Die Wirkung von Estramustin, Etoposid und des synthetischen MMP-Inhibitor
ICOL-51 (ein MMP-2 und MMP-9 selektiver Inhibitor) wurde am Duningtumor in
vivo und in vitro, als Mono,- und Kombinationstherapie untersucht. In vitro
konnte nur eine Wirkung von Etoposid auf das Tumorzellwachstum nachgewiesen
werden, ebenso ein synergistischer Effekt von Estramustin auf Etoposid.
ICOL-51 hingegen blieb ohne Wirkung. In vivo zeigte Estramustin keine Wirkung
auf das Tumorwachstum, Etoposid eine Tumorreduktion und ICOL-51 vgl. mit
Etoposid eine noch stärkeren Einfluss auf das Tumorwachstum. Die Kombination
von Etoposid und ICOL-51 zeigte keinen zusätzlichen synergistischen Effekt.
Die selektive Hemmung der MMP Aktivität ist ein neuer, viel versprechender
therapeutischer Ansatz und lässt für weitere Studien am hormonrefraktären,
metastasiertem Prostatakarzinom hoffen.

The therapeutic efficacy of estramustine, etoposide and the synthetic
inhibitors of matrix-metalloproteinases ICOL-51 (high selectivity for MMP2,
MMP9) was evaluated on the dunningtumor in vitro and in vivo as monotherapy
and combinationtherapy. In vivo only etoposide has shown an effect on
tumorgrowth, a synergistic effect of estramustine on etoposid could be shown
as well. ICOL-51 had no effect on tumorgrowth. In vivo estramustine had no
effect, etoposide an effect on tumorgrowth and ICOL-51 compared with etoposid
an even stronger influence on the tumorgrowth. The combination of etoposide
and ICOL-51 could not show further synergistic effect. Selective inhibition of
MMP activity is a novel therapeutic approach, which bears promise for studies
in patients with prostate cancer.
