Das Melanom ist ein hoch maligner Tumor, der sich gegenüber gängigen
Behandlungsmethoden weitgehend resistent erweist. Die Therapieresistenz beruht
dabei vor allem auf der Entwicklung einer Apoptosedefizienz. Ziel dieser
Arbeit war die Darstellung einzelner apoptotischer Regulationsprozesse im
Gesamtkontext der Apoptose. Hierzu wurde einerseits die Wirksamkeit Apoptose-
induzierender Stimuli auf Melanozyten und Melanomzellen quantifiziert und
andererseits deren Einfluss auf die Expression einzelner Apoptoseregulatoren
in kultivierten normalen humanen Melanozyten und Melanomzellen untersucht und
verglichen. Die Melanomzelllinien zeigten zum Teil Resistenzen gegenüber der
Apoptoseinduktion mit CH-11, IFN-γ (Interferon-γ), LPS (Lipopolysaccharide)
und TNF-α (Tumornekrosefaktor-α), so dass zwischen resistenten und sensitiven
Melanomzelllinien unterschieden werden konnte. In allen untersuchten
Melanomzelllinien konnte im Gegensatz zu Melanozyten eine stärkere Expression
der IAPs (inhibitors of apoptosis proteins) sowie von cFLIP (cellular FLICE
inhibitory protein) und Smac (second mitochondria-derived activator of
caspases) gefunden werden, wobei Livin nicht in allen Melanomzelllinien
exprimiert wurde. Die allgemeine Überexpression der IAPs in Melanomzellen
könnte durch die fehlende iNOS (induzierbare Stickstoffmonoxid-
Synthase)-Expression begründet sein. Das proapoptotische AIF (apoptosis
inducing factor) wurde hingegen in Melanomzellen schwächer als in Melanozyten
exprimiert. Da sich zwischen sensitiven und resistenten Melanomzelllinien
deutliche Expressionsunterschiede der anti- und proapoptotischen Proteine
fanden, konnten mehrere Hypothesen zur Resistenzentwicklung aufgestellt
werden. So könnte die Überexpression von Survivin für die Apoptoseresistenz
bestimmter Tumoren verantwortlich sein. So fand sich in den untersuchten
Zelllinien eine gegenläufige Expression des antiapoptotischen Survivin und des
proapoptotischen Smac. In resistenten Melanomzelllinien lag eine starke
Survivin- und schwache Smac- Expression vor. Survivin interagiert mit XIAP
(X-linked inhibitor of apoptosis protein) und unterbindet auf diesem Weg die
apoptotische Wirkung von Smac. Es gibt in der vorangegangenen Literatur jedoch
auch Hinweise für eine direkte Interaktion mit Smac. Weiterhin konnten in
dieser Untersuchung der negative Einfluss von Survivin auf die AIF-Expression
bestätigt werden. Survivin antagonisiert das proapoptotische Verhalten von
AIF. Eine Erklärung für die Resistenzentwicklung ließ sich hierdurch jedoch
nicht finden. AIF und Smac zeigten eine gegenläufige Expression, für die in
der bisherigen Literatur durch eine direkte Interaktion keine Erklärung
gefunden werden konnte. Übereinstimmend mit vorangegangenen Untersuchungen
ließ sich auch für Livin und Smac eine gegenläufige Expression finden. In den
Melanomzelllinien mit starker Livin- Expression fand sich nur eine schwache
Smac-Expression. Livin dient bereits als therapeutischer Marker. Eine weitere
Begründung für die Resistenzentwicklung von Melanomzellen könnte in der hohen
cFLIP-Expression gefunden werden. Bei nur schwacher Expression von cFLIP
zeigten sich die untersuchten Melanomzelllinien, bis auf eine Zelllinie,
sensitiv gegenüber den Apoptosestimuli. Zur Untersuchung der Überexpression
von CD95L auf die Apoptoseregulation wurde ein Tetracyclin-abhängiges
Expressionssystem genutzt. Verwendet wurde die Melanomzelllinie SK-Mel-13, die
mit dem Gen für CD95L stabil transfiziert wurde. Die Melanomzellen wurden
dadurch für zusätzliche Apoptosestimuli signifikant sensitiviert, wobei sich
die zweifach transfizierte Zelllinie SKM13-CD95L vor Aktivierung des Promotors
mit Doxycyclin auf die Induktion der Apoptose resistent zeigte. Die
überlebenden Zellen könnten auf Resistenz gegenüber dem selbst produzierten
CD95L selektiert worden sein und somit die Apoptoseresistenz erklären. Es wird
dabei von einer geringen Basalexpression des Transgens ausgegangen
(„leaky“-Expression). Die Resistenzentwicklung scheint dabei unabhängig von
den Expressionsveränderungen der pro- und antiapoptotischen Proteine zu sein.
