Die Arbeit besteht aus drei Teilen. Zuerst wurde der Einfluss von Ethanol auf
die mit Retinsäure hervorgerufene dopaminerge Differenzierung der humanen
Neuroblastomzelllinie SH-SY5Y untersucht. Es konnte gezeigt werden, dass die
Expression der kurzen Splicevariante des Dopamin D2 Rezeptors unter
Ethanolexposition für 28 Tage gegenüber der langen Splicevariante vermindert
war und dass sich diese Differenz auch nach 24 h Entzug nicht normalisiert
hatte. Morphologisch zeigten die Zellen eine gestörte neuronale
Differenzierung. Diese Störung ging einher mit einer kontinuierlich
reduzierten Expression des Raf-Kinase-Inhibitorproteins (RKIP;
Beobachtungszeitraum: 4 Wochen). Durch RKIP-Überexpression und siRNA-
vermittelte Herunterregulierung konnte die Bedeutung des RKIP für die
Differenzierung näher charakterisiert werden. Im zweiten Teil stehen die
-Carboline und ihre mögliche Bedeutung für die Pathogenese der Parkinsonschen
Erkrankung im Mittelpunkt. Es konnte herausgearbeitet werden, dass vor allem
solche -Carboline neurotoxisch wirken, die eine permanente kationische
Struktur aufweisen. Gleichzeitig konnte gezeigt werden, dass nichtkationische
-Carboline vergleichsweise wenig oder gar nicht toxisch sind und auch nicht
zu einer erhöhten ROS-Produktion führen. Davon ausgehend wurden
neuroprotektive und neuroregenerative Eigenschaften eines nichtkationischen
-Carbolins, dem 9-Methylnorharman, untersucht. In murinen embryonalen
mesenzephalen Primärzellen führte die Inkubation mit 9-Methylnorharman zu
einer Zunahme dopaminerger Neurone. In einem in vivo rescue Experiment wurde
Ratten für 4 Wochen kontinuierlich MPP+ in den linken Seitenventrikel
infundiert. Anschließend wurde den Tieren für 14 Tage 9-Methylnorharman oder
physiologische Kochsalzlösung infundiert. Die Behandlung mit 9-Methylnorharman
führte zu einer Normalisierung der Anzahl dopaminerger Neurone in der
Substantia nigra und des Dopamingehaltes im Striatum. Dies ging einher mit
einer verstärkten Expression neurotropher Faktoren wie BDNF, CNTF und ARMETL
(CDNF). Außerdem wurde ein Anstieg der Expression von Precerebellin
beobachtet. Wir haben hier erstmalig eine Substanz identifiziert, die auf
mehreren Ebenen in den Prozess der Degeneration dopaminerger Neurone eingreift
und in vivo neuroregenerative Wirkung zeigt.

dc.description.abstract
The work is devided in three topics. First, the impact of ethanol has been
investigated on retinoic acid induced differentiation of the human
neuroblastoma cell line SH-SY5Y. The expression of the short splice variant of
the dopamine D2 receptor was diminished compared to the long splice variant
during a 28 day ethanol exposition. This difference was not rescued after a 24
h withdrawal period. The morphology showed a disturbed neuronal
differentiation. This disturbance was accompanied by a continuous reduced
expression of the Raf-kinase inhibitor protein (RKIP). The importance of RKIP
for the neuronal differentiation was demonstrated by RKIP overexpression and
siRNA-mediated knock-down experiments showing accelerated and impaired
neuronal differentiation, respectively. The PKC-mediated ERK-1/2-activation
correlated with the RKIP-expression. Furthermore, the basal and forskolin-
stimulated intra cellular cAMP-value was twofold and tenfold enhanced in RKIP
over-expressing cells. The second part is dealing with -carbolines and their
importance for the pathogenesis of Prakinson’s disease. It has been shown that
those -carbolines, which are permanent cations, are neurotoxic, and those
which are non-cationic are mainly non-toxic and do not increase ROS-
production. Based on these findings, the neuroprotective and neuroregenerative
properties of the non-cationic -carboline 9-methyl-norharman have been
investigated in great detail. The incubation of murine embryonic mesencephalic
primary cells with 9-methyl-norharman increased the number of dopaminergic
cells. In an in vivo rescue experiment the neurotoxin MPP+ was continuously
infused into the left ventricle of rats for 28 days. The followed infusion of
9-methyl-norharman for 14 days led to a normalization of the number of
dopaminergic neurones in the substantia nigra and the contents of dopamine in
the striatum. This was accompanied by an enhanced expression of neurotrophic
factors like BDNF, CNTF and ARMETL1 (CDNF). Furthermore, the expression of
precerebellin was enhanced. For the first time, we identified a substance,
which interferes with several processes of the degeneration of dopaminergic
neurones and which demonstrated being neuroregenerative in vivo.
