Light is essential for photosynthetic oxygen evolution not only as a source of energy to oxidize water, but also as a stimulus to activate the system for oxygen evolution. The water oxidizing complex of chloroplast photosystem II (PSII) is located on the lumenal side of the thylakoid membrane, where it performs the decomposition of water into molecular oxygen, protons and electrons. The active site is composed of a Mn4Ca cluster that can accumulate four oxidizing redox equivalents. These are neutralized through the oxidation of two water molecules. The precise organization of the Mn4Ca cluster and its association with the PSII polypeptides are still under debate. The present work includes the results of recent experimental studies on disassembly and reassembly of the Manganese complex (Mn complex) in PSII and their biological implications. Combining a variety of biochemical and biophysical methods enabled me to examine in detail the intermediate states of both processes. By employing a trigger event characterized by a stepwise increase in temperature (temperature jump to 47°C) it was possible to initiate a relatively slow and stepwise disassembly of the Mn complex of PSII, involving several distinct intermediates. By combining oxygen polarography, X-ray absorption spectroscopy (XAS) at the Mn K-edge, electron paramagnetic resonance (EPR), atomic absorption spectroscopy (AAS) and recombination chlorophyll fluorescence techniques, it was shown that the heat-induced disassembly process involves at least three steps. (1) The first most rapid step occurs within ~5 min and is coupled to the release of the extrinsic protein with 18 kDa molecular weight from PSII; circumstantial evidence suggested that concomitantly the affinity of the essential Ca2+ ion which is bound at the Mn complex is lowered, leading to the observed inactivation of oxygen evolution. (2) In a second step, two Mn ions are reduced to MnII and released from their binding sites into the bulk within ~15 min; the remaining binuclear complex contains two MnIII ions connected by a di-µ-oxo bridge. (3) The third step occurs only within hours and leads to the reduction and liberation of the remaining two Mn ions as MnII whereby apo-PSII with an empty Mn/Ca site is formed. The second part of this work includes the results obtained on the photoassembly process of the Mn4Ca complex. The complex spontaneously is assembled in the light; a process denoted as photoactivation. Recovery of oxygen evolution activity after photoactivation was monitored by O2-polarography and delayed chlorophyll fluorescence measurements, starting from various states as obtained by the previous heat treatment. Oxidation state and structural features of the intermediates of the Mn complex during photoactivation were assayed by X-ray absorption spectroscopy at the Mn K edge. The MnIII2 complex obtained after ~15 min of heating and apo-PSII with an empty Mn/Ca site present after 2 h of heating effectively could be photoactivated to form the O2-evolving tetra-Mn complex within ~40 min in the presence of stoichiometric amounts of two and four respectively exogenous MnII ions per PSII. Starting with apo-PSII, a lag-phase of about 5 min duration was detected in the recovery of O2 activity during photoactivation. Within this lag phase a similar semi-stable MnIII2 complex is created as obtained after ~15 min of heating. Therefore, the MnIII2(di-µ-oxo) state is a key intermediate both in the assembly and disassembly of the Mn complex.
