The nuclear periphery including the nuclear lamina is one of the main nuclear sub-compartments that facilitate the complex yet dynamic chromatin organization in eukaryotes. The concept of plant lamina is relatively recent, as true lamin homologs are absent in plants, a thorough understanding of its functionality is not fully delved into. In Arabidopsis thaliana it has been shown previously that PWO1, an interactor of Polycomb repressive complex 2 (PRC2), collaborates with the nuclear lamina protein CRWN1 to mediate gene regulation. This study has demonstrated that the SWI/SNF chromatin remodeler subunit AtBAF60 interacts with PWO1 and CRWN1 by employing protein-protein interaction studies, gene expression analysis, and nuclear phenotype measurements using confocal microscopy. Transcriptome and genome-wide analyses coupled with immunocytochemical-high resolution microscopy studies have revealed the functional interplay between the interacting partners suggesting their role in stress responses and heterochromatin organization. In a nutshell, the present study discovered a novel interrelationship between SWI/SNF chromatin remodeler complex subunit AtBAF60 with the nuclear periphery and PWO1, thereby interconnecting three major players of gene regulation and chromatin organization in A. thaliana
Die Kernperipherie einschließlich der Kernlamina ist eines der wichtigsten Kernunterkompartimente, die die komplexe und dennoch dynamische Chromatinorganisation bei Eukaryoten ermöglichen. Das Konzept der Pflanzenlamina ist relativ neu, da echte Laminhomologe bei Pflanzen fehlen und ihre Funktionalität noch nicht vollständig verstanden ist. Bei Arabidopsis thaliana wurde bereits gezeigt, dass PWO1, ein Interaktor des Polycomb-Repressivkomplexes 2 (PRC2), mit dem Kernlaminaprotein CRWN1 zusammenarbeitet, um die Genregulation zu vermitteln. Diese Studie hat gezeigt, dass die SWI/SNF-Chromatin-Remodeler-Untereinheit AtBAF60 mit PWO1 und CRWN1 interagiert, indem Protein-Protein-Interaktionsstudien, Genexpressionsanalysen und Messungen des Kernphänotyps mittels konfokaler Mikroskopie eingesetzt wurden. Transkriptom- und genomweite Analysen in Verbindung mit immunzytochemischen hochauflösenden Mikroskopiestudien haben das funktionelle Zusammenspiel der interagierenden Partner enthüllt, was auf ihre Rolle bei Stressreaktionen und der Heterochromatinorganisation hindeutet. Kurz gesagt entdeckte die vorliegende Studie eine neuartige Wechselbeziehung zwischen der SWI/SNF-Chromatin-Remodeler-Komplex-Untereinheit AtBAF60 mit der Kernperipherie und PWO1 und verbindet damit drei Hauptakteure der Genregulation und Chromatinorganisation in A. thaliana.
