Für verschiedene Tumorentitäten konnte gezeigt werden, daß Endothelin-1 (ET-1) sowohl bei der Tumorproliferation wie auch bei der Tumorangiogenese involviert ist. Der Endothelin-Rezeptoren A (ETA) scheint dabei eine entscheidende Rolle zu spielen. Für die Inhibierung der sog. ET-1–ETA–Achse wird daher eine neue Therapieoption postuliert. Auch Zellen des humanen Pankreaskarzinoms sezernieren ET-1. Ziel dieser Arbeit war, die Rolle des ET-Systems für die Pathogenese bzw. die Therapie des Pankreaskarzinoms zu evaluieren. Hierzu wurden humane Pankreaskarzinomzelllinien unterschiedlicher Differenzierungsgrade entweder mit exogenem ET-1 oder mit selektiven ETA- Antagonisten (LU-302146, BQ485) in vitro und in vivo behandelt. Bei den rezeptorpositiven Tumorzellen (MiaPaCa-2, AsPC-1, HPAF-2) gelang es über eine selektive ETA-Inhibition Tumorzellproliferation, Tumorwachstum und Tumordisseminierung dosisabhängig zu reduzieren. Auch die mikrovaskuläre Gefäßdichte konnte supprimiert werden, die erwartete Interaktion mit dem vaskulären Schlüsselfaktor VEGF konnte in vitro allerdings nicht verifiziert werden. Die Ergebnisse zeigen eine klinisch relevante Rolle der ET-1–ETA–Achse beim humanen Pankreaskarzinom. ETA-Antagonisten stellen damit eine potentiell neue Therapiestrategie für das Pankreaskarzinom dar, die insbesondere in Kombination mit konventionellen Therapieformen eine Option bieten könnte.
Endothelin-1 (ET-1) increases proliferation and angiogenesis in various types of cancer. Endothelin receptor A (ETA) seems to have a pivotal function. Therefore inhibition of the so-called ET-1-ETA-axis could be an innovative approach in anti-cancer therapy. Human pancreatic cancer cells also produce ET-1. The aim of this study was to analyze the role of the ET-system in pancreatic cancer and to evaluate whether there is a therapeutic implication. We treated human pancreatic cancer cell lines of varying differentiations either with exogenous ET-1 or with selective ETA antagonist (LU-302146, BQ485) in vitro and in vivo. In receptor-positive cell lines (MiaPaCa-2, ASPC-1, HPAF-2) we achieved reduction of tumour cell proliferation, tumour growth and tumour dissemination by ETA-blockade in a dose depending manner. Even microvessel density was reduced; however, an expected interaction with VEGF, a vascular key factor, could not be verified in vitro. The results confirm a clinically relevant role of ET-1-ETA-axis in human pancreatic carcinoma. ETA- antagonists represent a potential new therapeutic strategy for pancreatic carcinoma. Especially in combination with conventional forms of therapy ETA- inhibition provides a novel option.
