
Weniger anzeigenHere we describe the novel, multi-point Comet Interceptor mission. It is dedicated to the exploration of a little-processed long-period comet, possibly entering the inner Solar System for the first time, or to encounter an interstellar object originating at another star. The objectives of the mission are to address the following questions: What are the surface composition, shape, morphology, and structure of the target object? What is the composition of the gas and dust in the coma, its connection to the nucleus, and the nature of its interaction with the solar wind? The mission was proposed to the European Space Agency in 2018, and formally adopted by the agency in June 2022, for launch in 2029 together with the Ariel mission. Comet Interceptor will take advantage of the opportunity presented by ESA's F-Class call for fast, flexible, low-cost missions to which it was proposed. The call required a launch to a halo orbit around the Sun-Earth L2 point. The mission can take advantage of this placement to wait for the discovery of a suitable comet reachable with its minimum Delta V capability of 600 ms(-1). Comet Interceptor will be unique in encountering and studying, at a nominal closest approach distance of 1000 km, a comet that represents a near-pristine sample of material from the formation of the Solar System. It will also add a capability that no previous cometary mission has had, which is to deploy two sub-probes - B1, provided by the Japanese space agency, JAXA, and B2 - that will follow different trajectories through the coma. While the main probe passes at a nominal 1000 km distance, probes B1 and B2 will follow different chords through the coma at distances of 850 km and 400 km, respectively. The result will be unique, simultaneous, spatially resolved information of the 3-dimensional properties of the target comet and its interaction with the space environment. We present the mission's science background leading to these objectives, as well as an overview of the scientific instruments, mission design, and schedule.
Weniger anzeigenContext. M-dwarf stars are the most common of potential exoplanet host stars in the Galaxy. It is therefore very important to understand planetary systems orbiting such stars and to determine the physical parameters of such planets with high precision. Also with the launch of the James Webb Space Telescope (JWST) the observation of atmospheric parameters of planets orbiting these stars has begun. It is therefore required to determine properties of potential targets.
Aims. Two planets around the red dwarf TOI-776 were detected by TESS. The objective of our study was to use transit observations obtained by the CHEOPS space mission to improve the current precision of the planetary radii, as well as additional radial velocity (RV) data in order to improve mass estimates of the two planets. Using these quantities, we wanted to derive the bulk densities of those planets, improving the precision in earlier results, and use this information to put them in context of other exoplanetary systems involving very low mass stars.
Methods. Utilizing new transit data from the CHEOPS satellite and its photometric telescope, we obtained very high precision planetary transit measurements. Interpretation of these provides updated planetary radii, along with other system parameters. A concurrent ESO large observing program using the high precision spectrograph HARPS has doubled the available radial velocity data. Calculating the power spectrum of a number of stellar activity indices we update the previously estimated stellar rotation period to a lower value.
Results. The CHEOPS data provide precise transit depths of 909 and 1177 ppm translating into radii of Rb = 1.798−0.077+0.078 R⊕ and Rc = 2.047−0.078+0.081 R⊕, respectively. Our interpretation of the radial velocities and activity indicator time series data estimates a stellar rotation period for this early M dwarf of ~21.1 days. A further multi-dimensional Gaussian process approach confirm this new estimate. By performing a Skew-Normal (SN) fit onto the Cross Correlation Functions we extracted the RV data and the activity indicators to estimate the planetary masses, obtaining Mb = 5.0−1.6+1.6 M⊕ and Mc = 6.9−2.5+2.6 M⊕.
Conclusions. We improve the precision in planetary radius for TOI-776 b and c by a factor of more than two. Our data and modelling give us parameters of both bodies consistent with mini-Neptunes, albeit with a relatively high density. The stellar activity of TOI-776 is found to have increased by a factor larger than 2 since the last set of observations.

WASP-76 b has been a recurrent subject of study since the detection of a signature in high-resolution transit spectroscopy data indicating an asymmetry between the two limbs of the planet. The existence of this asymmetric signature has been confirmed by multiple studies, but its physical origin is still under debate. In addition, it contrasts with the absence of asymmetry reported in the infrared (IR) phase curve.
