Several Enterobacteriaceae are capable of producing phosphoethaolamine (pEtN) celluloses as part of the protective biofilms they construct in challenging environment. The composite of proteins (e.g. curli) and pEtN cellulose promotes biofilms with enhanced elasticity and generates hallmark macrocolonies with wrinkling and ridging phenotype. Genetic analysis revealed that approximately half of glycosyl units in celluloses are functionalized with pEtN at C-6 position. Yet, no systematic insights into the modification pattern of cellulose and its impact on biofilm formation exist, since the complete structural information is altered during isolation and purification from the microbial matrix. Synthetic pEtN oligosaccharides are therefore ideal references to deepen the chemical characterization of Escherichia coli (E. coli) biofilm. The ultimate goal of this thesis is to establish a structure-property correlation in bacterial biofilms and produce pEtN-cellulose based material with tunable properties. To achieve this goal, a collection of well-defined pEtN oligosaccharides are prepared using automated glycan assembly (AGA) along with post-AGA manipulations, and used to shed light on how the substitution pattern modulates structure and biological and materials properties. In chapter 2, I developed a general on-resin method to access zwitterionic pEtN oligosaccharides. After obtaining the glycan backbone by AGA, I addressed several issues related to synthesis of pEtN oligosaccharides on solid support including the phosphorylation, deprotection and purification steps. On-resin phosphorylation was performed using H-phosphonate or phosphoramidite reagents. The utilization of H-phosphonate resulted in higher isolated yields compared to the operation with phosphoramidite. Treatment with sodium methoxide led to a short deprotection step. Final global deprotection and purification guaranteed a repertoire of pEtN oligosaccharides with different amounts and patterns of pEtN oligosaccharides in a quick fashion. In chapter 3, three systems with different levels of complexity were introduced to understand biofilm formation. All systems were based on synthetic pEtN oligosaccharides with different lengths, amounts and patterns of substitutions. Nanocomposites consisting of amyloidogenic peptide R5 co-assembled with pEtN celluloses revealed that the pEtN pattern affected the peptide fiber growth rate and modulated the adhesion of the matrices. The co-assembly of amyloidogenic protein CsgA and some pEtN hexasaccharide showed structural changes in the amide III band, indicating that the glycans affected the conformation of CsgA. Lastly, I applied synthetic pEtN celluloses to specific E. coli biofilms to study the changes in biofilm organization. Addition of different pEtN glycans onto bacteria resulted in less biofilm matrix production and granular patterns when treated with longer pEtN glycan chains with free-reducing end, suggesting that the natural zwitterionic glycopolymers were capable of disrupting E. coli biofilm production. In chapter 4, I employed pEtN celluloses to reveal the importance of pEtN substitution patterns in protein-glycan interactions. The biosynthesis of pEtN cellulose in bacteria requires multiple enzymes. BcsZ hydrolyzes internal β-1,4-glycosidic bond in cellulose during biosynthesis. Different constructs of catalytically inactive BcsZ and pEtN hexasaccharide complex demonstrated that certain pEtN patterns were decisive for BcsZ recognition and activity. Other protein-pEtN cellulose interactions were identified using the glycan array technology. Linker-coupled pEtN oligosaccharides were printed onto glass slides and incubated with various lectins. Binding to humoral lectins suggested that the interactions were specific and seemed to be related to the amount and pattern of pEtN substitution. Combined, the synthetic pEtN oligosaccharides prepared and investigated in this thesis served as starting point to drive the development of novel pEtN cellulose-based materials.

