Die Prävalenz der Infektion mit Toxoplasma gondii steigt beim Menschen
proportional mit dem Lebensalter um etwa ein Prozent pro Jahr an. In
Deutschland weisen im Durchschnitt 60-70% der erwachsenen Bevölkerung
Antikörper gegen T. gondii auf. Die Standardtherapie aller Verlaufsformen der
Toxoplasmose besteht aus der Kombination von Pyrimethamin und Sulfadiazin,
obwohl diese Therapie von schwerwiegenden Nebenwirkungen begleitet wird. In
bis zu 50% der Fälle werden allergische Reaktionen beobachtet, die zum
Absetzen bzw. Umstellen der Therapie führen. Auch die Teratogenität dieser
Therapie stellt in der Frühschwangerschaft ein Problem dar. Die mitochondriale
Atmungskette von Toxoplasmen weist eine Komponente auf, die in Säugerzellen
nicht vorkommt und deshalb einen vielversprechenden Angriffspunkt für
Antiparasitika darstellt: T. gondii besitzt anstelle des Komplexes I eine
alternative NADH-Dehydrogenase. Die Substanz
1-hydroxy-2-dodecyl-4(1H)-Quinolon (HDQ) wurde in Enzymassays als hoch affiner
Inhibitor der alternativen NADH-Dehydrogenase des Hefepilzes Yarrowia
lipolytica beschrieben. Die vorliegende Arbeit hatte zum Ziel, die
Wirkungsweise von neuen Hydroxyquinolonen in verschiedenen Modellen der T
.-gondii-Infektion zu untersuchen. Der Austausch eines einzelnen
Wasserstoffatoms durch eine Methylgruppe an Position C-3 des HDQ bei Substanz
A sowie der Austausch einer Dodecyl- und Methylgruppe des HDQ bei Substanz B
resultierten in einer deutlich gesteigerten Wirksamkeit gegen den Parasiten im
Vergleich zu HDQ in einem akuten Modell der Infektion sowie in einem Modell
der Reaktivierungstoxoplasmose. Die Substanz B erschien in den vorliegenden
Untersuchungen der Substanz A teilweise überlegen. Der therapeutische Effekt
war dem der Standardtherapie mit Pyrimethamin und Sulfadiazin ähnlich, jedoch
geringer als der des Atovaquon. Verbesserungen der Pharmakokinetik durch
modifizierte Präparationen der Substanzen könnten die Wirksamkeit weiter
steigern. Zusammenfassend verdeutlicht diese Arbeit, dass die neuen Quinolone,
insbesondere Substanz A und Substanz B, effektiv gegen T. gondii in klinisch
relevanten Mausmodellen sind.

dc.description.abstract
The prevalence of the infection with Toxoplasma gondii in humans increases
proportionally with age by about one percent per year. In Germany, on average,
60-70% of the adult population is positive for antibodies against T. gondii.
The standard course of therapy of all forms of toxoplasmosis is a combination
of pyrimethamine and sulfadiazine, although this therapy is accompanied by
serious side effects. In up to 50% of all cases, allergic reactions are
observed, leading to discontinuation or change of the therapy. The
teratogenicity of this therapy in early pregnancy is also a problem. The
mitochondrial respiratory chain of T. gondii has a component that does not
occur in mammalian cells and therefore represents a promising target for anti-
parasitics: T. gondii possess an alternative NADH dehydrogenase instead of the
complex I. The substance 1-hydroxy-2-dodecyl-4(1H)-quinolone (HDQ) was
described in enzyme assays as high-affinity inhibitor of alternative NADH
dehydrogenase of the yeast Yarrowia lipolytica. The present study has the goal
to prove the effect of new hydroxyquinolones in two different models of T.
gondii infection. The replacement of a single hydrogen atom by a methyl group
at position C-3 of HDQ by substance A and the replacement of a methyl and
dodecyl group of HDQ by substance B has resulted in a significantly enhanced
efficacy of these substances against the parasite in comparison to HDQ. This
study demonstrates that the new quinolones, especially substance A and
substance B, are effective against T. gondii infection, both in an acute model
and in a reactivation model of the infection.