Aims. We provide a more comprehensive dataset of WASP-76 b with the goal of drawing a complete view of the physical processes at work in this atmosphere. In particular, we attempt to reconcile visible high-resolution transit spectroscopy data and IR broadband phase curves.
Methods. We gathered 3 phase curves, 20 occultations, and 6 transits for WASP-76 b in the visible with the CHEOPS space telescope. We also report the analysis of three unpublished sectors observed by the TESS space telescope (also in the visible), which represents 34 phase curves.
Results. WASP-76 b displays an occultation of 260 ± 11 and 152 ± 10 ppm in TESS and CHEOPS bandpasses respectively. Depending on the composition assumed for the atmosphere and the data reduction used for the IR data, we derived geometric albedo estimates that range from 0.05 ± 0.023 to 0.146 ± 0.013 and from <0.13 to 0.189 ± 0.017 in the CHEOPS and TESS bandpasses, respectively. As expected from the IR phase curves, a low-order model of the phase curves does not yield any detectable asymmetry in the visible either. However, an empirical model allowing for sharper phase curve variations offers a hint of a flux excess before the occultation, with an amplitude of ~40 ppm, an orbital offset of ~ −30°, and a width of ~20º. We also constrained the orbital eccentricity of WASP-76 b to a value lower than 0.0067, with a 99.7% confidence level. This result contradicts earlier proposed scenarios aimed at explaining the asymmetry observed in high-resolution transit spectroscopy.
Conclusions. In light of these findings, we hypothesise that WASP-76 b could have night-side clouds that extend predominantly towards its eastern limb. At this limb, the clouds would be associated with spherical droplets or spherically shaped aerosols of an unknown species, which would be responsible for a glory effect in the visible phase curves.
Weniger anzeigenAcid ceramidase (Ac) is a lysosomal enzyme catalyzing the generation of sphingosine from ceramide, and Ac inhibitors are currently being investigated as potential cancer therapeutics. Yet, the role of the Ac in immune responses, particularly anti-viral immunity, is not fully understood. To investigate the impact of Ac expression on various leukocyte populations, we generated a tamoxifen-inducible global knockout mouse model for the Ac (iAc-KO). Following tamoxifen administration to healthy mice, we extracted primary and secondary lymphoid organs from iAc-KO and wild-type (wt) littermates and subsequently performed extensive flow cytometric marker analysis. In addition, we isolated CD4 + T cells from the spleen and lymph nodes for sphingolipid profiling and restimulated them in vitro with Dynabeads™ Mouse T-activator CD3/CD28. Intracellular cytokine expression (FACS staining) was analyzed and secreted cytokines detected in supernatants. To study cell-intrinsic effects, we established an in vitro model for iAc-KO in isolated CD4 + T and B cells. For CD4 + T cells of iAc-KO versus wt mice, we observed reduced Ac activity, an increased ceramide level, and enhanced secretion of IFNγ upon CD3/CD28 costimulation. Moreover, there was a marked reduction in B cell and plasma cell and blast numbers in iAc-KO compared to wt mice. To study cell-intrinsic effects and in line with the 3R principles, we established in vitro cell culture systems for iAc-KO in isolated B and CD4 + T cells. Our findings pinpoint to a key role of the Ac in mature B and antibody-secreting cells and in IFNγ secretion by CD4 + T cells.
Weniger anzeigenThe use of taboo words represents one of the most common and arguably universal linguistic behaviors, fulfilling a wide range of psychological and social functions. However, in the scientific literature, taboo language is poorly characterized, and how it is realized in different languages and populations remains largely unexplored. Here we provide a database of taboo words, collected from different linguistic communities (Study 1, N = 1046), along with their speaker-centered semantic characterization (Study 2, N = 455 for each of six rating dimensions), covering 13 languages and 17 countries from all five permanently inhabited continents. Our results show that, in all languages, taboo words are mainly characterized by extremely low valence and high arousal, and very low written frequency. However, a significant amount of cross-country variability in words’ tabooness and offensiveness proves the importance of community-specific sociocultural knowledge in the study of taboo language.
