Die CLCA-Genfamilie mutmaßlicher Ca2+-aktivierter Cl--Kanäle zählt zu den
eventuellen modulatorischen Genen, die maßgeblich den Phänotyp der Zystischen
Fibrose beeinflussen. Aufgrund der bisherigen Erkenntnisse der Zystischen-
Fibrose-Forschung weisen zahlreiche Beispiele auf eine zentrale Bedeutung von
modifizierenden Genen bei der Pathogenese der Erkrankung hin, die außerhalb
des cftr-Locus gelegen sind. Anhand elektrophysiologischer Daten, mRNA-
Expressionsanalysen und kürzlich veröffentlichter Allelvariationsstudien mit
Mikrosatellitenmarkern ergeben sich zunehmend Hinweise, dass ein Teil dieser
modifi-zierenden Einflüsse von Mitgliedern der CLCA-Genmilie, möglicherweise
durch die Vermittlung einer alternativen, Ca2+-aktivierter Cl--Leitfähigkeit,
mediiert werden könnte. In der vorliegenden Arbeit wurde die mRNA-
Expressionsrate der murinen mCLCA1- , mCLCA2- , mCLCA3- und mCLCA4-Homologen
mittels quantitativer real-time RT-PCR im Dünndarm von CF-
(cftrTgH(neoim)1Hgu, cftrtm1Cam) und WT-BALB/cJ-, -C57BL/6J-, -DBA/2J- und
-NMRI-Mäusen quantifiziert. Dabei wurden große Unterschiede der mRNA-
Expressions-raten aller vier CLCA-Homologen zwischen den vier WT-Mausstämmen
detektiert. Die mRNA-Expressionsraten von mCLCA1 und mCLCA4 waren bei CF- und
WT-Mausgruppen ähnlich hoch. Dagegen war die mCLCA3-mRNA-Expressionsrate bei
allen CF-Mausmo-dellen deutlich erhöht. Die immunhistochemische Detektion von
mCLCA3-Protein und die Identifizierung von Becherzellen mittels der PAS-
Reaktion zeigten ähnlich starke Zunahmen an mCLCA3-positiven Becherzellen bei
den CF-Mausmodellen. Deswegen kann mit großer Wahrscheinlichkeit angenommen
werden, dass die erhöhte mCLCA3-mRNA-Expression eine Folge der
Becherzellhyperplasie war und nicht auf Regulationsmechanismen auf dem
Transkriptionslevel beruhten. Die beobachteten Zunahmen an mCLCA2-mRNA-
Kopienzah-len bei den CF-Mausmodellen dagegen scheinen tatsächlich auf einer
hochregulierten Transskription zu beruhen. So waren Veränderungen der mRNA-
Kopienzahlen nicht mit veränderten Zellkinetiken des Darmepithels assoziiert,
wie mit Hilfe einer immunhistochemi- schen Detektion des phospho-
Histon3-Proteins und des aktivierte-Caspase3-Proteins bestätigt wurde.
Insgesamt kann aufgrund dieser Ergebnisse geschlossen werden, dass mCLCA2 und
mCLCA3 potentiell durch ihre verstärkte Expression als modifizierende Gene des
Phänotyps von CF-Mausmodellen im Darm in Frage kommen. Mit Hilfe der
Sequenzierung des offenen Leserahmens von mCLCA3 konnte keine Allel-variation
im mclca3-Gen, zumindest nicht in der kodierenden Region und bei diesen vier
Mausstämmen, festgestellt werden, die mit diesen Expressionsunterschieden in
Zusam-menhang zu bringen gewesen wären.

Members of the family of calcium-activated chloride channels (CLCA) have been
implicated as modulators of the phenotype in cystic fibrosis (CF). Results of
several different studies have shown that CF disease severity is modulated by
other genetic factors than CFTR. Electrophysiological, mRNA-expression and
allelic variation studies seem to imply that, at least in part, members of the
CLCA gene family could be responsible for this modulation by mediating an
alternative calcium-activated chloride conductance. Here, the expression
levels of the murine mCLCA1, mCLCA2, mCLCA3 and mCLCA4 were quantified by
real-time RT-PCR in the small intestines of CF (cftrTgH(neoim)1Hgu,
cftrtm1Cam) and wild type BALB/cJ, C57BL/6J, DBA/2J und NMRI mice. Markedly
different expression levels of all four CLCA-homologs were observed between
the different wild type strains. Expression of mCLCA1 and mCLCA4 was similar
in CF versus wild type mice. In contrast, mCLCA3 mRNA copy numbers were
increased up to three-fold in all CF models. Immunohistochemichal detection of
mCLCA3- and PAS-positive cells on consecutive tissue sections identified a
similar increase in mCLCA3-expressing goblet cells, suggesting that elevated
mRNA copy numbers of mCLCA3 are due to goblet cell hyperplasia rather than
transcriptional regulatory events. Increased mCLCA2 mRNA copy numbers,
however, were considered more likely to be due to transcriptional
upregulation. Changes in mRNA copy numbers were not associated with altered
cell kinetics as determined by immunohistochemistry using antibodies to
phospho-histone 3 and activated caspase 3. The results suggest that both
mCLCA2 and mCLCA3 may act as modifiers of the intestinal phenotype in CF.
Sequence analysis of the open reading frame of mCLCA3 failed to identify
allelic variations in the coding region between the four different mouse
strains and thus variations in coding sequences were not associated with
differences in mCLCA-expression levels.