Licht ist essentiell sowohl als Energieressource für die photosynthesische Wasseroxidation als auch als Stimulus für die Aktivierung des Sauerstoffbildungssystems. Der Wasseroxidationskomplex des Photosystems II (PSII) auf der Lumenseite der Thylakoidmembran im Chloroplasten spaltet Wasser in molekularen Sauerstoff, Protonen und Elektronen. Der Mn4Ca-Komplex des aktiven Zentrums kann vier oxidierende Redoxäquivalente akkumulieren, die simultan durch die Oxidation zweier Wassermoleküle neutralisiert werden. Der genaue Aufbau des Mn4Ca- Komplexes und seine Assoziierung mit den PSII Polypeptiden werden noch kontrovers diskutiert. Die vorliegende Arbeit beinhaltet die Ergebnisse experimenteller Untersuchungen zur Disassemblierung und zur Assemblierung des Mangankomplexes (Mn Komplex) im PS II. Die Kombination vielfältiger biochemischer und biophysikalischer Methoden ermöglichte detaillierte Untersuchungen der intermediären Stadien beider Prozesse. Durch die Anwendung einer Temperaturerhöhung (Temperatursprung auf 47°) wurde es möglich, die relative langsame schrittweise Zerlegung des Mn Komplexes im PS II zu studieren, wobei verschiedene Intermediatzustände auftreten. Zunächst wurden die Inaktivierung und der schrittweise Abbau des Mn-Komplexes durch Erwärmung auf 47°C für ansteigende Zeitintervalle untersucht. Die Wärmebehandlung wurde gewählt, um den normalerweise langsamen Abbauprozess des Mn Komplexes zu beschleunigen und klar vom Abbauprozessen der PSII-Proteine abzugrenzen. Durch die kombinierte Anwendung von Polarographie, Röntgenabsorptionsspektroskopie (XAS) an der Mn K-Kante, Paragmanetisches Elektronen resonanz (EPR), Atomabsorptionsspektroskopie (AAS) und Rekombinations-Chlorophyll-Fluoreszenztechniken konnte gezeigt werden, dass beim wärmeinduzierten Abbau mindestens drei Schritte ablaufen. (1) Der schnellste Schritt ereignet sich innerhalb von fünf Minuten und ist an die Freisetzung des extrinsischen 18 kDa Proteins des PSII gekoppelt. Die vorliegenden Ergebnisse legen nahe, dass gleichzeitig die Affinität des essentiellen Ca2+ Ions am Mn Komplex reduziert wird und zu der beobachteten Inaktivierung der Sauerstoffbildung führt. (2) In einem zweiten Schritt, werden innerhalb von fünfzehn Minuten zwei Mn Ionen zu MnII reduziert und aus ihrer Bindung gelöst: der verbleibende binukleare Komplex beinhaltet zwei über eine di-µ-oxo Brücke verbundene MnIII Ionen. (3) Der dritte Schritt erfolgt erst nach Stunden und führt zur Reduktion und Freisetzung der verbleibenden beiden Mn Ionen als MnII, wobei sich der apo-PSII mit einer leeren Mn/Ca Bindungsstelle bildet. Der zweite Teil dieser Arbeit beinhaltet die Ergebnisse zur Assemblierung des Mangankomplexes. Der Aufbau erfolgt im Licht durch die sogenannte Photoaktivierung. Ausgehend von den durch Temperatursprung erhaltenen Intermediaten wurde die Wiederherstellung der Sauerstoffbildungsaktivität nach Photoaktivierung durch Sauerstoffpolarographie und Messungen der verzögerten Fluoreszenz beobachtet. Der Oxidationszustand und strukturelle Eigenschaften der Intermediate des Mn Komplexes wurden mit Röntgenabsorptionsspektroskopie an der Mn K-Kante untersucht. Sowohl der nach ca. fünfzehn Minuten Wärmebehandlung erzeugte MnIII2 Komplex als auch das nach zweistündiger Wärmebehandlung erzeugte apo- PSII mit leerer Mn/Ca Bindungsstelle konnten zum sauerstoffbildenden Mn Komplex innerhalb von ca. vierzig Minuten bei Anwesenheit stoichiometrischer Mengen von zwei bzw. vier exogenen MnII Ionen pro PSII photoaktiviert werden. Ausgehend vom apo-PSII wurde eine Verzögerung von ca. fünf Minuten bis zur Wiederherstellung der Sauerstoffbildungsaktivität durch Photoaktivierung beobachtet. In dieser Verzögerungsphase wurde ein semi-stabiler MnIII2 Komplex gebildet. Ein ähnlicher Komplex entsteht auch nach ca. fünfzehn Minuten Wärmebehandlung. Das MnIII2(di-µ-oxo) Stadium ist also ein Schlüsselintermediat sowohl im Abbau- als auch im Photoaktivierungsprozess des Mangankomplexes.
