dc.description.abstract
Seit Ende 2002 wurde in Deutschland bei Mastküken aus verschiedenen Herden ab
dem sechsten Lebenstag ein Krankheitsgeschehen beobachtet, das durch Durchfall
und anschließende Wachstumsdepression gekennzeichnet ist. Der
Krankheitskomplex wird in der Literatur als Malabsorptionssyndrom (MAS)
bezeichnet. Da eine antimikrobielle Therapie keinen Erfolg brachte, wurde eine
virale Ätiologie vermutet. Aus 17 Broiler-Beständen mit aktueller MAS-
Problematik wurden aus 56 von 68 Organproben Reoviren isoliert und mittels
Indirektem Immunfluoreszenztest sowie elektronenmikroskopisch identifiziert.
In neun ausgewählten Proben, aus denen später ausschließlich Reoviren isoliert
werden konnten, wurden in der elektronenmikroskopischen Untersuchung 4x
Rotaviren, 1x Reoviren, 1x Rota- und Reoviren, 1x Calici- sowie einzelne Rota-
und Reoviren und 1x Paramyxovirus-ähnliche Partikel nachgewiesen. In einer
untersuchten Probe konnte kein Virus gefunden werden. 24 Reovirus-Isolate
wurden mit monoklonalen Antikörpern (mAk) untersucht, die eine Einteilung der
Isolate in enteric reovirus strains (ERS) und Reoviren („nicht-ERS“) erlauben.
16 Isolate erwiesen sich als ERS. Eine weitere Differenzierung ausgewählter
Isolate wurde mittels der vergleichenden Morphologie der cpE sowie der
Plaquegrößen, mit Plaquereduktions-Testen und mit der RNA-Polyacrylamid-
Gelelektrophorese (PAGE) durchgeführt. Die Ergebnisse, die mit diesen
morphologischen, serologischen und molekularbiologischen Methoden erzielt
wurden, haben keine einheitliche Einteilung der MAS Isolate in pathogen bzw.
nicht pathogen zugelassen. Zur Bestimmung der Pathogenität ausgewählter
Isolate in vivo wurden einen Tag alte SPF-Broiler oral infiziert. Alle Küken
der Gruppe, die mit dem ERS-Vergleichsstamm aus Polen 238/98 infiziert worden
waren, verstarben innerhalb von sechs Tagen p.i. Das beschriebene
Krankheitsbild konnte vollständig reproduziert werden, während dies mit dem
ERS-Feldisolat 120/03-1 aus Deutschland und dem Reovirus-Prototyp-Stamm S1133
nicht möglich war. Das „nicht-ERS“-Feldisolat 259/03 aus Deutschland führte zu
einer signifikanten Reduktion des Körpergewichts der Tiere und zu klinischen
Erscheinungen, die jedoch nicht mit denen der ursprünglichen Herdenproblematik
übereinstimmten. Der Rückgang der Virusausscheidung anhand der Ergebnisse der
Virusreisolierung aus Kloakentupfern korrelierte innerhalb der Gruppen
entsprechend mit dem Einsetzen bzw. dem Anstieg der AK-Bildung. Bei den
histopathologischen Untersuchungen wurden in der Gruppe 238/98 v.a. Nekrosen
in Leber, Milz und Bursa gefunden, während in den Gruppen S1133, 120/03-1,
120/03-1 (AM) und 259/03 primär Myo- und Epikarditiden konstatiert wurden. Bei
der Gruppe 259/03 wurden zusätzlich Gastritiden beobachtet. Mittels
Immunhistochemie konnten vor allem in Milz, Bursa Fabricii sowie Blinddarm und
Blinddarmtonsillen Virus Antigen nachgewiesen werden. Aus am Ende des Versuchs
entnommenen Organprobenpools der mit dem deutschen ERS-Feldisolat 120/03-1,
dessen Ausgangsmaterial 120/03-1 (AM) sowie dem Reovirus-Prototyp-Stamm S1133
infizierten Tiere wurde sporadisch Virus aus Blinddarm und Blinddarmtonsillen
sowie der Milz reisoliert. Bei den mit dem „nicht-ERS“-Feldisolat 259/03
infizierten Tieren wurde aus den untersuchten Organen weder Virus reisoliert
noch Antigen nachgewiesen. Aus jedem Organpool der Tiere, die mit dem ERS-
Vergleichsstamm aus Polen 238/98 infiziert worden waren und bis zum sechsten
Tag p.i. starben, konnte Virus reisoliert und in den Organen exklusive des
Gehirns Reovirus Antigen detektiert werden. Die erzielten Ergebnisse zeigen,
dass es mit den verwendeten labordiagnostischen Methoden nicht möglich war,
die Pathogenität von Reoviren ohne Tierversuche eindeutig zu bestimmen. Es
konnte in diesen Untersuchungen kein Zusammenhang zwischen Sero- Elektrophero-
und Pathotyp ermittelt werden. Die Heterogenität der untersuchten Isolate
spricht gegen ein monokausales, durch bestimmte Reoviren verursachtes MAS-
Geschehen, darüber hinaus sind nicht alle ERS-Isolate als pathogen zu
betrachten. Aus den Untersuchungen wird deutlich, dass mit keiner der
angewandten Untersuchungsmethoden ein gemeinsames Merkmal aller MAS
verursachenden Isolate gefunden werden kann. Der verfügbaren Literatur zufolge
ist dies die erste Arbeit, in der eine experimentelle Infektion mit SPF-
Broilern durchgeführt wurde.

