Die Oxidation von Wasser durch Cyanobakterien, Algen und Pflanzen stellt einen zentralen Schritt in der oxygenen Photosynthese dar. Das katalytische Zentrum – ein Mangan-Calcium-Komplex und seine Proteinumgebung – im Photosystem II (PSII) durchläuft zyklisch vier semistabile Zustände (S0 bis S3). Der vollständige Reaktionszyklus umfasst das Entfernen von 4 Elektronen und 4 Protonen von dem Mn-Komplex. Mit zeitaufgelösten photothermischen Experimenten konnte ich Volumenänderungen des PSII-Proteins beobachten, die mit lokaler Ladungsbildung durch lichtinduzierten Elektronentransfer (Kontraktion) und ladungs-kompensierenden Protonenbewegungen (Expansion) zusammenhängen. Zwei zuvor unsichtbare Protonenabgaben des Mn-Komplexes konnten so beobachtet und zwei Lücken im Reaktionszyklusmodell der photosynthetischen Wasseroxidation gefüllt werden. Im S2→S3-Übergang des klassischen S-Zyklus wird durch Deprotonierung ein Intermediat deutlich vor dem Elektronentransfer (ET) zum Oxidans (YZox) gebildet. Charakteristisch für den ratenlimitierenden Elementarschritt (τ ≈ 35 µs bei 20 °C) der Überführung des Protons vom Mn- Komplex zur Wasser-Protein-Grenzschicht sind: eine hohe Aktivierungsenergie (Ea = 0,46 ± 0,05 eV), ein ausgeprägter H/D-Isotopenaustausch-Effekt (KIE ≈ 5) und eine deutliche pH-Abhängigkeit (τ ≈ 20 µs bei pH 7,0 und τ ≈ 60 µs bei pH 5,5). Die Eigenschaften eines Protonentransferschritts im S0→S1-Übergang waren ähnlich (τ ≈ 100 µs bei 20 °C, Ea = 0,34 ± 0,08 eV, KIE ≈ 3, τ5.5/τ7.0 ≈ 1,5). Die Abgabe des Protons erfolgte hier jedoch nach dem ET zu YZox. Durch die Entdeckung zweier weiterer Intermediate im Reaktionszyklus der photosynthetischen Wasseroxidation wird eine zeitliche Sequenz strikt alternierender Entfernung von Elektronen und Protonen vom Mn-Komplex etabliert. In einem zweiten experimentellen Ansatz konnten Prozesse im Hundert-ns-Zeitbereich mit der Methode der Photoakustik beobachtet werden, die auf der Detektion einer Druckwelle nach Anregung von PSII-Proben mit ns- Laserpulsen basiert. Diese Prozesse mit Zeitkonstanten von ~350 ns bei 25 °C gehen ET-Reaktionen am Mn-Komplex deutlich voraus und sind in der Vergangenheit als energetische Relaxation des oxidierten YZox interpretiert worden. Die photoakustischen Experimente zeigen, dass speziell bei Anregung von PSII im S2-Zustand nukleare Umordnungen ausgelöst werden, die zu einer Kontraktion von mindestens 30 Å3 führen. Im S3-Zustand fiel die ns-Kontraktion fünfmal kleiner aus, in S0 und S1 war keine Volumenänderung in diesem ns- Zeitbereich zu beobachten. Daher wird in der Schrift diskutiert, dass die Bildung eines Intermediatzustands spezifischer atomarer Struktur das oxidierte Tyrosin YZox durch energetische Relaxation stabilisiert und den Mn-Komplex auf die folgende Entfernung des Protons und Elektrons vorbereitet.
Water oxidation by cyanobacteria, algae, and plants is pivotal in oxygenic photosynthesis, the process that powers life on earth. The catalytic site, a manganese-calcium complex and its protein environment in photosystem II (PSII), cycles through four semi-stable states (S0 to S3). Each turnover of the reaction cycle of photosynthetic water oxidation requires the removal of 4 electrons and 4 protons from the Mn-complex. In time-resolved photothermal beam deflection (PBD) experiments, it was possible to monitor the apparent volume changes of the PSII protein that are associated with charge creation by light-induced electron transfer (contraction) and charge-compensating proton relocation (expansion). Two previously invisible proton removal steps were detected, thereby filling two gaps in the basic reaction-cycle model of photosynthetic water oxidation. In the S2→S3 transition of the classical S-state cycle, an intermediate is formed by deprotonation clearly before electron transfer to the oxidant (YZox). The rate-determining elementary step (τ ≈ 35 µs at 20 °C) in the long-distance proton relocation toward the protein-water interface is characterized by a high activation energy (Ea = 0.46 ± 0.05 eV), strong H/D kinetic isotope effect (KIE ≈ 5), and pronounced pH dependence (τ of 20 µs at pH 7.0 vs τ of 60 µs at pH 5.5). The characteristics of a proton transfer step detected in the S0→S1 transition are similar (τ ≈ 100 µs at 20 °C, Ea = 0.34 ± 0.08 eV, KIE ≈ 3, τ5.5/τ7.0 ≈ 1.5); however the proton removal from the Mn-complex proceeds after electron transfer to YZox. By discovery of the transient formation of two further intermediate states in the reaction cycle of photosynthetic water oxidation, a temporal sequence of strictly alternating removal of electrons and protons from the catalytic site is established. In a second experimental approach, it was possible to monitor processes in the time domain of hundreds of nanoseconds by a photoacoustic method involving pressure-wave detection after excitation of PSII membrane particles by ns-laser flashes. Those processes with time constants of ~350 ns at 25 °C precede charge transfer reactions of the Mn complex clearly and have been assigned previously to energetic relaxation of the oxidized tyrosine (YZox). Evaluation of photoacoustic experiments shows that especially for excitation of PSII in the S2 state, nuclear rearrangements are induced which amount to a contraction of PSII by at least 30 Å3. In the S3 state, the 350-ns-contraction is about 5 times smaller whereas in S0 and S1 no volume changes are detectable in this ns-time domain. It is discussed that aside from stabilization of YZox by energetic relaxation the formation of an intermediate state of specific (but still unknown) atomic structure prepares the Mn-complex entity for the subsequent proton removal and electron transfer.
