Localized signaling and communication during yeast mating

Abstract

Communication between cells within the same population is an essential requirement for the dynamic adaptation of a cellular phenotype to other individuals in the same population. This thesis studies the pheromone response of budding yeast, a prototypical eukaryotic signaling pathway employing cell- cell communication. We developed a non-invasive assay to directly quantify extracellular concentrations of signaling molecules in vivo. This assay was used to show that budding yeast cells actively regulate their extracellular pheromone profile in order to divide the population into growing and diploid- forming cells. This allows the population to execute a "division of labor", where diploids are selectively formed in spots with a high local cell density, whereas the remaining areas remain in a continued mitosis of haploid cells. Furthermore, I present computational results mapping distinct parts of the pheromone signaling cascade to dynamic functions. By using stochastic mathematical modeling I show that the receptor and G protein activation in early parts of the signaling cascade have an intrinsic ability to reduce noise by accumulating extracellular signals. However, this accumulation can not keep up a tight localization of later signaling components regulating the change of cellular shape in the direction of a mating partner. This can be achieved by the formation of a large multi-protein complex, as it is showed by analysis of partial differential equation systems treating the formation of those complexes only. The results of this thesis illustrate how yeast cells actively regulate their intercellular communication within a population and how distinct parts of the signaling cascade are employed for efficient detection of the extracellular pheromone signal. The developed methods and frameworks are applicable to a wide range of biological problems.

Die Kommunikation zwischen unterschiedlichen Zellen der gleichen Population ist eine essentielle Vorraussetzung für die dynamische Anpassung eines Zellphänotyps abhängig vom Rest der Population. Diese Arbeit beschäftigt sich mit der Pheromonantwort der Hefezelle, ein prototypischer eukaryontischer Signalweg, welcher Zell-Zell-Kommunikation realisiert. Wir haben eine nicht- invasive Methode entwickelt die es erlaubt extrazelluläre Signalmoleküle in vivo zu quantifizieren. Diese Methode wurde verwendet um zu zeigen dass Hefezellen ihr extrazelluläres Pheromonprofil optimieren um die Gesamtpopulation in weiter wachsende und diploid formende Teilpopulationen zu spalten. Dies erlaubt der Population eine "Arbeitsteilung" zu realisieren in der Diploide selektiv in Gebieten mit hoher lokaler Zelldichte geformt werden, wobei der Rest der Population in der haploiden Mitose verbleibt. Weiterhin präsentiere ich Simulationsergebnisse welche bestimmte Teile der Pheromonantwort direkt mit biologischen Funktionen assoziieren. Mit einem stochastischen mathematischen Modell konnte ich zeigen, dass die Rezeptor- und G-Protein-Aktivierung während der frühen Pheromonantwort eine intrinsische Fähigkeit zur Störungskompensierung haben indem sie extrazelluläres Signal über die Zeit akkumulieren. Diese Akkumulation kann aber keine starke Lokalisierung der späteren Proteinkomplexe in Richtung der Partnerzelle aufrecht erhalten. Dies kann durch die Bildung eines Multi-Protein-Komplexes erreicht werden wie ich es durch Analyse partieller Differentialgleichung zeige welche ausschließlich die Bildung dieses Komplexes behandeln. Die Ergebnisse dieser Arbeit zeigen wie Hefezellen aktive ihre interzelluläre Kommunikation regulieren und wie spezifische Teile der Signalkaskade genutzt werden um das extrazelluläre Pheromonsignal effizient wahrzunehmen. Die entwickelten Methoden und Analysen sind auf eine große Klasse biologischer Problem anwendbar.
