Das kolorektale Karzinom (CRC) stellt immer noch eine der häufigsten Todesursachen von Krebs beim Menschen dar. Um die sporadische Entstehung des CRC im Menschen zu imitieren haben wir neue Mausmodelle generiert, welche durch gerichtete Stammzelltransformation durch das SV40 T-Antigen (Tag) in der putativen Stammzellregion der intestinalen Krypten spontan Tumore entwickeln. Mit Hilfe der Lgr5-Cre-ERT2 x LoxP-Tag-Maus haben wir die Immunantwort gegen spontan entstehende Tumoren des gastrointestinalen Traktes analysiert und einander gegenübergestellt. Diese sind entweder durch Applikation von niedrig dosiertem Tamoxifen oder durch eine Tamoxifen-unabhängige stochastische Tag- Expression entstanden. Weiterhin haben wir die Induktion anti-Tag-spezifischer zytotoxischer T-Lymphozyten und IgG in Lgr5-Cre-ERT2 x LoxP-Tag-Mäusen analysieren. Dabei fanden wir heraus, dass sowohl junge als auch einige wenige ältere Lgr5-Cre-ERT2 x LoxP-Tag transgene Mäuse noch eine signifikante zytotoxische T-Zellantwort gegen das dominante Epitop IV von Tag aufrecht behielten. Des Weiteren zeigten diese Mäuse einen mit zunehmendem Alter ansteigenden anti-Tag spezifischen IgG-Titer. Im Vergleich zu Villin-Cre-ERT2 x LoxP-Tag-Mäusen, welche Tag auch außerhalb der Stammzelle in den Mikrovilli des epithelialen Bürstensaumes im gastrointestinalen Trakt exprimieren (Czéh et al., 2010, Oncogene, 29: 6591), führt die spatio-temporale Tamoxifen- induzierbare Tag-Expression in Lgr5-Cre-ERT2 x LoxP-Tag-Mäusen zu einer längeren Tumorlatenz und somit Überlebenszeit. In dem letzteren Modell entwickeln die Mäuse gastrointestinale Tumoren zum Beispiel im Darm, Magen, Pankreas und in der Leber. Die Tamoxifengabe kann eine signifikante ZTL- Antwort in diesen Mäusen hervorrufen und führt zu der Aktivierung von CD4+ und CD8+ T-Zellen im Darm, die mit Hilfe der Marker CD44 und CD69 nachgewiesen werden konnte. Die beschriebenen Tumormodelle ermöglichen ein besseres Verständnis sowohl der lokalen als auch systemischen Immunantwort während des langen Prozesses der Tumorentstehung und erlauben es uns neue Therapeutika zu testen.
Colorectal cancer (CRC) is still one of the most common causes of death from cancer in humans. In order to mimic the sporadic nature of human colon carcinoma development we generated novel mouse models with spontaneous tumour growth by directing the expression of SV40 T antigen (Tag) into the putative stem cell region of the intestinal crypts. With the help of Lgr5-Cre-ERT2 x LoxP-Tag transgenic mice we analyzed and compared the anti-tumour immune response against colorectal cancer directed towards spontaneously arising tumours either throughout the activation of the Cre-recombinase by low-dose Tamoxifen administration or via a stochastic Tag expression in a Tamoxifen- independent fashion. We furthermore aimed to analyze the induction of CTL tolerance towards Tag in Lgr5-Cre-ERT2 x LoxP-Tag transgenic mice at different ages and to follow their anti-Tag IgG antibody response. We found in the stochastic model that young as well as some of the aged Lgr5-Cre-ERT2 x LoxP- Tag transgenic mice still retained CTL responses against the dominant epitope IV of Tag. Furthermore these mice showed an increasing anti-Tag IgG response with age. When comparing Lgr5-Cre-ERT2 x LoxP-Tag mice to the Villin-Cre-ERT2 x LoxP-Tag transgenic mice which express Tag in the microvilli of the brush border epithelial cells lining the gut (Czéh et al., 2010, Oncogene, 29: 6591) in the sporadic Tamoxifen-induced tumour model we found that Lgr5-Cre-ERT2 x LoxP-Tag mice live significantly longer and start to develop tumours at a later time point. In this model Lgr5-Cre-ERT2 x LoxP-Tag mice develop tumours in the gastrointestinal tract such as stomach cancer, pancreatic cancer, cancer of the intestine and liver cancer. The administration of Tamoxifen is able to induce a significant CTL response in these mice and leads to the activation of T cells in the intestine identified by the markers CD69 and CD44. The described colon tumour models offer the opportunity for a better understanding of local as well as systemic immune responses throughout the long process of tumorigenesis and allow testing of novel therapeutics.
