Künstliche Nukleasen stellen ein interessantes Werkzeug für biochemische
Anwendungen dar, wobei als Target der Träger der Erbinformationen in den
Zellen, die Desoxyribonukleinsäure (DNA), dient. Mithilfe solcher Nukleasen
können DNA-Moleküle gespalten werden, was u. a. in der Entwicklung von neuen
Chemotherapeutika von Bedeutung ist. Sigman und Mitarbeiter entdeckten bereits
Ende der 1970er Jahre die oxidative Spaltung von Nukleinsäuren durch
Bis-(1,10-phenanthrolin)kupfer(I) {[Cu(phen)2]+}, der ersten Kupfer-basierten
künstlichen Nuklease. Seitdem beschäftigten sich zahlreiche Arbeiten mit
diesem System, um die Mechanismen der DNA-Spaltung aufzuklären und/oder die
Nukleaseaktivität zu verbessern. Keine dieser Arbeiten beinhaltete jedoch den
Einfluss von Fluorierungen des Liganden auf die Bioaktivität, trotz der
verbreiteten Anwendung von Fluor- und Trifluormethylsubstituenten zur
Verbesserung der pharmakologischen Eigenschaften von bioaktiven Molekülen. Im
Rahmen dieser Arbeit wurde nun der Effekt der Einführung von fluorhaltigen
Substituenten am phen-Gerüst auf die Nukleaseaktivität und die DNA-
Bindungseigenschaften der jeweiligen Kupferkomplexe untersucht. Zunächst
wurden verschiedene fluorhaltige phen-Derivate mithilfe einer modifizierten
Skraup-Reaktion dargestellt. Es konnten fünf Liganden der Zusammensetzung
5-X-phen (X = F, CF3,OCF3, SCF3, SF5) und 5,6-Difluorphen synthetisiert
werden, wobei vier der Derivate (X = CF3, SCF3, OCF3, SF5) zum ersten Mal
dargestellt wurden. Die Liganden konnten mit Kupfer(II)nitrat zu Komplexen mit
der Grundstruktur [Cu(Xphen)2]2+ (X = F, F2, CF3, SCF3, SF5) umgesetzt werden.
Die Komplexe zeigten in Gegenwart von Ascorbinsäure als Reduktionsmittel eine
hohe Nukleaseaktivität, wenngleich diese für die fluorhaltigen Komplexe
geringer als für den unsubstituierten Komplex [Cu(phen)2]2+ ausfiel.
Zusätzlich wurden DNA-Bindungsstudien mit den Kupfer(II)-Komplexen
durchgeführt. Dabei zeigte sich, dass die DNA-Affinität mit steigender
Substituentengröße abnimmt. Die gleichzeitige Abnahme von DNA-Affinität und
Nukleaseaktivität der Komplexe weist auf einen engen Zusammenhang zwischen
diesen beiden Parametern hin. Die fluorhaltigen Kupfer(II)-Komplexe fungierten
interessanterweise, im Gegensatz zu [Cu(phen)2]2+, als selbstaktivierende
Nukleasen und zeigten DNA-Spaltaktivität in Abwesenheit von Reduktionsmittel.
Dabei konnte ein Zusammenhang zwischen dem Redoxpotential der Kupferkomplexe
und deren DNA-Spaltung aufgezeigt werden. So wies der Komplex [Cu(F2phen)2]2+
gleichzeitig die höchste Nukleaseaktivität und das positivste Redoxpotential
der untersuchten Komplexe auf. Das zweite zu untersuchende System beschäftigte
sich mit den Kupfer(II)-Komplexen von 2-Methyl-1,10-phenanthrolin (Mephen) und
dessen Derivaten 5(6)-X-2-Mephen (X = fluorhaltiger Substituent). Die Synthese
der Liganden mit unterschiedlichen Substituenten (F, CF3, SCF3, SF5) gelang
mittels Doebner-von Miller-Reaktion, einer Modifikation der Skraup-Reaktion.
Bis auf SF5Mephen konnten für alle Mephen-Derivate definierte Komplexe der
Zusammensetzung [Cu(XMephen)2]2+ erhalten werden. Diese Kupfer(II)-Komplexe
zeigten in Gegenwart von Ascorbinsäure jeweils eine hohe Nukleaseaktivität,
wobei auch hier DNA-Affinität und DNA-Spaltaktivität miteinander korrelierten.
