Das TT-Virus, Torque Teno Virus, (TTV) wurde 1997 als erstes einzelsträngiges
zirkuläres DNA-Virus aus einem Patienten isoliert. Es weist eine Seroprävalenz
von über 90% auf. TTV wird gemeinsam mit dem Torque-teno-mini-virus (TTMV),
dem Small Anellovirus (SAV) und dem Torque-teno-midi-virus (TTMDV) in das
Genus Anellovirus eingeordnet. Das hüllenlose TT-Virus ist etwa 30-32nm groß.
Das Genom besitzt ca. 3800nt und ist negativ orientiert. Der codierende
Bereich umfasst ca. 2600nt während die nichtkodierende Region (non-coding
region; NCR) etwa 1200nt lang ist. Die NCR beinhaltet einen 113nt langen
Abschnitt, der einen GC-Gehalt von bis zu 90% aufweist. Innerhalb der hoch-
konservierten GC-reichen Region befinden sich Strukturen, die für die virale
Replikation und Transkription notwendig sind. TTV wird in fünf Genogruppen
eingeteilt, denen mindestens 39 Genotypen angehören. Die Divergenz auf DNA-
Ebene beträgt zwischen den Genotypen bis zu 40%. Für den TTV-Genotyp P/1C1
wurde die Synthese von vier mRNA-Klassen nachgewiesen (2,8kb, 1,2kb, 1,0kb und
0,6kb) von denen ausgehend sieben Proteine exprimiert werden. Die Struktur des
Proteins das durch den Offenen Leserahemen 1 (ORF1) codiert wird, lässt eine
Funktion als Kapsidprotein oder Replikase vermuten. In der vorliegenden Arbeit
wurden die TTV-Isolate P/1C1 und J01B6 untersucht, die nach phylogenetischen
Analysen, in denen die Aminosäuresequenz des ORF1 von 31 TTV-Isolaten
verglichen wurden, der Genogruppe 1 (P/1C1) und J01B6 der Genogruppe 5 (J01B6)
zuzuordnen sind. Es wurden zwei C-terminale Fragmente von ORF1-P/1C1 und ein
C-terminales Fragment von ORF1-J01B6 als Fusionsproteine an eine
Gluthation-S-Transferase (GST) gekoppelt in E. coli-Zellen exprimiert. Die
exprimierten Proteine wurden mittels SDS-PAGE von den bakteriellen Proteinen
getrennt, aus dem Gel eluiert und mit den rekombinanten Proteinen Kaninchen
zur Gewinnung von polykonalen Antiseren für 80 bzw. 81 Tage immunisiert. Alle
hergestellten Antiseren detektieren im Western-Blot die viralen Proteine, die
zur Immunisierung eingesetzt wurden. Gleichfalls wurden Antikörper gegen das
Fusionsprotein GST gebildet. Die gewonnenen Antiseren zeigen keine
Kreuzreaktivität mit bakteriellen Proteinen. Auch das ORF1 Protein des jeweils
anderen Genotyps wurde nicht erkannt, wie die Western-Blot-Analysen zeigten.
Für Immunfluoreszenzuntersuchungen wurden Plasmide, die die DNA-Sequenz des
ORF1 eines der beiden Virusisolate enthielt, in 293- bzw. HeLa-Zellen
transfiziert. In der anschließenden Immunodetektion konnte gezeigt werden,
dass jeweils das spezifische Antiserum, das gegen Proteine von P/1C1 bzw.
J01B6 gerichtet ist, ein positives Signal in Kompartimenten innerhalb des
Zellkerns liefert. Eine Kreuzreaktivität mit zellulären Proteinen wurde nicht
beobachtet. Eine Lokalisation von ORF1 im Zellkern ist sowohl mit der Funktion
als Kapsidprotein als auch als Replikase vereinbar. Die in der vorliegenden
Arbeit hergestellten Antiseren sind in der Lage, ORF1-Proteine der TTV-
Genotypen P/1C1 und J01B6 sensitiv und spezifisch sowohl im Western-Blot als
auch in Immunofluoreszenzanalysen zu detektieren. Damit stellen sie ein
wichtiges Werkzeug für weitergehende Untersuchungen an TT-Viren dar, die dazu
beitragen werden, Erkenntnisse über die Molekularbiologie und Pathogenität von
TTV zu gewinnen.

The TT-virus, Torque Teno Virus, (TTV) was isolated from a patient in 1997. It
is the first single-stranded circular DNA-Virus in humans. Its seroprevalence
lies over 90%. TTV is a member of the Genus Anellovirus that also contains the
Torque-teno-mini-virus (TTMV), the Small Anellovirus (SAV) and the Torque-
teno-midi-virus (TTMDV). The 30-32nm small TT-Virus is non-enveloped and the
negatively orientated genome contains approx. 3800nt. The coding region is
2600nt, and the non-coding region (NCR) is about 1200nt long. Within the NCR
lies a GC-rich part of 113nt which shows motives that are responsible for
replication and transcription. There are five genogroups of TTV with at least
39 genotypes. On DNA-level they diverge up to 40%. For the Genotype P/1C1 four
classes of mRNA was shown (2.8kb, 1.2kb, 1.0kb and 0.6), from which seven
proteins are expressed. The protein corresponding to the openreading frame 1
(ORF1) is supposed to work as a capsid protein or a replicase. In this work
the TTV-isolataes P/1C1 and J01B6 were investigated. Analysing the amino acid
sequence of 31 different TTV isolates, P/1C1 can be assigned to Genogroup 1
and J01B6 to Genogroup 5. Two c-terminal parts of ORF1-protein of P/1C1 and
one c-terminal fragment of the ORF1-protein of J01B6 were linked to a
Gluthation-S-transferase-(GST-) protein and expressed in E. coli-cells. The
GST-fusion proteins were separated from bacterial proteins by SDS-PAGE and
elution from the gel afterwards. The recombinant proteins were injected into
rabbits to immunise the animals and gain polyclonal antibodies against the
viral proteins. Period of immuinisation was 80 or 81 days respectively. The
won antisera did not show cross reactivity with bacterial proteins or the ORF1
protein of the other viral genotype in Western-Blot-Analyses. For
immunofluorenscence investigations a plasmid carrying the complete ORF1-DNA-
sequence of each viral isolate was transfected into 293- and HeLa-cells
respectively. In the following detection antisera detected viral proteins
specifically without crossreaction against cell proteins or ORF1 proteins of
the other viral genotype. The reaction of the sera was located in
compartiments within the cell nucleus. These findings are consistent with the
thesis that the ORF1-proteins work as a capsidprotein or replicase. The
generated antisera are able to detect ORF1-proteins of the TTV-genotypes P/1C1
and J01B6 with a high sensitivity and specificity in Western-Blot- and
Imnnofluorescence-Analyses. So they are helpful tools to carry out further
investigations to advance the knowledge about TTV’s molecular biology and
pathogenicity.