Die antiapoptotischen Caspaseninhibitoren der Gruppe der IAPs, FLIP sowie die
proapoptotischen Proteine AIF und Smac haben als Regulatoren der Apoptose
entscheidenden Einfluss auf die Resistenzentwicklungen des malignen Melanoms.
Mit Hinblick auf die Entwicklung therapeutischer Strategien wäre es
interessant und wünschenswert die Ansätze dieser Arbeit weiter auszubauen. Nur
durch die Aufklärung der genauen zellulären Abläufe können alte Therapieformen
optimiert und neue Therapiekonzepte entwickelt werden.

dc.description.abstract
Melanoma is a highly malignant tumor which is resistant to most usual
therapies. This resistance is mainly due to a reduction of apoptosis in
melanoma cells. Aim of this study was to analyze specific pathway steps of
apoptosis in normal melanocytes and malignant melanoma cells. We therefore
quantified and compared the efficacy of apoptosis inducing stimuli on
melanocytes and melanoma cells. Furthermore we analyzed changes in expression
of regulators of apoptosis in both cell types. Since the melanoma cells showed
partially resistances to the induction of apoptosis by CH-11, IFN-γ
(Interferon-γ), LPS (lipopolysaccharide) und TNF-α (tumor necrosis factor-α)
we could distinguish resistant and sensitive melanoma cell lines. In contrast
to melanocytes we found in all examined melanoma cells a stronger expression
of IAPs (inhibitors of apoptosis proteins), of cFLIP (cellular FLICE
inhibitory protein) and of Smac (second mitochondria-derived activator of
caspases). However, Livin was not expressed in all melanoma cell lines. The
overall increased expression of IAP in melanoma cells may be explained by a
missing iNOS (inducible nitric oxide synthase)-expression in these cells. On
the other hand, the pro-apoptotic AIF (apoptosis inducing factor) was
expressed much weaker in melanoma cells than in normal melanocytes. As we
found strong distinctions of expression of anti- and proapoptotic proteins
between sensitive and resistant melanoma cells, we can formulate several
hypothesis that may explain how resistance of melanoma cells to induction of
apoptosis may have developed. One explanation could be an overexpression of
the anti-apoptotic protein Survivin. We found a reciprocal expression of
antiapoptotic Survivin and of proapoptotic Smac. Within the resistant melanoma
cell lines we observed a strong expression of Survivin and a weak expression
of Smac. Survivin inhibits the apoptotic impact of Smac by interacting with
XIAP (X-linked inhibitor of apoptosis protein). Furthermore it inhibits AIF-
expression and antagonizes the pro-apoptotic effect of AIF. Yet, this
mechanism does not completely explain the development of resistance of
melanocytes towards apoptosis induction. AIF and Smac showed reciprocal
expression for which up to today there has not been found a proper
explanation. The reciprocal expression of Livin and Smac on the other hand has
been described by others previously. In this study, melanoma cell lines with
strong Livin expression showed only mild Smac expression. Livin is already
being used as a diagnostic marker. Another reason for the development of
resistance to apoptosis may be the high cFLIP expression on melanoma cells.
When showing a weak expression of cFLIP all but one of the melanoma cell lines
we investigated, were found to be sensitive to stimuli of apoptosis. In order
to investigate the effect of CD95L-overexpression on apoptosis regulation we
used a tetracyclin regulated expression system. SK-Mel-13, a melanoma cell
line that is transfected with the gene encoding for CD95L, was used for this
investigation. Through this transfection the melanoma cells became more
sensitive to apoptosis stimuli. The twice transfected cell line SKM13-CD95L in
contrast was – prior to activation the promotor gene of apoptosis with
doxycycline - resistant to apoptosis induction. A possible explanation may be
that the survived cell lines had been selected in advance due to their
resistance to their own CD95L. A feeble basic expression of the transgene is
quite assumable („leaky“-expression). The development of resistance seemed to
develop independently from altered expression of pro- and anti-apoptotic
proteins. The anti-apoptotic IAPs and FLIP as well as the pro-apoptotic
proteins AIF and Smac do have a major influence on regulating apoptosis and on
the development of resistance in malignant melanoma cells. Further studies
will be needed to understand the exact cellular and molecular mechanisms of
apoptosis in order to optimize and develop new strategies to treat malignant
melanoma.
