Brain oscillations, as measured by electro- and magnetoencephalography (EEG/MEG), mediate brain functions such as perception, cognition, and learning. Transcranial alternating current stimulation (tACS) can interact with brain oscillations to influence behavior and has shown promise in the treatment of brain disorders. However, its underlying mechanisms are poorly understood, and stimulation effects often vary considerably within and between participants. To better understand the mechanisms of tACS and reduce effect variability by adapting stimulation to ongoing brain oscillations, it would be necessary to measure brain oscillations during stimulation. However, stimulation artifacts in simultaneously recorded EEG/MEG have prevented assessment of brain oscillations during tACS. Here, stimulation artifact source separation (SASS), an adaptive spatial filter that enables assessment of single-trial brain oscillations targeted by amplitude-modulated tACS (AM-tACS), is introduced. Using SASS, it is shown that AM-tACS enhances and suppresses brain oscillations in a phase-dependent manner. Such modulation of brain oscillations is linked to conscious visual perception during a binocular rivalry task. These results show that phase-dependent enhancement and suppression of brain oscillations is a key mech-anism of AM-tACS and pave the way for the development of closed-loop protocols where the phase of AM-tACS is adapted to ongoing brain oscillations in real-time. Closed-loop approaches are expected to reduce variability of tACS effects by enabling selective enhancement and suppression of brain oscillations, increasing its clinical applicability in the treatment of brain disorders.

Die transkranielle Wechselstromstimulation (tACS) kann Gehirnoszillationen modulieren, um Verhalten zu beeinflussen, und hat in der Behandlung von Gehirnerkrankungen vielversprechende Ergebnisse gezeigt. Die zugrunde liegenden Mechanismen sind jedoch schlecht verstanden, und Stimulationseffekte variieren oft erheblich innerhalb und zwischen Teilnehmern. Um die Mechanismen der tACS besser zu verstehen und die Effektvariabilität durch Anpassung der Stimulation an Gehirnoszillationen zu reduzieren, wäre es notwendig, Gehirnoszillationen während der Stimulation zu messen. Elektrische Stimulationsartefakte in gleichzeitig aufgezeichneten EEG/MEG Signalen verhindern jedoch die Messung von Gehirnoszillationen während der tACS. Hier wird Stimulation Artifact Source Separation (SASS), ein adaptiver räumlicher Filter zur Messung von Gehirnoszillationen unter tACS, vorgestellt. Mit SASS wird gezeigt, dass AM-tACS Gehirnoszillationen in einer phasenabhängigen Weise verstärkt und unterdrückt. Die Modulation von Gehirnoszillationen wird mit bewusster visueller Wahrnehmung während der binokularen Ri-valität verknüpft. Diese Ergebnisse zeigen, dass die phasenabhängige Verstärkung und Unterdrückung von Gehirnoszillationen ein Schlüsselmechanismus von AM-tACS ist. So-mit wird der Weg für die Entwicklung von Closed-Loop Protokollen geebnet, bei denen die Phase von AM-tACS in Echtzeit an modulierte Gehirnoszillationen angepasst wird. Es wird erwartet, dass solche Closed-Loop-Ansätze die Variabilität von tACS-Effekten reduzieren, indem sie eine selektive Verstärkung und Unterdrückung von Gehirnoszillationen ermöglichen, und damit die Anwendbarkeit von tACS bei der Behandlung von Gehirnerkrankungen erhöht wird.
