dc.description.abstract
Die rechtzeitige Detektion von Tumorerkrankungen könnte in Zukunft die
Möglichkeit erhöhen, Patienten effektiver und schneller zu heilen und so ihre
Überlebenschancen zu erhöhen. Bisher gibt es für viele Tumorarten nicht die
geeignete Diagnosemethode, um eine frühzeitige und spezifische Diagnose zu
gewährleisten. Daher ist der Bedarf groß neue, schnelle sowie spezifische
Methoden zu etablieren, mit denen es möglich ist, bereits aus Seren von
Tumorpatienten potentielle Biomarker zu entdecken. Es ist auch von
entscheidender Bedeutung, Biomarker für die Bestimmung der Art und des
Stadiums der Tumorerkrankung zu identifizieren. Die Veränderung der
Glykosylierung auf Zelloberflächen während der Entwicklung, Differenzierung
und Erkrankung wird zunehmend als potentielle spezifische Biomarkerkandidaten
für viele Krebsarten und Entzündungskrankheiten betrachtet. Borderline-
Ovarialkarzinom Im Rahmen dieser Arbeit wurde das N-Glykosylierungsmuster von
Borderline-Ovarialkarzinom-Patientinnen analysiert um herauszufinden, ob es
mögliche Unter-schiede zwischen Ovarial (Ovca)- und Borderline-Ovarialkarzinom
(BOT) Patientin-nen gibt. Zuerst wurde das IgG-Glykom unter Verwendung von
Trypsin analysiert. Es konnten insgesamt sechs signifikante
Glykopeptidstrukturen ermittelt werden, die in Kombination mit dem gängigen
Biomarker CA-125 die beste Spezifität sowie Sensi-tivität aufweisen, um OvCa
sowie BOT-Serenproben von gesunden Serenproben zu unterscheiden. Mit dieser
Kombination war es möglich, alle BOT-Proben als krank zu klassifizieren. Es
war aber nicht möglich, zwischen OvCa und BOT zu unterscheiden. Im nächsten
Schritt wurde das N-Glykom des gesamten Serums von OvCa- und BOT Patientinnen
mit ihren entsprechenden altersangepassten gesunden Kontrollen analysiert und
verglichen. N-Glykane wurden selektiv mit dem Enzym PNGase F aus menschlichen
Blutseren freigesetzt, isoliert und anschließend mittels MALDI-TOF-MS
gemessen. Um alle N-Glykanstrukturen einschließlich diejenigen mit negativ
geladenen Sialinsäuren, zu detektieren, wurden sie zuvor mit Methyliodid
derivatisiert. Diese Derivatisierungsreaktion ermöglicht eine Messung im
positiven Ionisations-modus. Ihre Strukturen wurden mit MALDI-TOF/TOF-MS
Fragmentierungs-Analysen bestätigt. Die Arbeitsgruppe hat bereits einen Serum
Biomarker namens GLYCOV spezifisch für OvCa entwickelt. Für BOT-Proben wurden
auch die GLYCOV-Werte bestimmt. Allerdings war eine Differenzierung zwischen
OvCa- und BOT nicht möglich. Ein neuer Wert, GLYCOV_B, wurde aus den
statistisch signifikanten Strukturen in BOT etabliert, um diese Kohorte besser
zu bestimmen und einzugrenzen. Mit dem neu berechneten Wert wurde keine
einzige Probe als gesund eingestuft. Die Haupt-komponentenanalyse bestätigt
nochmals, dass beide maligne Kohorten sich voneinander unterscheiden. Eine
solche Klassifizierung wurde bisher noch nicht in der Literatur beschrieben.
Kolonkarzinom Desweiteren wurde in dieser Arbeit das N-Glykosylierungsprofil
aus Seren von Kolonkarzinom (CRC)-Patienten untersucht, um potentielle
glykanbasierende Biomarker zu identifizieren. Diese Art von Tumorerkrankung
gehört weltweit zu den führenden Todesursachen. Die Überlebenschancen steigen
mit der frühzeitigen Erkennung um mehr als 90%. Daher sind robuste Biomarker
mit hoher Empfindlichkeit sowie Spezifität sehr hilfreich. Nach der
enzymatischen Freisetzung wurden die N-Glykane gereinigt, permethyliert und
mittels MALDI-TOF-MS analysiert. Die signifikanten Strukturen wurden mit
MALDI-TOF/TOF-MS bestätigt. In CRC Seren sind zwei monofucosylierte
biantennäre Strukturen herunterreguliert, wohingegen zwei difucosylierte,
mono- und diantennäre Strukturen und drei sialylierte monofucosylierte
Strukturen hochreguliert sind. Aus den sieben bedeutenden Strukturen wurde ein
sogenannter GlycoColon-Wert berechnet, der mit CEA, dem aktuellen Biomarker
für Darmkrebs, verglichen wurde. Sowohl die Spezifität, als auch die
Sensitivität der GlycoColon-Werte waren besser als die vom CEA Biomarker.
