Frühere Arbeiten zeigten vielfältige Interaktionen zwischen dem
Fettgewebshormon Leptin und dem Immunsystem. In Bezug auf T-Lymphozyten konnte
unter Leptineinfluss eine Präferenz zugunsten der IFN-γ dominierten
Th1-Antwort gezeigt werden, während Th2-spezifische Immunreaktionen
gleichzeitig gehemmt wurden. Das Ziel der vorliegenden Arbeit war, den
Einfluss von Leptin auf die in vitro Polarisierung von T-Helferzellen zu
charakterisieren. Naive T-Helferzellen wurden hierzu nach Präinkubation in An-
oder Abwesenheit von Leptin unter antigenspezifischen sowie -unspezifischen
Bedingungen polarisiert. Quantitative Auswertungen erfolgten anhand
durchflusszytometrischer Analysen. Zusammenfassend konnte mit der vorliegenden
Arbeit der Einfluss von Leptin auf die in vitro Polarisierung von naiven
T-Helferzellen eindeutig gezeigt werden. Während im antigenunspezifischen
System die Th1-Polarisierung gefördert und die Th2-Polarisierung supprimiert
wird, ist der Einfluss auf das antigenspezifische System vielfältiger und
stärker abhängig von den Kofaktoren der Antigenpräsentation sowie der Art der
Lymphozytenstimulation. Die antigenspezifische Stimulation kommt im Vergleich
zur unspezifischen Stimulation in vivo Vorgängen näher und gibt einen Einblick
darauf, dass die systemischen Einflüsse von Leptin auf immunologische Prozesse
weitaus komplexer und facettenreicher sind, als bisher in der Literatur
postuliert wurde.

dc.description.abstract
Previous publications have shown a wide range of interaction between leptin
and the immune system. In terms of T-lymphocytes leptin modulates immune
responses in favor of a IFN-γ dominated Th1 response. Further investigation
show that Th2 lymphocytes are inhibited by leptin. The aim of this study was
to investigate the influence of leptin on the in vitro polarization of
T-helper cells. Naive T-helper cells were preincubated in the presence or
absence of leptin. Afterwards polarization was carried out under non-specific
or antigen-specific conditions. The quantitative evaluation was accomplished
by flow cytometric analysis. In the present study, the influence of leptin on
the in vitro polarization of naive T-helper cells was clearly demonstrated.
While in the non-specific system Th1 polarization was promoted and Th2
polarization was actively suppressed, the effect of leptin on the antigen-
specific system was more diverse and dependent on the cofactors of antigen
presentation and lymphocyte stimulation. The antigen-specific stimulation is
comparable to in vivo processes and gives an insight to the fact that the
systemic effects of leptin on immunological processes are more complex than
previously postulated in the literature.
