dc.description.abstract
Die photosynthetische Wasseroxidation des Mn4Ca-Komplexes im Photosystem II
(PSII) ist für die Atmosphäre und das Leben auf der Erde von fundamentaler
Bedeutung. Jedoch ist dieser Prozess nur unzureichend verstanden. Mit Hilfe
von Röntgenabsorptionsspektroskopie (X-ray Absorption Spectroscopy, XAS) zur
Untersuchung der Struktur des Mn4Ca-Komplexes und insbesondere der
strukturellen Veränderungen im katalytischen Zyklus wurden wichtige Resultate
erzielt um den Mechanismus der photosynthetischen Wasseroxidation zu
enträtseln. In der hier vorliegenden Arbeit werden zwei spezielle Beiträge
dazu vorgestellt. 1\. Raumtemperatur- und zeitaufgelöste XAS-Messungen zu
Intermediatzuständen des katalytischen Zyklus Durch präparative Untersuchungen
konnte in PSII-Membranfragmenten ein minimaler Wassergehalt auf 50 Gewichts-%
erzielt werden, der für ein optimales Signal-zu-Rausch-Verhältnis erforderlich
ist. Durch die Nutzung eines elektronischen Dosiersystems konnte außerdem die
automatisierte, schnelle und reproduzierbare Auftragung von PSII-Suspension
auf einen speziell entworfenen Probenträger erreicht werden. Ferner wurde eine
Apparatur für den automatisierten Probenwechsel während der XAS-Messungen
entwickelt. Damit war es möglich, einzelne Proben mit Laserblitzsequenzen zu
belichten und simultan XAS-Daten aufzunehmen. Nur mit der Hilfe dieser
verbesserten Methodik wurde eine Reihe von Untersuchungen am katalytischen
Zyklus des PSII-Komplexes möglich. Dadurch konnten Geschwindigkeitskonstanten
und Temperaturabhängigkeit der durch Röntgenstrahlung induzierten
Photoreduktion des Mn4Ca-Komplexes ermittelt werden. Außerdem wurde zum ersten
Mal die Struktur und die Oxidationszustände des Mn4Ca-Komplexes bei
Raumtemperatur (funktionale und quasi-native Bedingungen) für alle vier
semistabilen Intermediatzustände charakterisiert und mit Daten, die bei 20 K
ermittelt wurden, verglichen. Die zeitaufgelösten XAS-Messungen mit einer
Auflösung von 10 μs ermöglichten die Entdeckung und Identifizierung eines
neuen Reaktionszwischenzustandes. Diese Ergebnisse führten zu einer
Erweiterung des fundamentalen S-Zyklus und sind von wesentlicher Bedeutung für
den Entwicklung mechanistischer Modelle zur photosynthetischen
Wasseroxidation. 2\. XAS an der Ca-K-Kante zur Bestimmung der Position und der
Koordination von Calcium Durch die Optimierung der Prozedur zur Entfernung von
überschüssigen Calcium konnte der Ca2+-Gehalt auf weniger als 2 - 3 Ca/PSII
reduzieren werden, ohne signifikanten Verlust der Sauerstoffaktivität. Trotz
der experimentellen Schwierigkeiten konnten Ca-XAS-Spektren aufgenommen und
analysiert werden, und zwar für PSII-Proben im S1-Zustand. Es wurden außerdem
erste Versuche zur Charakterisierung des S2-Zustandes unternommen. Es zeigte
sich, dass das Ca2+-Ion sich ein einem Abstand von ca. 3,3 Å zu mindestens
einem Manganatom befindet. Dieses Ergebnis ist in guter Übereinstimmung mit
den neusten kristallographischen Daten und mit den XAS-Ergebnissen aus anderen
Gruppen.

dc.description.abstract
Photosynthetic water oxidation at the Mn4Ca-cluster of Photosystem II (PSII),
a process of fundamental importance for the atmosphere and biosphere on the
earth, is only insuffciently understood. X-ray absorption spectroscopy (XAS)
on the structure of the Mn4Ca-cluster and, in particular, on the structural
changes in the catalytic cycle has resulted in central contributions to
unravel the mechanism of photosynthetic water oxidation. In the presented
thesis two specific contributions are described. 1\. Room-temperature and
time-resolved XAS on intermediates in the catalytic cycle Through preperative
investigations a minimal water content of PSII membrane particles could
possible down to 50 % (weight per weight). This is requered for an optimal
signal-to-noise ratio. Using a electronic dispensing system the automated
preparation of PSII samples on specifically designed samples stripes has been
achieved. An apparatus for automated sample change during the XAS measurements
has been developed that facilitated Laser-flash application and XAS
measurements on up to 10,000 PSII samples per beamtime period. Only by means
of this improved methodology, a series of investigation on the Mn4Ca-cluster
in its catalytic cycle became possible. Thereby the rate and temperature
dependence of the X-ray induced reduction of the Mn4Ca-cluster was thoroughly
characterized. And for the first time, structure and oxidation state of the
Mn4Ca-cluster at room temperature (functional and quasi-native conditions) has
been characterized for all four semi-stable intermediate states of the S-state
cycle and compared to data collected at 20 K. Time-resolved XAS measurements
at 10 μs resolution facilitated discovery and identification of a novel
reaction intermediate. The results lead to an extension of the fundamental
S-state cycle and bear important mechanistic implications. 2\. XAS at the Ca
K-edge on the position of Ca coordination environment By optimization of a
procedure for removal of excessive calcium, the Ca2+ content could be reduced
down to a level of 2 - 3 Ca per PSII, without significant activity loss. In
spite of the experimental difficulties, it became possible to collect and
analyze XAS data for PSII samples in the S1-state. First attempts to
characterize the S2-state were made. One result is, that the calcium is at a
distance of ca. 3.3 Å to more than one manganese ion. This result is in good
agreement with recent crystallographic data and with the XAS results of other
research groups.
