Einleitung: Sialinsäuren umfassen eine Gruppe von meist terminal gebundenen
Aminozuckern, die an einer Vielzahl von biologischen Prozessen wie der
Zellerkennung, Zelladhäsion und Proliferation, aber auch an pathologischen
Vorgängen, beispielsweise der Virusbindung oder Tumormetastasierung, beteiligt
sind. Beim metabolischen Oligosaccharid-Engineering (MOE) werden modifizierte
Sialinsäurevorläufer wie N-Propanoylmannosamin (ManNProp) in die zelluläre
Sialinsäurebiosynthese eingeschleust, zu modifizierten Sialinsäuren
metabolisiert und in Sialoglycane eingebaut. Ziel der Arbeit war die
Etablierung eines Zellsystems mit einer nahezu homogenen Population
strukturell veränderter Sialinsäuren auf der Zelloberfläche, um den Einfluss
modifizierter Sialoglycane auf biologische Vorgänge besser untersuchen zu
können. Methoden: Die hyposialylierten Subklone BJA-B K20 und HL60-I zweier
hämatopoetischer Zelllinien, die keine UDP-GlcNAc-2-Epimerase, das
Schlüsselenzym der Sialinsäure-biosynthese, exprimieren, wurden wie die normal
sialylierten Subklone BJA-B K88 und HL60-II mit 10 mM des natürlichen
N-Acylmannosamins ManNAc oder des unnatürlichen ManNProp über 72 h inkubiert.
Änderungen der Zelloberflächensialylierung und die Inkorporation modifizierter
Sialinsäuren wurden mittels HPLC und Durchflusszytometrie bestimmt.
Ergebnisse: Nach Inkubation mit ManNProp enthielten die Membranglycoproteine
von HL60-I- und BJA-B K20-Zellen einen Anteil von bis zu 85 % modifizierter
Sialinsäuren, die HL60-II und BJA-B K88 jedoch nur 15 % bzw. 56 %. Nach N
-Acylmannosamin-Behandlung wurde bei den HL60-I-Zellen ein signifikanter
Anstieg von Sialyl-Lewis X-Epitopen nachgewiesen. Vergleiche von Änderungen
des CMP-Sialinsäure-Gehalts, der Sialyl-Lewis X-Expression sowie der
Lektinbindungsmuster von ManNAc- und ManNProp-behandelten Zellen belegen, dass
ManNProp von den sonst permissiven Enzymen der CMP-Sialinsäurebiosynthese
weniger effizient metabolisiert wurde. Jedoch fanden sich gleichzeitig
Hinweise, dass offenbar manche Sialyltransferasen bevorzugt modifizierte CMP-
Sialinsäuren übertrugen. In dieser Arbeit konnte außerdem die lange infrage
gestellte Aufnahme freier Sialinsäureanaloga gezeigt werden.
Sialinsäureanaloga mit Modifikationen an C-5 oder C-9 wurden mit einem Anteil
von bis zu 95 % in die Membranglycoproteine von HL60-I- und BJA-B K20-Zellen
effizient inkorporiert. Schlussfolgerung: Durch den Einsatz freier
Sialinsäureanaloga lässt sich das Spektrum an Modifikationen in Sialoglycanen
deutlich erweitern. Die Inkorporation modifizierter N-Acyl-mannosamine sowie
freier Sialinsäureanaloga ist abhängig von der Aktivität der UDP-
GlcNAc-2-Epimerase. Das hier etablierte und charakterisierte UDP-GlcNAc-2
-Epimerase-defiziente Zellsystem ist daher für ein effizientes MOE besonders
gut geeignet.

Introduction: Sialic acids comprise a group of usually terminally bound amino
sugars, which are involved in a variety of biological processes such as cell
recognition, cell adhesion and proliferation but also in pathological
processes such as virus binding and tumor metastasis. Modified sialic acid
precursors such as N-propanoylmannosamine (ManNProp) are introduced by
metabolic oligosaccharide engineering (MOE) into the cellular sialic acid
biosynthesis, metabolized to modified sialic acids and incorporated into
sialoglycans. Aim of this work was the establishment of a cell system with an
almost homogeneous population of structurally altered sialic acids on the cell
surface in order to investigate the influence of modified sialoglycans on
biological processes. Methods: The hyposialylated subclones HL60-I and BJA-B
K20 of two hematopoietic cell lines that do not express UDP-GlcNAc
2-epimerase, the key enzyme of the sialic acid, were incubatetd as well as the
normally sialylated subclones BJA-B K88 and HL60-II with 10 mM of the natural
N-acylmannosamine ManNAc or the unnatural ManNProp for 72 h. Changes in cell
surface sialylation and the incorporation of modified sialic acids were
determined by HPLC and flow cytometry. Results: After incubation with ManNProp
the membrane glycoproteins of HL60-I and BJA-B K20 cells contained a
proportion of up to 85 % of modified sialic acids, but the HL60-II and BJA-B
K88 only 15 % and 56 %. After N-acylmannosamine-treatment, a significant
increase of sialyl Lewis X epitopes was demonstrated in the HL60-I cells.
Comparisons of changes in CMP-sialic acid content, sialyl Lewis X expression
and lectin binding patterns of ManNAc- and ManNProp-treated cells demonstrate,
that ManNProp was less efficiently metabolized by the enzymes of the otherwise
permissive CMP-sialic acid biosynthesis than natural ManNAc. However, also
evidence was found that some sialyltransferases apparently prefered to
transfer modified CMP-sialic acids. In this work, the long questioned uptake
of free sialic acid analogs could also be shown. Sialic acid analogs carrying
modifications at C-5 or C-9 were efficently incorprated resulting in a
proportion of up to 95 % of modified sialic acids in membrane glycoproteins of
HL60-I and BJA-B K20 cells. Conclusions: By using free sialic acid analogs the
range of modifications in sialoglycans can be significantly expanded. The
incorporation of N-acylmannosamine analogs and free sialic acid analogs
depends on the activity of UDP-GlcNAc 2-epimerase. The UDP-GlcNAc 2-epimerase-
deficient cell system established and characterized in this study is therefore
particularly suitable for efficient MOE.
