Aufgrund vorliegender Ergebnisse unserer Arbeitsgruppe mit Propranolol, einem spezifischen Inhibitor der Phosphatidat Phosphohydrolase, und D609, einem spezifischen Inhibitor der Phospholipase C konnte eine noch unbekannte Funktion des Diacylglycerols im Kontext mit dem Invasionsprozess von E. coli Shigella vermutet werden. Zur Klärung wurde ein Assay, bestehend aus Infektion von Hela-Zellkulturen mit E. coli Flexneri, Phospholipidextraktion, radioaktiver Phosphorylierung von Diacylglycerol mit der Diacylglycerol- Kinase, Chromatographie und Röntgenfilmexposition zur Detektion des durch Enzymkatalyse entstandenen Diacylglycerolphosphats, entwickelt. Im Rahmen der Optimierung des Versuchsaufbaus konnte Cardiolipin neben seiner bekannten Funktion als Aktivator der Diacylglycerol-Kinase erstmalig als Substrat des Enzyms nachgewiesen werden. Infektionsexperimente mit dem invasiven E coli Flexneri Stamm SC301 und dem non-invasiven Stamm SC300 zeigten eindeutige Banden auf Höhe des Diacylglycerolphosphat-Standards. Einen von der Infektionszeit abhängigen Anstieg der Diacylglycerol-Konzentration konnte für SC301 nicht nachgewiesen werden. Ebenso zeigten sich keine Unterschiede hinsichtlich der Diacylglycerol-Konzentration zwischen dem invasiven Stamm SC301 und dem nicht invasiven Stamm SC300. Die möglichen Ursachen der gefundenen Ergebnisse werden im Kontext der vorhandenen Literatur diskutiert.
Based on previous findings by our team involving propranolol, a specific inhibitor of phosphatidate phosphohydrolase, and D609, a specific inhibitor of phospholipase C, a hitherto unknown function of diacylglycerol in context with the invasion process of E. coli Shigella could be assumed. For clarification purposes, an assay, consisting of an infection of HeLa-cell cultures with E. coli Flexneri, phospholipid extraction, radioactive phosphorylation of diacylglycerol with diacylglycerol kinase, chromatography and X-ray film exposition was developed to detect the diacylglycerol-phosphate produced by enzyme catalysis. As part of the optimisation of the experimental setup, it was possible to establish for the first time that cardiolipin, besides its known function as an activator of diacylglycerol kinase, is a substrate of the enzyme. Infection experiments with the invasive E coli Flexneri strain SC301 and the non-invasive strain SC300 displayed distinct bands at the level of the diacylglycerol phosphate standard. For SC301, no increase controlled by the time of infection could be determined in the diacylglycerol concentration. At the same time, there was no difference regarding the diacylglycerol concentration between the invasive strain SC301 and the non-invasive strain SC300. Possible causes for these findings are discussed in the context of the available literature.
