Claudins are considered the backbone of the tight junctions (TJ), sealing the gap between two adjacent cells and preventing the passage of solutes across epithelia. Interestingly, claudin-2 (Cldn2) forms channels at the TJ that allow the paracellular passage of cations and water. This protein is co-expressed in some epithelia, e.g., proximal nephron and small intestine, along with the barrier formers claudin-3 (Cldn3) and claudin-1 (Cldn1). However, the impact that the interaction of these claudins may have on the properties of Cldn2 channels is unclear.
Therefore, the aim of this dissertation was to determine whether co-expression of Cldn2 with Cldn3 or Cldn1 affects the ion and water permeability of Cldn2 channels. To achieve this goal, two knockout cell models (MDCK C7/Cldn3KO and MDCK II/quinKO) were used to express Cldn2 and co-express Cldn2/Cldn3. Additionally, Cldn2/Cln1 was co-expressed in MDCK II/quinKO cells. After characterizing the different cell lines by Western blot and different microscopy techniques (confocal, STED, and freeze-fracture electron), different electrophysiological assays were performed to evaluate the functions of Cldn2 channels in cell monolayers. The results of these experiments showed that co-expression of Cldn2/Cldn3 in both cell models, and co-expression of Cldn2/Cldn1 in MDCK II/quinKO cells reduced the absolute ion permeability of Cldn2 channels without altering their charge- or size-selectivity or ion dehydration patterns. However, co-expression of Cldn2/Cldn3 did not affect water flux in MDCK C7/Cldn3KO cells.
These findings suggest that Cldn2 channel properties are regulated by hetero-oligomerization of Cldn2 and either Cldn3 or Cldn1, forming claudin-intermixed channels that are permeable to water but not cations. These hybrid channels may coexist in the TJ meshwork with homo-oligomeric Cldn2 channels, which is supported by the results of this study and complementary experiments demonstrating Cldn2-Cldn3 trans– and Cldn2-Cldn1 cis-interactions.
Claudine werden als das Rückgrat der Tight Junction (TJ) bezeichnet, da sie den Zellzwischenraum abdichten und so den parazellulären Transport von Soluten über Epithelien verhindern. Interessanterweise bildet Claudin-2 (Cldn2) jedoch Kanäle innerhalb der TJ, die den parazellulären Durchtritt von Kationen und Wasser ermöglichen. In bestimmten Epithelien, wie dem proximalen Tubulus oder dem Dünndarm, wird Cldn2 gemeinsam mit den barrierebildenden Claudinen Claudin-3 (Cldn3) und Claudin-1 (Cldn1) coexprimiert. Der Einfluss dieser Co-Expressionen auf die funktionellen Eigenschaften der Cldn2-Kanäle ist bislang jedoch unklar.
Dementsprechend war das Ziel dieser Arbeit, zu untersuchen, ob die Co-Expression von Cldn2 mit Cldn3 oder Cldn1 die Ionendurchlässigkeit und den Wassertransport von Cldn2-Kanälen beeinflusst. Hierzu wurden zwei Knockout-Zellmodelle (MDCK C7/Cldn3KO und MDCK II/quinKO) verwendet, in denen Cldn2 alleine oder in Kombination mit Cldn3 exprimiert wurde. Zusätzlich wurde in MDCK II/quinKO-Zellen Cldn2 gemeinsam mit Cldn1 exprimiert. Nach der Charakterisierung der verschiedenen Zelllinien mit Hilfe von Western Blot und unterschiedlichen mikroskopischen Verfahren (konfokale Mikroskopie, STED und Gefrierbruch-Elektronenmikroskopie) wurden elektrophysiologische Messungen durchgeführt, um die Funktion der Cldn2-Kanäle in Zellmonolayern zu analysieren. Die Ergebnisse zeigten, dass die Co-Expression von Cldn2/Cldn3 in beiden Zellmodellen, sowie von Cldn2/Cldn1 in MDCK II/quinKO-Zellen die absolute Ionendurchlässigkeit der Cldn2-Kanäle reduziert, ohne deren Ladungs- oder Größenselektivität oder Dehydrierungsmuster der Ionen zu verändern. Der Wassertransport blieb bei der Co-Expression von Cldn2/Cldn3 in MDCK C7/Cldn3KO-Zellen unverändert.
Diese Befunde deuten darauf hin, dass die Eigenschaften des Cldn2-Kanals durch Hetero-Oligomerisierung mit Cldn3 oder Cldn1 beeinflusst werden. Dabei entstehen gemischte Claudin-Kanäle, die für Wasser, jedoch nicht für Kationen durchlässig sind. Solche Hybrid-Kanäle könnten daher innerhalb des TJ-Netzwerks zusammen mit homooligomeren Cldn2-Kanälen coexistieren. Diese Annahme wird durch die vorliegende Arbeit, sowie durch ergänzende Experimente zur trans-Interaktion von Cldn2/Cldn3 und zur cis-Interaktion von Cldn2/Cldn1 gestützt.
