dc.description.abstract
Aufgabenstellung der vorliegenden Arbeit war die Ermittlung der
dreidimensionalen Struktur des bakteriellen epsilon (e), zeta (z)-Antitoxin /
Toxin-Proteinkomplexes (TA-System) kodiert auf dem Plasmid pSM19035 aus
Streptococcus pyogenes. Das hier vorgestellte Strukturmodell ist die erste
Struktur eines TA-Komplexes. Röntgenbeugungsdaten wurden an Selenomethionin-
derivatisiertem Protein (max. Auflösung.: 3.10 Å) und Wild-Typ-Protein (max.
Auflösung.: 1.95 Å) mit Synchrotronstrahlung gesammelt. Die Lösung des
Phasenproblems erfolgte nach der MAD-Methode (Multiple wavelength Anomalous
Diffraction). Die Gene e und z wurden in Escherichia coli in Methionin- oder
Selenomethionin-haltigen Anzuchtsmedien exprimiert. Zur Reinigung des e,z
Proteinkomplexes wurde ein Protokoll entwickelt und optimiert. Homogenes
Protein wurde kristallisiert, an Kristallen wurden Beugungsdaten gesammelt.
Nach Lösung des Phasenproblems mit MAD wurde ein Strukturmodell erstellt und
verfeinert. Zusätzlich erfolgte auch die Lösung des Phasenproblems (mit der
Methode des Molekularen Ersatzes) und anschließende Verfeinerung von
Kristallstrukturen des Komplexes in zwei weiteren polymorphen (polytypen)
Modifikationen. Die auftretenden Konformationsänderungen wurden als
Packungseffekte identifiziert. Das Antitoxin ( neutralisiert im e2z2-Komplex
die toxische Wirkung von z. Dies erfolgt mit Hilfe von Seitenketten der
N-terminalen Helix a von e, welche durch sterische Hinderung und repulsive
Wechselwirkungen einer ATP-Bindung von z entgegenwirken und das Protein somit
inaktivieren. Die inaktive Form liegt in vivo solange vor, als die das Operon
kodierenden Gene im Zellcytosol vorhanden sind. Strukturvergleiche mit in der
Proteindatenbank (PDB) veröffentlichten Proteinen ergaben, dass das Protein z
mit Phosphotransferasen strukturell verwandt ist, wenngleich nur sehr geringe
Aminosäure-Sequenzidentität (max. 15 %) vorhanden ist. Dennoch wurden
basierend auf diesen strukturellen Erkenntnissen in z katalytisch wichtige
Aminosäuren identifiziert. Ausgehend von diesen Ergebnissen wurden zusätzlich
zur gestellten Aufgabe ortsgerichtete Mutagenese-Studien und Strukturanalysen
an diesen mutierten Proteinen durchgeführt. Alle Mutationen führten zu
inaktivem, nicht-toxischem z Protein. Deshalb wird in dieser Arbeit eine
Phosphotransferase-Aktivität als toxischer Effekt von z vorgeschlagen. Damit
wird zum ersten Mal der Wirkungsmechanismus eines TA-Systems sowohl
molekularbiologisch als auch strukturell untersucht.

dc.description.abstract
The aim of this work was the determination of the three dimensional structure
of the bacterial epsilon (e), zeta (z) Toxin / Antitoxin complex (TA system)
encoded by the plasmid pSM19035 of Streptococcus pyogenes. The described
structural model is the first reported structure of a TA complex. X-ray
diffraction data on crystals of a selenomethionine - derived protein (max.
resolution: 3.10 Å) and the wild-type protein (max. resolution: 1.95 Å) were
collected using synchrotron sources and the phase problem was solved utilising
the Multiple wavelength Anomalous Diffraction (MAD) method. The genes encoding
the antitoxin e and toxin z were transformed into E. coli and the proteins
overproduced in methionine or selenomethionine containing media. A protocol
for the purification of the protein complex was developed and optimised.
Homogeneous protein complex e,z was crystallised and X-ray diffraction data
collected. Following solution of the phase problem with MAD, a structural
model was built and refined. Additionally, the phase problems of two further
polymorphous modifications were solved and the structures refined. Apparent
conformational changes were confirmed as effects of crystal packing. The
short-lived antitoxin ( neutralises the toxic effects of z in the e2z2
complex. This is achieved by side chains of the N-terminal helix a of e, which
blocks the putative ATP binding pocket z by steric hindrance and repulsive
interactions, thereby inactivating the toxin. The inactive form exists in vivo
as long as the gene encoding e is present and expressed in the cell cytosol.
Structural comparisons with published proteins deposited in the Protein
Database (PDB) revealed significant structural homology of z to
phosphotransferases, although the sequence identity is low (max. 15%). Despite
the latter, the structural information obtained permitted determination of the
catalytically important residues in z. Expansion of the originally defined
project involved site-directed mutagenesis studies and structural analysis of
the resultant mutant proteins. All mutations of the putative catalytic
residues produced inactive, non-toxic z protein, supporting the postulation
that the toxic effect of z is caused by a phosphotransferase activity. This is
the first account of utilisation of both molecular biological and protein
structural methods to describe the functional mechanism of inactivation of a
TA system.
