Einleitung: Die Reduktion der Mismatch-Negativität (MMN) stellt einen
gesicherten Befund bei Schizophrenie dar. Rs4680 beeinflusst als genetische
Variante des Enzyms die Aktivität der Catechol-O-Methyltransferase (COMT) bei
Schizophrenie-Patienten und bei Gesunden. Unter Nutzung des Endophänotyps-
Konzepts in der Schizophrenie sollte der Einfluss von rs4680 auf die MMN
überprüft werden. Methoden: Bei 77 Patienten mit Schizophrenie und 92 gesunden
Kontrollen erfolgte die Bestimmung der MMN sowie des COMT-Genotyps. Es wurden
außerdem ein psychopathologisches Rating mittels PANSS (Positive and Negative
Syndrome Scale) durchgeführt sowie biografische Daten erhoben. Ergebnisse:
Rs4680 modulierte die MMN sowohl bei den gesunden Kontrollen als auch bei den
Schizophrenie-Patienten. Unterschiedliche Effekte des COMT-Genotyps auf die
MMN zwischen beiden Gruppen konnten nicht gefunden werden. Schlussfolgerungen:
Der Nachweis, dass der modulierende Einfluss von rs4680 auf die MMN bei
Vorliegen von Schizophrenie anders ausfällt als bei Gesunden konnte nicht
erbracht werden. Rs4680 konnte somit kein Einfluss auf die Lebenszeit-
Prävalenz von Schizophrenie nachgewiesen werden. Zukünftig könnte die
Zusammenfassung mehrerer Endophänotypen-Merkmale zur Aufklärung der Genese von
Schizophrenie beitragen.

Introduction: Reduction of Mismatch negativity (MMN) in schizophrenic patients
is a well approved finding. Rs4680 influences as genetic variant of the enzyme
the activity of Catechol-O-Methyltransferase (COMT) in schizophrenic patients
and healthy controls. By using the concept of endophenotype in Schizophrenia
the influence of Rs4680 on MMN should be analysed. Methods: MMN and COMT
genotype of 77 schizophrenic patients and 92 healthy controls were analysed.
Psychopathological rating by PANSS was conducted and biographical data was
raised. Results: Rs4680 modulated MMN in schizophrenic patiens and healthy
controls. There was no difference of influence of rs4680 on MMN between both
groups. Conclusions: It could not be shown that rs4680 influences MMN
differently in schizophrenic patients than in healthy controls. Rs4680 could
not be proven to influence the life-time prevalence of Schizophrenia. In
future the pooling of several endophenotypical parameters could contribute to
the understanding of genesis of Schizophrenia.