Darüber hinaus konnte gezeigt werden, dass die Kombination von beiden
Biomarkern eine Sensitivität von 92% und eine Spezifität von 96% liefert. In-
Gel-Analytik Um eine schnelle und effiziente Methode zur Freistzung und
Analyse von geringen Mengen an N-Glykanen zu etablieren, wurde die Detektion
von In-Gel verdauten N-Glykanen durch die Zugabe von OGP, einem nichtionischen
Detergens und der Nutzung von Anchorchip-Target mit D-Arabinosazon als Matrix
verbessert. Es wurden drei verschiedene Modellproteine (RNase B, AGP und
Fetuin) in SDS-PAGE-Gelen in Anwesenheit von OGP mit PNGase F verdaut. Mit
dieser Optimierung konnte eine Nachweisgrenze von mindestens 10 pmol des
Modellproteins bestätigt werden. Die MALDI-TOF-MS-Spektren mit höheren
Signlintensitäten bestätigen, eine verbesserte Freisetzung und Extraktion von
N-Glykanen aus im Gel verdauten Proteinen durch die Zugabe von OGP. Glykan-
Mikroarrays für die serumbasierte N-Glykananalytik Die Mikroarraytechnologie,
die eine schnellere und effizientere Analyse ermöglicht, wird zunehmend für
neuartige diagnostische Anwendungen eingesetzt. Sie ist auch ein
weitverbreitetes Glykomik-Werkzeug geworden, um biologische Fragen zu
entschlüsseln. In dieser Studie wurden N-Glykane aus Seren isoliert, gereinigt
und anschließend markiert. Die für die Mikroarray-Analyse benötigten optimalen
Bedin-gungen wie Labelreagenz, Oberfläche und Bindungsprotokoll, wurden
mithilfe von Glucoseoligomeren bestimmt. Es konnte gezeigt werden, dass
anstelle des üblichen Labelreagenz 2AB, DAP-Glucoseoligomere viel effizienter
an epoxyaktivierten Glasträgern binden. Danach wurden Seren von OvCa-
Patientinnen und ihren entsprechenden alters-angepassten gesunden Kontrollen
mithilfe von Glykan-Mikroarrays untersucht. Mannosestrukturen und komplexe
N-Glykanstrukturen (Hex6HexNAc5Neu5Ac3-2AB (m/z 3764.6) und
Hex5HexNAc4Neu5Gc2-2AB (m/z 3014.6)) wurden zuvor durch HPLC-Fraktionierung
isoliert und auf epoxyaktivierte Glasträger gedruckt. Nachdem die Glykan-
Mikroarrays mit Serum beschichtet wurden, wurde ein zweiter fluores-zierender
Antikörper, der gegen Immunglobuline gerichtet war, angewendet. Die
resultierenden Fluoreszenzsignale wurden unter Verwendung eines Fluoreszenz-
scanners gemessen. Die Signalintensitäten von Highmannosen und
Hex5HexNAc4-Neu5Gc2-2AB waren auf OvCa-Arrays signifikant stärker als auf den
gesunden Arrays. Zusammenfassend kann gesagt werden, dass Anti-Highmannose-
und Anti-NeuGc-Autoantikörper potentielle Serum-Biomarker für OvCa sind.

dc.description.abstract
Most cancerous tumors are detected at a late-stage because of the lack of
suitable diagnostic methods to ensure an early and specific diagnosis.
