Das Amyloid-Vorläuferprotein (APP) und die APP-ähnlichen Proteine 1 und 2
(APLP1 und APLP2) bilden eine evolutionär hoch konservierte Familie von
Transmembranproteinen bei Säugetieren. Es wird angenommen, dass APP der
entscheidende molekulare Faktor bei der Ausbildung der Alzheimer-Krankheit
ist, da es das neurotoxische Abeta-Peptid enthält, das bei Alzheimer-Patienten
zu Plaques aggregiert. APP, APLP1 und APLP2 bilden in lebenden Zellen native
Homo- und Heterodimere und enthalten Bindungsstellen für diverse
extrazelluläre Liganden. Die Auswirkungen der Ligandenbindung sind bislang
jedoch nicht verstanden. In dieser Arbeit wurde der Einfluss von Metallionen
auf die Struktur und Funktionen von APP und APLPs in der Zelle untersucht. Es
konnte in Messungen an lebenden Zellen gezeigt werden, dass APP, APLP1 und
APLP2 in Gegenwart von Zink innerhalb von Sekunden oligomere Komplexe aus
Homo- und Heterooligomeren ausbilden. Dabei wurde bereits bei niedrigen
mikromolaren Zinkkonzentrationen die Bildung von APP- und APLP1-Oligomeren
induziert, wohingegen deutlich höhere Zinkkonzentrationen für die
Oligomerisierung von APLP2 benötigt wurden. Die Zink-abhängige
Oligomerisierung von APLP1 wird über eine neue Zinkbindungsstelle im
C-terminalen Bereich der E2-Domäne vermittelt, wobei bei hohen
Zinkkonzentrationen weitere intermolekulare Kontakte auftreten können. Der
Mechanismus der Zink-abhängigen Oligomerisierung von APP und APLP1 wurde mit
rekombinanten E2-Domänen charakterisiert. Zinkionen banden direkt an die
E2-Domänen von APP, APLP1 und APLP2 und bewirkten strukturelle Änderungen in
der Domäne. Durch die Zinkionen erfolgte eine Dimerisierung der E2-Domänen von
APP und APLP1, aber nicht von APLP2. Die zusätzliche Dimerisierungsstelle
induzierte eine Oligomerisierung der dimeren Volllängenproteine. Die
physiologische Relevanz der Zink-abhängigen Oligomerisierung wurde durch ihren
Einfluss auf die Lokalisation von APLP1 in der Plasmamembran gezeigt.
Zinkionen vermittelten die Anreicherung von APLP1 in Zellkontaktbereichen bei
Zinkkonzentrationen, die bei neuronaler Aktivität in der Synapse entstehen.
APLP1 wird exklusiv im zentralen Nervensystem exprimiert und liegt vorwiegend
in der Plasmamembran vor. Dies legt eine Funktion von APLP1 als ein durch Zink
reguliertes synaptisches Adhäsionsprotein nahe.

dc.description.abstract
The amyloid precursor protein (APP) and the APP-like proteins 1 and 2 (APLP1
and APLP2) are the mammalian representatives of an evolutionarily conserved
family of transmembrane proteins. APP is hypothesized to be the decisive
molecular factor in the etiology of Alzheimer Disease by harboring the
neurotoxic Abeta peptide, which aggregates to plaques in Alzheimer Disease
patients. In living cells, APP, APLP1 and APLP2 form native homo- and
heterodimers and they contain binding sites for several extracellular ligands.
However, the consequences of ligand binding are not understood. Here, the
influence of metal ions on structure and function of APP and APLPs in the cell
was studied. It was shown by live-cell analyses that in presence of zinc APP,
APLP1 and APLP2 form homo- and heterooligomeric complexes within seconds.
Whereas low micromolar zinc concentrations induced formation of APP and APLP1
oligomers, far higher concentrations were required to mediate APLP2
oligomerization. Zinc-induced oligomerization is mediated by a novel zinc
binding site in the C-terminal region of the E2 domain. At higher zinc
concentrations formation of additional intermolecular contacts was observed.
The mechanism of zinc-mediated oligomerization was characterized with
recombinant E2 domains of APP, APLP1 and APLP2. Zinc ions bound directly to
the E2 domains of APP, APLP1 and APLP2 and induced structural rearrangements
of the domain. A dimerization of APP and APLP1 E2 domains was induced by zinc
ions, but no dimerization of the APLP2 E2 domain was observed. The additional
dimerization site led to oligomerization of the dimeric full length proteins.
The physiological relevance of zinc-mediated oligomerization was shown by its
influence on the localization of APLP1 in the plasma membrane. Zinc ions
mediated enrichment of APLP1 in cell contacts at concentrations occuring at
the synapse during neuronal activity. APLP1 is exclusively expressed in the
central nervous system and mainly localized to the plasma membrane. This
suggests a function for APLP1 as a zinc-regulated synaptic adhesion protein.
