Innovative partikuläre Trägersysteme ermöglichen durch eine verzögerte
Wirkstofffreisetzung das Erreichen von einem konstanten Medikamentenspiegel.
Dies ist von größter Bedeutung für die lokale Dermatotherapie. Verbesserte
Penetrations- und Depoteigenschaften sowie auch ihre Funktion als
Impfstoffadjuvantien untermauern ihre Rolle als potentielle Kandidaten für das
gezielte Heranbringen von Wirkstoffen an bestimmte Zellpopulationen. In der
Tat können durch das Targeting der antigen-präsentierenden Zellen (APZ) der
Haut potente und effiziente Immunantworten hervorgerufen werden. Aus diesen
Gründen und zur Beseitigung von Sicherheitsbedenken wurde im Rahmen dieser
experimentellen Arbeit die Rolle der biologisch degradierbaren PLA-Partikel
untersucht. Ziele: Die Untersuchung der Penetrationseigenschaften der PLA-
Partikel in exzidierter menschlicher Haut, der möglichen Wirkstofffreisetzung
aus den Partikeln und der Aufnahme der Partikel durch die APZ der Haut mit dem
Ziel derer Anwendung als Träger für die lokale Dermatotherapie und als
Antigenträger für die transkutane Vakzinierung. Methoden: Untersuchung der
Penetrationseigenschaften der Partikel in exzidierter menschlicher Haut mit
der Hilfe der Fluoreszenzmikroskopie. Untersuchung der Wirkstofffreisetzung
mit Anwendung eines biphasischen (lipophil/hydrophil) Systems zur Simulation
der Interaktion der hydrophilen Partikelsuspension mit der lipophilen Haut.
Untersuchung der Partikelaufnahme durch APZ der Haut ex vivo am Modell der
exzidierten menschlichen Haut und in vitro in der Zellkultur mit der Hilfe der
Durchflusszytometrie. Ergebnisse und Diskussion: Nach Auftragen der
Partikelsuspension auf exzidierte menschliche Haut erfolgt die selektive
intrafollikuläre Penetration, eine Aggregatebildung durch Destabilisierung der
Partikel und anschließend die Farbstofffreisetzung welche zu einer Verfärbung
nicht nur der Epidermis sondern auch des follikulären Epithels und der
Talgdrüse führt. Die Aggregate im Infundibulum fungieren als Depot für die
verzögerte Freisetzung des Farbstoffes. Eine Penetration durch die
Hautbarriere und eine Aufnahme durch die APZ erfolgt nicht, sodass PLA
Partikel gute Kandidaten für die Entwicklung neuer Trägersysteme für die
lokale Therapie von Hautkrankheiten und Talgdrüse-assoziierten Erkrankungen
sind. Für Ihre Anwendung als Antigenträger für die transkutane Vakzinierung
müssen sie jedoch zuerst stabilisiert werden um die Aggregatebildung zu
vermeiden und die Aufnahme durch APZ der Haut zu ermöglichen.

dc.description.abstract
Novel drug delivery systems (DDS) enable a sustained drug release, thus
maintaining constant drug levels. The latter is of greatest importance for the
local dermatotherapy. Furthermore strong and efficient immune responses can be
induced by targeting vaccines to skin antigen-presenting cells. Because
vaccination efficacy has been shown to increase with particle-bound antigen,
this aspect of drug delivery with use of particulate antigen carriers is
especially interesting for the design of transcutaneous (t.c.) vaccination
systems. The need for a biodegradable DDS and antigen carrier eliminating
safety concerns set the focus on PLA-particles. Aims: In this study, we
investigated the penetration properties of polylactic acid (PLA) particles in
human skin explants and particle uptake by epidermal antigen presenting cells
(APCs). The aim was to assess the ability of biodegradable PLA-particles to
deliver incorporated fluorescent dyes to hair follicle-associated structures
and the suitability of polymeric particles as antigen-carrier systems for t.c.
vaccination. Methods: Penetration and kinetic of dye release from PLA-
particles was analyzed on cryosections of particle-treated skin explants using
fluorescence microscopy. The dye release was confirmed in vitro with use in a
hydrophilic/lipophilic system, which we designed to mimic the environment in
the hair follicle ducts. Particle translocation and uptake was studied by flow
cytometry and fluorescence microscopy of APCs isolated from skin explants
treated with PLA-particles. Results: We observed clustering of particles
leading to partial release of incorporated dye and staining of epidermis, hair
follicles and sebaceous glands. The latter retained dye for up to 24 hours.
Microscopic analysis of particles added to the biphasic in vitro system
revealed the formation of conglomerates at the interface between water and
organic solvent. Although PLA-particles could be taken up by APCs in vitro, no
particle uptake by APCs was detected after their topical application on
excised human skin. Conclusions: Upon application of PLA particles on human
excised skin, particles destabilization occurred with concomitant release of
the loaded dyes. The prolonged release of the incorporated drug and the drug
diffusion would enable to achieve constant drug levels within the hair
follicle, which is useful for the treatment of hair related diseases.
Furthermore, targeting of the sebaceous gland is of utmost importance for the
treatment of sebaceous gland related disorders. Thus PLA-particles represent
promising candidates for an efficient drug delivery in topical dermatotherapy.
For their use as antigen carriers though stabilization of PLA-particles has to
be achieved, since the cluster formation hindered the translocation of PLA-
particles through the skin barrier and their uptake by APCs.
