dc.description.abstract
1\. Microparticles prepared by the solvent evaporation (cosolvent) method:
Adjusting parameters, which affected the PLGA precipitation kinetics, provided
efficient ways to increase the encapsulation efficiency and control the
initial release. Addition of NaCl to the external aqueous phase increased the
encapsulation efficiency. The presence of ethanol in the external phase led to
porous microparticles with an increased initial release but a decreased
encapsulation efficiency. The initial release also decreased with decreasing
volume of the external phase and homogenization speed, as well as with
covering the preparation apparatus; however, these variations showed no
significant effect on the encapsulation efficiency. Varying formulation and
processing parameters (e.g., drug loading, volume of the external phase, using
low molecular weight PLGA) affected the initial release of microparticles but
not the drug release thereafter. The inclusion of medium chain triglycerides
(MCT) successfully shifted the tri-phasic release pattern of microparticles to
a continuous release profile. MCT led to the formation of microparticles with
an irregular surface and a highly porous inner structure. Differential
scanning calorimetry (DSC) revealed a high entrapment of MCT in the
microparticles and an unchanged glass transition temperature (Tg) of the PLGA.
2\. In situ microparticle (ISM) system: ISM showed a high initial release due
to the high porosity of microparticles. The initial release could be reduced
by increasing the polymer concentration, increasing the volume and viscosity
of the oil phase, and decreasing the drug loading. Introducing a partial
water-miscible solvent reduced the initial release from ISM, which could be
attributed to the formation of less porous microparticles. In vivo study in
rabbits showed a suppression of testosterone until day 29 after single
administration of ISM prepared with a solvent mixture of 80% w/w NMP and 20%
w/w triacetin. For ISM, the low molecular weight PLGA resulted in a lower
initial release than the high molecular weight PLGA. ISM prepared with PLGA
combinations showed a decreasing initial release with increasing low molecular
weight PLGA content. A slower solvent diffusion from the low molecular weight
PLGA solution droplets into the release medium led to a less porous structure
of the resulting microparticles, thus explaining the lower initial release.
PLGA with free carboxylic acid endgroups led to a slower drug release compared
to PLGA with esterified end groups. 6-month controlled release leuprolide ISM
could be obtained by blending poly(lactides) (PLA) with different molecular
weights.

1\. Mikropartikel, welche durch die Lösungsmittel-Evaporations-Methode mittels
Kosolventien hergestellt wurden: Die Einstellungen verschiedener Parameter,
die die PLGA-Ausfällung beeinflussten, erwiesen sich als effiziente Werkzeuge,
um die Verkapselungseffizienz und die Kontrolle der initialen Freisetzung zu
verbessern. Die Zugabe von NaCl zur externen wässrigen Phase erhöhte die
Verkapselungseffizienz. Die Anwesenheit von Ethanol in der äußeren Phase
erhöhte initialer Freisetzung, allerdings verringerter Verkapselungseffizienz.
Die initiale Freisetzung nahm auch bei einer Verringerung des Volumens der
äußeren Phase und der Homgenisationsgeschwindigkeit ab, ebenso bei Abdeckung
der Herstellungsapparatur. Diese Modifikationen wirkten sich nicht signifikant
auf die Verkapselungseffizienz aus. Veränderung der Formulierungs- und
Herstellungsparameter (z.B. Wirkstoffgehalt, Volumen der äußere Phase, PLGA
geringen Molekulargewichts) beeinflussten zwar die initiale Freisetzung
(Burst), jedoch nicht die nachfolgende. Mittelkettige Triglyzeride (MCT)
konnten erfolgreich eingesetzt werden, um eine kontinuierliche Freisetzung zu
erhalten. Sie führten zur Bildung von Mikropartikeln mit ungleichmäßiger
Oberfläche und einer hochporösen inneren Struktur. Mit der Differential-
Wärmestrom-Analyse (DSC) konnte der Einschluss von MCT bei gleichbleibender
Glasübergangstemperatur (Tg) von PLGA beobachtet werden. 2\. In situ
Mikropartikel (ISM) Systeme: ISM-Systeme zeigten eine hohe initiale
Freisetzung, welche wahrscheinlich auf die hohe Porosität der Mikropartikel
zurückzuführen ist. Die initiale Freisetzung konnte durch Erhöhung der
Polymer-Konzentration, der Menge und der Viskosität der Öl-Phase und durch
Senkung der Arzneistoffbeladung reduziert werden. Die Einführung eines
teilweise mit Wasser mischbaren Kosolvenz reduzierte die initiale Freisetzung
von Mikropartikel. Dieses könnte auf die in situ Bildung weniger poröser
Mikropartikel zurückzuführen sein. In vivo Untersuchungen in Hasen zeigten
eine Verringerung der Testosteron- Plasmakonzentration bis zum 29. Tag nach
einer einmaligen Verabreichung von ISM, die mit einem Lösungsmittelgemisch aus
80% w/v NMP und 20% w/v Triacetin hergestellt wurden. Resultierte die
Verwendung von PLGA mit niedrigem Molekulargewicht in ISM eine niedrigeren
initialen Freisetzung als ISM, welche mit PLGA höheren Molekulargewichts
formuliert wurden. Aus PLGA-Kombinationen hergestellte ISM zeigten eine
abnehmende initiale Freisetzung mit ansteigendem Gehalt an PLGA niedrigen
Molekulargewichts. Diese niedrigere initiale Freisetzung lässt sich durch die
langsamere Diffusion des Lösungsmittels von den niedriger molekulargewichtigen
PLGA-Lösungstropfen in das Freisetzungsmedium erklären, was zu einer weniger
porösen Struktur der resultierenden Mikropartikel führte. PLGA mit freien
Carboxylsäure-Endgruppen zeigte eine langsamere Arzneistoff-Freigabe im
Vergleich zu PLGA mit veresterten Endgruppen. Ein ISM System, welches
Leuprolid über 6 Monate kontrolliert freisetzte, konnte durch das Mischen von
Polylaktiden (PLA) mit unterschiedlichen Molekulargewichten erhalten werden.