Weniger anzeigenA change of orbital state alters the coupling between ions and their surroundings drastically. Orbital excitations are hence key to understand and control interaction of ions. Rare-earth elements with strong magneto-crystalline anisotropy (MCA) are important ingredients for magnetic devices. Thus, control of their localized 4f magnetic moments and anisotropy is one major challenge in ultrafast spin physics. With time-resolved x-ray absorption and resonant inelastic scattering experiments, we show for Tb metal that 4f-electronic excitations out of the ground-state multiplet occur after optical pumping. These excitations are driven by inelastic 5d-4f-electron scattering, altering the 4f-orbital state and consequently the MCA with important implications for magnetization dynamics in 4f-metals and more general for the excitation of localized electronic states in correlated materials.
Weniger anzeigenInsect monitoring is pivotal for assessing biodiversity and informing conservation strategies. This study delves into the complex realm of insect monitoring in the Global South—world developing and least-developed countries as identified by the United Nations Conference on Trade and Development—highlighting challenges and proposing strategic solutions. An analysis of publications from 1990 to 2024 reveals an imbalance in research contributions between the Global North and South, highlighting disparities in entomological research and the scarcity of taxonomic expertise in the Global South. We discuss the socio-economic factors that exacerbate the issues, including funding disparities, challenges in collaboration, infrastructure deficits, information technology obstacles and the impact of local currency devaluation. In addition, we emphasize the crucial role of environmental factors in shaping insect diversity, particularly in tropical regions facing multiple challenges including climate change, urbanization, pollution and various anthropogenic activities. We also stress the need for entomologists to advocate for ecosystem services provided by insects in addressing environmental issues. To enhance monitoring capacity, we propose strategies such as community engagement, outreach programmes and cultural activities to instill biodiversity appreciation. Further, language inclusivity and social media use are emphasized for effective communication. More collaborations with Global North counterparts, particularly in areas of molecular biology and remote sensing, are suggested for technological advancements. In conclusion, advocating for these strategies—global collaborations, a diverse entomological community and the integration of transverse disciplines—aims to address challenges and foster inclusive, sustainable insect monitoring in the Global South, contributing significantly to biodiversity conservation and overall ecosystem health.
Weniger anzeigenRecognition of mitotic figures in histologic tumor specimens is highly relevant to patient outcome assessment. This task is challenging for algorithms and human experts alike, with deterioration of algorithmic performance under shifts in image representations. Considerable covariate shifts occur when assessment is performed on different tumor types, images are acquired using different digitization devices, or specimens are produced in different laboratories. This observation motivated the inception of the 2022 challenge on MItosis Domain Generalization (MIDOG 2022). The challenge provided annotated histologic tumor images from six different domains and evaluated the algorithmic approaches for mitotic figure detection provided by nine challenge participants on ten independent domains. Ground truth for mitotic figure detection was established in two ways: a three-expert majority vote and an independent, immunohistochemistry-assisted set of labels. This work represents an overview of the challenge tasks, the algorithmic strategies employed by the participants, and potential factors contributing to their success. With an score of 0.764 for the top-performing team, we summarize that domain generalization across various tumor domains is possible with today’s deep learning-based recognition pipelines. However, we also found that domain characteristics not present in the training set (feline as new species, spindle cell shape as new morphology and a new scanner) led to small but significant decreases in performance. When assessed against the immunohistochemistry-assisted reference standard, all methods resulted in reduced recall scores, with only minor changes in the order of participants in the ranking.
Weniger anzeigenThe transition from a humid green Sahara to today’s hyperarid conditions in northern Africa ~5.5 thousand years ago shows the dramatic environmental change to which human societies were exposed and had to adapt to. In this work, we show that in the 620,000-year environmental record from the Chew Bahir basin in the southern Ethiopian Rift, with its decadal resolution, this one thousand year long transition is particularly well documented, along with 20–80 year long droughts, recurring every ~160 years, as possible early warnings. Together with events of extreme wetness at the end of the transition, these droughts form a pronounced climate “flickering”, which can be simulated in climate models and is also present in earlier climate transitions in the Chew Bahir environmental record, indicating that transitions with flickering are characteristic of this region.