dc.description.abstract
Since the end of 2002 the incidence of a syndrome characterized by diarrhoea
and subsequent stunting in broilers starting at six days of age has increased
drastically in Germany. In the literature this disease complex is called
malabsorption syndrome (MAS). A viral aetiology has been suspected because
antimicrobial therapy was not successful. In the present study 68 samples of
different organs from 17 broiler flocks with MAS history were investigated
virologically. In 56 out of 68 samples reoviruses were isolated and identified
by IIFT and electron microscopy. 9 selected samples of the original material,
from which only reoviruses had been isolated, were also examined by electron
microscopy. Following results were obtained: 4x rotavirus, 1x reovirus, 1x
rota- and reovirus, 1x calicivirus with some rota- and reovirus particles and
1x paramyxovirus-like particles. In one sample no any viruses were found. 24
reovirus isolates were selected and classified as enteric reovirus strains
(ERS) or reovirus (“non-ERS”) using monoclonal antibodies. 16 isolates were
found to be to ERS. For further characterization of selected isolates,
morphology of cytopathic effect, size of plaques, results of plaquereduction
tests and polyacrylamid gel electrophoresis of extracted RNA were compared.
The obtained results did not allow a classification of the reovirus isolates
in pathogenic and non-pathogenic. The pathogenicity in vivo of selected
isolates was investigated in orally infected one day old SPF-broiler chickens.
All birds of the group, which were infected with the ERS-prototype 238/98
(polish strain) died within six days p.i. and the described disease pattern
was reproduced. This was not possible with the German ERS-field isolate
120/03-1 as well as the reovirus-prototype-strain S1133. The German “non-
ERS”-isolate 259/03 caused a significant reduction of body weight and clinical
signs which did not correspond to those in the original flock. The examination
of cloacal swabs for virus shedding at different ages showed that a shedding
reduction correlated with the increase of antibody level. Histopathological
examinations showed in group 238/98 mainly necrosis in liver, spleen and Bursa
of Fabricius while in groups S1133, 120/03-1, 120/03-1 (original material, AM)
and 259/03 primarily myo- and epicarditis were seen. In addition in group
259/03 gastritis were observed. By immunohistochemistry at the end of the
experiment virus antigen could be detected mainly in spleen, Bursa of
Fabricius and caecum with caecal tonsils. In samples of organs from birds
infected with the German ERS-field isolate 120/03-1, its original material
120/03-1 (AM) as well as the reovirus-prototype-strain S1133 sporadically
virus could be reisolated from caecum with caecal tonsils and spleen. In birds
infected with the German “non-ERS”-isolate 259/03 neither virus could be
reisolated nor virus antigen could be detected in any organ at the end of the
experiment. From birds infected with the ERS-prototype 238/98 which died
before or on day six p.i. virus could be reisolated from all organs and virus
antigen could be detected in all organs except the brain. The results show
that it was impossible to determine the pathogenicity of reovirus using in
vitro methods used in this study without experimental infection in vivo. No
correlation between sero-, elektrophero-and pathotype could be found in this
study. The heterogeneity of the examined isolates makes it unlikely that MAS
is a disease caused only by reoviruses. Furthermore it was shown that not all
ERS isolates are pathogenic. The in vitro methods used in this study did not
allow to identify a common marker of reoviruses causing MAS. According to
available literature this is the first report on experimental infection with
reovirus in SPF-broilers.