Die Zusammensetzung aus Proteinen (z.B. Curli) und pEtN-Cellulose fördert Biofilme mit erhöhter Elastizität und erzeugt charakteristische falten- und rillenphänotypische Makrokolonien. Genetische Analysen ergaben, dass etwa die Hälfte der Glycosyl-Einheiten in Cellulosen an der C-6-Position mit pEtN funktionalisiert sind. Es existieren jedoch keine systematischen Einblicke in das Modifikationsmuster der Cellulose und deren Einfluss auf die Biofilmbildung, da die vollständigen Strukturinformationen während der Isolierung und Reinigung aus der mikrobiellen Matrix verändert werden. Synthetische pEtN-Oligosaccharide sind daher ideale Referenzen, um die chemische Charakterisierung von E. coli-Biofilmen zu vertiefen. Das Hauptziel dieser Arbeit ist es, eine Struktur-Eigenschafts-Beziehung in bakteriellen Biofilmen herzustellen und pEtN-Cellulose-basierte Materialien mit modifizierbaren Eigenschaften zu produzieren. Um dieses Ziel zu erreichen, wird eine Sammlung gut definierter pEtN-Oligosaccharide unter Verwendung der automatisierten Glykan-Assemblierung (AGA) sowie nach AGA-Manipulationen hergestellt und verwendet, um zu untersuchen, wie das Substitutionsmuster die Struktur und die biologischen und Materialeigenschaften verändert. In Kapitel 2 habe ich eine allgemeine On-Resin-Methode entwickelt, um zwitterionische pEtN-Oligosaccharide zugänglich zu machen. Nach Gewinnung des Glykan-Rückgrats durch AGA befasste ich mich mit mehreren Problemen in Zusammenhang mit der Synthese von pEtN-Oligosacchariden auf Festphase, einschließlich der Phosphorylierung, Entschützung und Reinigung, angesprochen. Die On-Resin-Phosphorylierung wurde unter Verwendung von H-Phosphonat- oder Phosphoramidit-Reagenzien durchgeführt. Die Verwendung von H-Phosphonat führte zu höheren Ausbeuten im Vergleich zur Arbeit mit Phosphoramidit. Die Behandlung mit Natriummethoxid führt zu einem schnellen Entfernen der Schutzgruppe. Die abschließende globale Entschützung und Reinigung ermöglichten ein Repertoire an pEtN-Oligosacchariden mit unterschiedlichen Mengen und Mustern von pEtN-Oligosacchariden in schneller Weise. In Kapitel 3 wurden drei Systeme mit unterschiedlicher Komplexität verwendet, um die Biofilmbildung zu verstehen. Alle Systeme basierten auf synthetischen pEtN-Oligosacchariden mit unterschiedlichen Längen, Mengen und Substitutionsmustern. Nanokomposite, bestehend aus amyloidogenem Peptid R5, das mit pEtN-Cellulosen zusammengebaut wurde, zeigten, dass das pEtN-Muster die Wachstumsrate der Peptidfasern beeinflusste und die Adhäsion der Matrizen veränderte. Der Zusammenbau des amyloidogenen Proteins CsgA und einiger pEtN-Hexasaccharide zeigte strukturelle Veränderungen in der Amide-III-Bande, was darauf hindeutet, dass die Glykane die Konformation von CsgA beeinflussten. Schließlich habe ich synthetische pEtN-Cellulosen auf spezifische E. coli-Biofilme angewendet, um die Veränderungen in der Biofilmorganisation zu untersuchen. Die Zugabe verschiedener pEtN-Glykane zu Bakterien führte zu einer geringeren Biofilmmatrixproduktion und granulären Mustern, wenn sie mit längeren pEtN-Glykan-Ketten mit freiem reduzierenden Ende behandelt wurden, was darauf hindeutet, dass die natürlichen zwitterionischen Glykopolymere in der Lage waren, die E. coli-Biofilmproduktion zu stören. In Kapitel 4 habe ich pEtN-Cellulosen verwendet, um die Bedeutung der pEtN-Substitutionsmuster bei Protein-Glykan-Interaktionen aufzuzeigen. Die Biosynthese von pEtN-Cellulose in Bakterien erfordert mehrere Enzyme. BcsZ hydrolysiert während der Biosynthese die interne β-1,4-glycosidische Bindung in Cellulose. Verschiedene Konstrukte des katalytisch inaktiven BcsZ und des pEtN-Hexasaccharid-Komplexes zeigten, dass bestimmte pEtN-Muster entscheidend für die Erkennung und Aktivität von BcsZ waren. Andere Protein-pEtN-Cellulose-Interaktionen wurden unter Verwendung der Glykan-Array-Technologie identifiziert. Linker-gekoppelte pEtN-Oligosaccharide wurden auf Glasplatten gedruckt und mit verschiedenen Lektinen inkubiert. Die Bindung an humorale Lektine deutete darauf hin, dass die Interaktionen spezifisch waren und mit der Menge und dem Muster der pEtN-Substitution zusammenzuhängen schienen. Zusammengefasst dienten die in dieser Arbeit hergestellten und untersuchten synthetischen pEtN-Oligosaccharide als Ausgangspunkt, um die Entwicklung neuartiger pEtN-Cellulose-basierter Materialien voranzutreiben.