Therefore, there is a crucial need to establish new and rapid method that
allows the early detection of neoplasia in the sera of tumor patients, which
could offer the possibility to heal them faster and to increase their chances
of survival. It is also of high interest to discover potential biomarkers that
determine the type and stage of the tumor disease prior to invasive
operations. Glycome changes during development, differentiation and disease
are increasingly recognized as biomarkers that are specific for many types of
diseases such as cancer and inflammatory diseases. Borderline ovarian cancer
In this study, the serum N-glycosylation pattern of BOT patients was analyzed
to find out, whether it was possible to differentiate between ovarian- and BOT
patients at the serum level. At first, the IgG glycome was measured using
tryptic glycopeptides, purified from serum using protein A. Six statistically
significant glycopeptide structures were detected by MALDI-TOF-MS using SPSS
analyses. The best specificity and sensitivity could be achieved with the
combination of the common used biomarker CA-125 and the six different
structures. With this combination, it was possible to classifiy all BOT
samples as ill but it was not possible to distinguish between OvCa and BOT
samples. In the next step, the N-glycome of total serum from OvCa, BOT and
age-matched healthy controls were analyzed. N-glycans were selectively
released with the enzyme PNGase F from human blood sera, isolated and
subsequently measured by MALDI-TOF-MS. In order to detect all N-glycan
structures including those with negatively charged sialic acid units,
N-glycans were previously derivatized with methyl iodide. This derivatization
reaction enables a measurement in positive ionization mode of all N-glycan
structures. Their structures were confirmed by MALDI-TOF/TOF-MS fragmentation
analysis. The working group has already developed a serum biomarker named
GLYCOV for OvCa. GLYCOV values were determined for BOT samples but they failed
to discriminate BOT from healthy controls. A new value, GLYCOV_B, was
calculated from the statistical differences observed between BOT and healthy
control samples. With the new calculated value, all BOTsamples were classified
as borderline and none of them were classified as healthy. The principal
component analysis makes clear that both disease cohorts differ from each
other. Such a classification has not been previously described in the
literature. Colon cancer Further, the N-glycosylation profile from sera of
colon cancer patients was examined to identify potential glycan-based
biomarkers. This type of neoplasia is among the world's third leading cause of
death. Survival chances increase with early detection by more than 90%.
Therefore, robust serum biomarkers with high sensitivity and specificity would
be very helpful. After enzymatic release, N-glycans were purified, derivatized
via permethylation and measured by MALDI-TOF-MS. Seven statistically
significant structures were identified by SPSS analyses and their structures
were confirmed by MALDI-TOF/TOF-MS. Two monofucosylated biantennary structures
were down-regulated whereas two difucosylated mono- and diantennary structures
and three sialylated monofucosylated structures were upregulated. A so-called
GlycoColon value was generated from the seven significant structures, which
was compared with CEA, the current biomarker for colorectal cancer. Both,
specificity and sensitivity of GlycoColon values, were better than of CEA. In
addition, when both biomarkers were combined together, the sensitivity reached
92% and the specificity 96%. In-gel N-glycan analysis To establish a rapid and
efficient method to release and analyze minute amounts of N-glycans, the
detection of in-gel digested N-glycans by MALDI-TOF was improved for model
proteins by the addition of OGP, a non-ionic detergent, and by measurements on
an Anchorchip target using D-arabinosazone as the matrix. Three different
model proteins (RNase B, AGP and Fetuin) were digested in SDS-PAGE gels in the
presence or absence of OGP. With this optimization, a limit of detection as
low as 10 pmol of the model glycoprotein could be achieved. The addition of
OGP enables an improved release and extraction of N-glycans from proteins
stacked within the gel as the MALDI-TOF-MS spectra show structures with higher
signal intensities. Glycan microarrays for serum-based diagnostics The
microarray technology that enables faster and more efficient analysis is
increasingly used for establishing novel diagnostic applications. In parallel,
it has become a wide¬spread glycomics tool to unravel biological questions. In
this study, N-glycans were isolated from human sources and subsequently
labeled. The optimum label, surface and binding protocols were selected using
glucose oligomers as model substances. It could be shown that, instead of the
usual label 2AB, DAP glucose oligomers were binding to the surface of the
slide much more efficiently. Thereafter, sera from OvCa patients and age-
matched healthy controls were investigated with the microarray technology.
High mannose N-glycans and complex-type N-glycanstructures
(Hex6HexNAc5Neu5Ac3-2AB (m/z 3764.6) and Hex5Hex-NAc4Neu5Gc2-2AB (m/z 3014.6))
were isolated by HPLC fractionation. Slides were coated with glycans, serum
was put on the slides and finally a secondary fluorescent antibody directed
against immunoglobulins was applied. The resulting fluorescent signals were
measured using a fluorescence scanner. The signal intensities of the high-
mannose N-glycans and Hex5HexNAc4Neu5Gc2-2AB were significantly stronger on
OvCa arrays than on healthy control arrays. In summary, anti-high-mannose and
anti-NeuGc autoantibodies are potential serum biomarkers for OvCa.
