Mit der Charakterisierung des Redifferenzierungsverhaltens dedifferenzierter Chondrozyten in 3D Kultursystemen konnte gezeigt werden, dass bei menschlichen Chondrozyten nach der 4. Passage in der Monolayerkultur eine irreversible Dedifferenzierung eintritt. Diese Zellen sind damit für die Verwendung als Basis für autologe Knorpelzelltransplantationen unbrauchbar, denn sie synthetisieren keine belastbare Knorpelmatrix mehr. Der Verlust des chondrogenen Potentials ging offensichtlich mit einer Hemmung der Interaktion von Signalproteinen des MAPKinase Signalweges einher. Durch eine Inhibition dieses essentiellen Signalweges kann in Chondrozyten direkt Apoptose induziert werden. Die Analyse spezifischer Integrinfunktionen zeigte, dass beta1-Integrine auf der Zelloberfläche von Chondrozyten mit weiteren Rezeptorkomplexen, die z. T. katabolen Systemen zuzuordnen sind, kooperieren, als Hinweis auf ihren multifunktionellen Charakter. Die Wirkungen pro- inflammatorischer Zytokine wie IL-1beta und TNFalpha in Knorpelzellkulturen beinhalteten eine Hemmung der Synthese knorpelspezifischer Matrixproteine, die Induktion von katabolen und inflammatorischen Enzymen (MMPs und COX-2), und die Einleitung einer Apoptose, erkennbar an typischen morphologischen Kennzeichen und einer vermehrten Caspase-3 Aktivität. Hierbei wurde deutlich, dass verschiedene Phytopharmaka die katabolen Zytokinwirkungen im Knorpel partiell inhibieren konnten und damit als Ergänzung für die Arthrosetherapie interessant sind. Als möglicher protektiver Wirkungsmechanismus wurde für Curcumin eine NF-κB Hemmung auf verschiedenen Ebenen diskutiert. Im Folgenden wurde die Idee einer lokalen Entzündungs-Hemmung durch anti-inflammatorische Zytokine im Knorpel verfolgt. Die Untersuchungen der direkten Wirkungen von rekombinanten IL-10 auf Knorpelzellen ergaben eine Hemmung TNFalpha induzierter pro-apoptotischer Wirkungen über die Beeinflussung intrinsischer Apoptosewege und eine Stimulation der Kollagen Typ II mRNA Expression durch IL-10. Eine Entzündungshemmung über die Suppression der Expression pro- inflammatorischer Zytokine durch IL-10 wie für andere Zelltypen beschrieben, deutete sich nur für IL-1beta und TNFalpha an. Experimente mit dem IL-10 Überexpressionsvektor führten zu einem effektiven Überexpressionsmodell für IL-10 in Knorpelzellen, in dem die in Experimenten mit rekombinanten IL-10 erzielten Ergebnisse weitgehend Bestätigung fanden und keine Minderung des differenzierten chondrozytären Phänotyps erkennbar war.
The characterization of the redifferentiation capacity of chondrocytes in alginate or high-density culture which were dedifferentiated during expansion in monolayer culture indicated that human articular chondrocytes undergo irreversible dedifferentiation after the fourth monolayer passage. Irreversible dedifferentiated chondrocytes produced a non-specific extracellular matrix. Hence, these cells are not suitable for the use in autologous chondrocyte transplantation approaches. The loss of their chondrogenic potential was strongly associated with an inhibition of the interaction of signalling proteins of the MAPKinase pathway. The inhibition of this signalling cascade induced chondrocyte apoptosis. The analysis of chondrocytes integrins revealed that β1-integrins which are important cell surface signal transduction receptors on chondrocytes intimately cooperate with other receptor complexes associated with catabolic processes suggesting their multifunctional character in chondrocytes. The effect of pro- inflammatory cytokines IL-1β and TNFα on chondrocyte cultures included the inhibition of cartilage-specific matrix synthesis, the induction of catabolic and inhibitory enzymes (e.g. matrix metalloproteinases and cyclo-oxygenase-2) as well as the induction of apoptosis. Chondrocyte apoptosis was characterized by typical morphological features and the induction of caspase-3 activation. Dietary phytopharmaca were able to partly inhibit the effects of pro- inflammatory cytokines on articular chondrocytes suggesting them as an additional tool in osteoarthritis therapy. Inhibition of the transcription factor NFκB was discussed as a possible anti-inflammatory mechanism of curcumin. Additionally, the hypothesis of chondroprotection by the anti- inflammatory cytokine IL-10 was further studied. IL-10 modulated TNFα-induced intrinsic apoptotic pathways in chondrocytes and directly stimulated gene expression of the cartilage-specific matrix protein type II collagen. Furthermore, IL-10 had inhibitory effects on the gene expression of pro- inflammatory cytokines TNFα and IL-1β. The analysis of the effects of adenoviral IL-10 overexpression in chondrocytes supported the results produced with recombinant cytokines.
