Das antidiuretische Hormon (ADH) spielt eine wichtige Rolle bei der
Harnkonzentrierung. Die ADH-abhängige Wasserresorption in den Hauptzellen des
Sammelrohrs wurde in den vorangegangenen Studien detailliert beleuchtet,
während die Wirkung des Hormons im distalen Nephron nur in wenigen Ansätzen
untersucht war. Die Expression des V2 Rezeptors für ADH im distalen Nephron
wurde ebenfalls bestritten. Distale Salztransporter, NKCC2 und NCC, sind
entscheidend in die Regulation der renalen Salzausscheidung involviert und
bestimmen somit das extrazelluläre Volumens und den Blutdruck mit. Die
vorliegende Habilitationsarbeit beschäftigt sich mit der ADH-abhängigen
Steuerung dieser Transporter. Die hier aufgeführten Originalarbeiten
beschreiben die Expression des V2 Rezeptors entlang dem distalen Nephron und
die ADH-abhängige Regulation von NKCC2 und NCC bei deren Phosphorylierung.

dc.description.abstract
AVP has been recognized as the most potent endocrine stimulus of the urinary
concentrating mechanism. Previous work has established the role of AVP in the
regulation of water reabsorption in principal cells of collecting duct,
whereas its action in the distal nephron was less characterized. In
particular, no conclusive information was available on the expression of the
vasopressin V2 receptor (V2R) and V2R-mediated effects along the distal
convoluted tubule. The distal salt transporters, NKCC2 and NCC, play an
important role in renal salt handling and volume regulation. The present
habilitation thesis aimed at elucidating the action of AVP along the distal
nephron. Therefore, effects of AVP on NKCC2 and NCC were studied, and the
molecular pathways involved in AVP signaling were characterized. The
publications presented in this thesis address critical issues in this field,
namely the characterization of the cellular distribution of V2R along the
nephron and the elucidation of the AVP-related kinase pathways mediating the
regulation of NKCC2 and NCC.
