Ein Schwerpunkt der vorliegenden Arbeit lag in der Entwicklung eines Verfahrens zur Quantifizierung der antioxidativen Kapazität von exzidierter Humanhaut, sowie deren Erhöhung durch neuartige Radikalfänger (Nitroxyl- Radikale). Ein weiteres Anliegen dieser Arbeit bestand in der Untersuchung der Penetrationseigenschaften ausgewählter Nitroxyl-Radikale mit Hilfe der ESR- Spektroskopie und Tomografie. Außerdem sollte das Verhalten der Nitroxide Humanhaut nach UV-Bestrahlung untersucht werden. Als wichtigste Ergebnisse der Arbeit sind folgende Punkte zu nennen: • Die antioxidative Kapazität der Epidermis von exzidierter Humanhaut konnte bestimmt und mit dem stabilen Modell-Radikals Diphenylpicrylhydrazyl, mittels der UV/VIS-Spektroskopie, quantifiziert werden. • Topisch applizierte Antioxidantien konnten mit diesem Verfahren in situ auf ihr reduktives Potential hin getestet werden. • Die, in dieser Arbeit, topisch applizierten Spinsonden zeigten ein erhöhtes Reduktionsvermögen gegenüber DPPH. Die Nitroxide TEMPOL (4-Hydroxy-2,2,6,6-tertramethylpiperidin-1-oxyl) und DHME (5-Doxylhexansäuremethylester) reduzierten das Modell-Radikal DPPH effektiver als das Standardantioxidans Ascorbinsäurepalmitat. • Die antioxidative Kapazität von Humanhaut ließ sich mit Thiolgruppen-Hemmstoffen auf ein Minimum reduzieren • Es konnte gezeigt werden, dass die Anzahl an UV-induzierten Sonnenbrandzellen in exzidierter Humanhaut durch die Substanzen wie Ascorbinsäurepalmitat, TEMPOL und DHME verringert werden konnte • Es konnte mit Hilfe der ESR-Tomografie die Penetration der Nitroxyl-Radikale TEMPOL und DHME verfolgt werden. Außerdem konnten unterschiedliche Verteilungsprofile der beiden Spinsonden erstellt werden. Dabei war zu beobachten, dass sich TEMPOL vermehrt im mittleren und unteren Bereich der Epidermis und DHME im oberen Bereich der Dermis aufhielt. • Mit Hilfe der Hyperfeinaufspaltung der ESR- Spektren von TEMPOL und DHME konnte ein Polaritätsprofil der unmittelbaren Umgebung der Spinsonden festgestellt werden. Dabei hielt sich das hydrophilere TEMPOL in einer polareren Umgebung auf als das lipophilere Nitroxid 5-Doxylhensäuremethylester. • Mit der Reaktion der Spinsonden, nach Bestrahlung von exzidierter Haut mit UV-Strahlung (UV A/B und C), zu ESR- stummen Verbindung über die gesamte vermessen Hauttiefe konnte gezeigt werden, dass der Wirkungsbereich von UV-Strahlung über die Eindringtiefe hinausgeht. • Es konnte eine verstärkte Reduktion der Spinsonden DHME und TEMPOL, in der Haut, nach UV-Bestrahlung und nach Hemmung des antioxidative Systems durch SH- Reagenzien festgestellt werden. UV-induzierte Folgeprodukte, wie reaktive Sauerstoffspezies, wurden vermehrt durch, in der Haut verteilte, Nitroxide abgefangen.
The present thesis was focused on the development of a method to quantify the antioxidant capacity of excised human skin, and increase by novel radical scavengers (nitroxyl radicals).A second aim of this work was to study the penetration properties of selected nitroxyl radicals using EPR spectroscopy and tomography. Finally, the behavior of the nitroxides in human skin after UV irradiation is investigated. The main results of the investigations are: • The antioxidant capacity of the epidermis of excised human skin was determined and quantified as equivalent of the stable radical Diphenylpicrylhydrazyl, by means of UV/VIS spectroscopy. • It was possible to estimate, in situ, the reductive potential of topically applied antioxidants. • The topically applied spin probes showed an increased reducing activity compared to untreated skin. The nitroxides DHME (5-doxyl-hexanoic acid methylester) and TEMPOL (4-hydroxy-2,2,6,6-tertramethylpiperidine-1-oxyl) reduced the model radical DPPH more effective than the antioxidans ascorbic acid palmitate (used as standard). • The antioxidant capacity of human skin could be reduced to a minimum with inhibitors of sulfhydril-groups • It was shown that the number of UV-induced sunburn cells in excised human skin could be reduced by substances such as ascorbic acid palmitate, TEMPOL and DHME • The penetration of the nitroxyl-radicals TEMPOL and DHME could be pursued by means of EPR tomography. Also there could be seen different distribution profiles of the two spin label. It was an increase amount of TEMPOL in the middle and bottom of the epidermis whereas DHME was staying in the top of the dermis. • Also there could be detected a polarity profile of the immediate environment of the spin probes by means of hyperfine splitting of ESR spectra of TEMPOL and DHME. As a hydrophilic spin label TEMPOL remains in polar environment compared to the lipophilic nitroxide 5-doxyl hexanoic acid methylester. • After UV-irradiation (UV A /B and C) of excised skin, the reaction lead to ESR-silent spin probes over the entire measured skin depth. It could be shown that the area of influence of UV radiation goes beyond the penetration depth. • There was a greater reduction of the spin probes TEMPOL and DHME, in the skin, after UV- irradiation and inhibition of the antioxidant system by SH-reagents. UV- induced secondary products, like reactive oxygen species, could be scavenged by nitroxides.