Sowohl Nukleaseaktivität als auch DNA-Affinität nahmen mit steigender
Substituentengröße (H < F < CF3 < SCF3) ab. Ohne Reduktionsmittel konnte hier
hingegen keine Spaltung von Plasmid-DNA beobachtet werden. Während die
Affinität der Komplexe des [Cu(XMephen)2]2+-Systems für DNA verglichen mit den
analogen Komplexen des nicht-methylierten Systems [Cu(Xphen)2]2+ (X = H, F,
CF3, SCF3) größer oder zumindest gleich groß ist, zeigen die Komplexe der
nicht-methylierten Derivate eine höhere Nukleaseaktivität. Beide
Komplexsysteme wiesen überdies eine hohe Zytotoxizität für MCF-7-Zellen
(Brustkrebszelllinie, IC50 < 10 μM) auf, wobei der genaue Wirkmechanismus in
der Zelle noch unklar ist und dessen Aufklärung weitere Untersuchungen
erfordert. DNA-Spaltaktivität und Zytotoxizität der Komplexe korrelierten
nicht direkt miteinander, ein Zusammenhang zwischen beiden Eigenschaften kann
jedoch trotzdem nicht ausgeschlossen werden, da andere Aspekte bisher noch
nicht betrachtet wurden (z. B. die Aufnahme und Verteilung der Komplexe in den
Zellen).

dc.description.abstract
Artificial nucleases are powerful tools for biochemical and medicinal
applications. They target one of the most fundamental biomolecules, the DNA,
which is carrying the genetic information. Its uniqueness in transferring
genetic information makes it fundamental for cancer research, having in mind
that modifications in DNA strands can be responsible for the conversion of
healthy into cancer cells. Artificial nucleases can initiate apoptosis in
cancer cells via cleaving the DNA of the cell. One of the first developed and
well understood systems is bis(phenanthroline)copper(I), [Cu(phen)2]+,
reported by Sigman and co-workers in the late 70's of the last century. In the
last three decades there was a lot of research to improve and understand this
system, but no one has looked into fluorination of the ligand system. This is
surprising, since fluorination is a very often used tool for enhancing
bioactivity of pharmaceuticals. Our focus was on studying the influence of
fluorine-containing groups located on the phenyl moiety (5-Xphen), especially
regarding the bioactivity of the copper-phen system including affinity towards
DNA, cleavage activity and cytotoxicity against specific types of cancer
cells. The first topic included the synthesis of phenanthroline derivatives
with different fluorine-containing substituents. This was achieved with a
modified Skraup reaction under milder conditions. We managed to generate five
fluorine-containing derivatives of the form 5-Xphen (X = F, CF3, SCF3, OCF3,
SF5) plus the difluorinated 5,6-diFphen, out of which four have not been
synthesized before (X = CF3, OCF3, SCF3, SF5). We could obtain copper(II)
complexes of the structure [Cu(Xphen)2]2+ for all ligands except OCF3phen. The
copper(II) complexes are excellent DNA cleavers in the presence of ascorbic
acid as reducing agent and show good binding abilities for DNA. Both cleaving
and binding ability decreased with expanding substituent size (H < F < CF3 <
SCF3 < SF5) so that we can conclude a strong relationship between binding
ability and cleavage activity. Interestingly the fluorine-containing complexes
showed self-activated nuclease activity without any reducing agent in contrast
to the unsubstituted [Cu(phen)2]2+. Probably this activity exists due to the
more positive redox potentials of fluorinated [Cu(Xphen)2]2+ complexes and an
easier activation of the copper(II) complex via reduction. Quenching
experiments suggest an oxidative cleavage mechanism. The reason for activation
of copper(II) is still unclear, possible considerations are reduction through
DNA components like bases (e. g. guanine). The second investigated system are
copper(II) complexes of 2-methyl-1,10-phenanthroline (Mephen) and certain
fluorine-containing derivatives. This system is interesting because no
previous studies with [Cu(Mephen2]+/2+ have been reported before and it is a
kind of structural intermediate of the inactive [Cu(2,9-dimethylphen)]+/2+ and
the highly active [Cu(phen2]+/2+ system. Moreover, the methyl group makes it
attractive due to possible functionalizations at this group. The complexes
show high nuclease activity in the presence of ascorbic acid and high binding
abilities for DNA. Just like in the [Cu(phen)2]2+ system binding ability and
cleavage activity decrease with increasing substituent size. In contrast to
the former system no hints for self-activated nuclease activity were found.
Both systems, [Cu(Xphen)2]2+ and [Cu(XMephen)2]2+, were found to be highly
cytotoxic to MCF-7-cells (breast cancer cells, IC50 ≤ 10 μM) with little
higher toxicity for the methylated analogues. In contrast, nuclease activity
of the non-methylated species is higher compared to the methylated ones. No
clear correlation between nuclease activity and cytotoxicity was found and the
mode of action for toxicity is still unclear. Nevertheless, a relation between
nuclease activity and cytotoxicity cannot be excluded due to other aspects (e.
g. uptake and distribution of the complexes in cells).
