Der Gastrointestinaltrakt dient dem HI-Virus als Eintrittspforte. HIV-
Patienten leiden häufig an Diarrhoe. Bei HIV-Patienten kommt es zur Erhöhung
bestimmter Zytokine, die an der Entstehung dieser Diarrhoe beteiligt sein
könnten. Daher wurde der Einfluss der HIV-typischen Zytokine TNF-alpha, IFN-
alpha, IFN-gamma und IL-1-beta auf die Restitution von Einzellläsionen mittels
Conductance scanning untersucht. Alle Zytokine gemeinsam bewirkten eine
signifikante Verzögerung der Restitution von Einzelzellläsionen von 21 %
gegenüber Kontrollmessungen. Jedes Zytokin für sich alleine hatte keinen
signifikanten Effekt auf die Restitution, sodass von einem synergistischen
Geschehen ausgegangen wird. Dieses Ergebnis weist darauf hin, dass das
verä¤nderte Zytokinmuster bei HIV-Infektion für die Entstehung der Diarrhoe
mitverantwortlich ist, weil durch entstehende Lecks Solute ins Lumen strömen
und Wasser binden. Eine Untersuchung der an der Reparatur beteiligten Proteine
mittels Immunfluoreszenz und konfokaler Fluoreszenzmikroskopie zeigte, dass
der Wiedereinbau des Tight junction-Proteins Occludin bereits 10 min nach
Setzen einer Einzelzellläsion einsetzte. Das Zellahahesionsmolekül E-Cadherin
konnte zu diesem Zeitpunkt nicht nachgewiesen werden. Es wurde
geschlussfolgert, dass Occludin im Gegensatz zu E-Cadherin an der
Frührestitution von Einzelzellläsionen beteiligt ist.

dc.description.abstract
The HI-virus may enter the body through the gastrointestinal system. HIV-
patients often suffer from diarrhoea. HIV positve patients manifest a release
of the cytokines TNF-alpha, IFN-alpha, IFN-gamma and IL-1-beta. In the present
study the conductance scanning technique was employed to investigate effects
of these cytokines on the restitution of single cell lesions in epithelial
cell layers. A combination of all HIV-typical cytokines caused a statistically
significant delay in the restitution of single cell lesions of 21%, compared
with control measurements. Single cytokines did not result in a significant
deceleration of restitution. Therefore, a synergistic effect was assumed.
These results indicate, that the induced cytokine pattern of HIV-patients may
be responsible for the mechanism of diarrhoea, because the resulting leakage
of solutes binds water in the intestinal lumen. Moreover, investigation of
proteins involved in restitution by immunofluorescence staining and confocal
fluorescence microscopy indicated a reappearance of the tight junction protein
occludin 10 minutes after inducing a single-cell-lesion. At that time the
cell-adhesion molecule E-cadherin was not detectable. It was concluded that
occludin participates in the early restitution of single cell lesions whereas
E-cadherine contributes to junction formation at a later stage of restitution.
