Sphärische, funktionsfähige Hämoglobin-Mikropartikel (Hb-MP) wurden
erfolgreich mittels einer neuen einfachen, effizienten und kosteneffektiven
drei-Schritt-Methode hergestellt: Copräzipitation von Hb mit CaCO3 zu Hb-
CaCO3-MP, Vernetzung des Hb und Auflösung des CaCO3 zu Hb-MP. Die Hb-
Einkapselungseffizienzen der MP variierten zwischen 8 und 50 % abhängig von
der Ausgangskonzentration der CaCl2 , Na2CO3 und Hb Lösungen. Die Hb-Beladung
kann mindestens ein Drittel der Hb-Konzentration in Erythrozyten erreichen.
Der MetHb-Gehalt war nach der Herstellung stark erhöht, konnte aber mittels
Ascorbinsäure in einen akzeptablen Bereich reduziert werden. Die
unmodifizierten Partikel waren durchschnittlich 3,2 µm groß; ein nicht
modifizierter Partikel enthielt durchschnittlich 2,2 pg Hb (ca. 2 • 107 Hb-
Tetramere). Die Partikel waren aufgrund der GA Vernetzung autofluoreszierend.
Die nicht modifizierten Hb MP hatten ein negatives Zetapotential. Nach der
Herstellung waren durchschnittlich 7 20 % der Partikel in Abhängigkeit von der
bei der Vernetzung eingesetzten GA-Konzentration aggregiert (> 10 µm). Mittels
Ultraschall-Behandlung konnten die Aggregate stark reduziert oder beseitigt
werden. Die Partikel wurden nach der Inkubation mit Vollblut oder
plättchenreichem Plasma in vitro analysiert. Je niedriger die eingesetzte GA-
Konzentration bei der Vernetzung war, desto weniger wurden die Partikel
phagozytiert, und desto schwächer aggregierten die Partikel in
plättchenreichem Plasma. Das Quenchen mit Glycin und die Reduktion mit NaBH4
brachten ebenfalls viele Vorteile. Die Partikel wurden weniger phagozytiert
und aggregierten weniger in plättchenreichem Plasma. Die Hb-MP hatten eine
starke Sauerstoffaffinität (also einen niedrigen p50). Wird das Hb mit 0,01 %
GA unter normalem Luftsauerstoffpartialdruck vernetzt, mit Glycin gequencht
und mit NaBH4 reduziert, so haben die Hb-MP einen p50 von weniger als 2 mmHg.
Der p50 betrug 7,0 mmHg, wenn die Partikel unter sauerstoffarmen Bedingungen
vernetzt wurden. Die PEGylierung der Partikel konnte zwar die Aggregation der
MP nach der Herstellung nicht reduzieren, die immunologischen Eigenschaften
der Partikel wurden jedoch positiv beeinflusst. Die PEGylierten Partikel
aggregierten kaum in plättchenreichem Plasma. Die Partikel hatten ein
reduziertes Zetapotential und eine erhöhte Sauerstoffaffinität mit einem p50
von 5,3 mmHg. Hb-Mischpartikel mit HA, HSA oder modifiziert mit Dx wiesen eine
reduzierte Aggregation der Partikel nach der Herstellung auf. Im Vergleich zu
Hb-MP ohne Modifikation, wurde das Zetapotential der Partikel moderat (bei Hb-
HSA-MP) oder stark (bei Hb HA MP oder Hb Dx MP) erhöht. Eine leicht erhöhte
Phagozytose der Hb HSA MP und eine stark erhöhte Phagozytose bei Hb-HA-MP oder
Hb-Dx-MP waren die Folge. Die modifizierte Copräzipitation mit HSA oder die
Modifikation mit Dx konnten die Aggregation der Partikel in plättchenreichem
Plasma nicht reduzieren. Es wurde jedoch keine Aggregation nach der Inkubation
der Hb-HA-MP mit plättchenreichem Plasma beobachtet. Nach der Einführung von
HA, HSA und der Modifikation mit Dx wurde die Sauerstoffaffinität noch einmal
erhöht; die p50 von diesen Partikeln waren kleiner als 4 mmHg. Aufgrund der
Größe im Mikrometer-Bereich und der hohen Sauerstoffaffinität ist zu vermuten,
dass die Partikel nicht in den interstitiellen Raum gelangen und damit ein
begrenztes NO-Scavenging haben, und dass sie den Sauerstoff nicht vorzeitig in
den präkapillaren Arteriolen freigeben. Somit würden die Partikel keine
Vasokonstriktion auslösen. Der niedrige p50 der Hb-MP würde den Einsatz der
Partikel bei starker Hypoxie oder in der Tumortherapie erlauben.

Spherical functioning hemoglobin microparticles (Hb-MPs) were successfully
produced by a new simple, efficient and cost-effective 3-step method: co-
precipitation of Hb with CaCO3 to form Hb-CaCO3 microparticles, cross-linking
of Hb and dissolution of CaCO3 template to obtain pure Hb-MPs. The Hb
encapsulation efficiencies ranged from 8 to 50 % of the initial amount of Hb
depending on the concentration of CaCl2, Na2CO3 or Hb solution used. The Hb-
loading of the CaCO3-MPs was at least one third of the Hb content of the RBCs.
The methemoglobin content was significantly increased after preparation, but
could be reduced to an acceptable range after a post-reduction of the Hb-MPs
with ascorbic acid at pH 7.4. The unmodified particles had an average diameter
of about 3.2 µm; one unmodified particle contained 2.2 pg Hb (approximately 2
• 107 Hb tetramers). The particles were auto-fluorescent due to the
glutaraldehyde (GA) cross-linking. The unmodified Hb-MPs had a negative zeta
potential. Depending on the GA concentration used for cross-linking, 7 20 % of
the particles were aggregated (> 10 µm). The amount of aggregates could be
reduced greatly or eliminated by sonication. The particles were analyzed after
incubation with whole blood or platelet rich plasma in vitro. The lower the
concentration of GA during cross-linking, the less particles were
phagocytized, and the weaker the particles aggregated in the platelet rich
plasma. Quenching with glycine and the reduction with NaBH4 also brought
benefits. The particles were less phagocytized and less aggregated in platelet
rich plasma. The Hb-MPs had a high oxygen affinity (low p50). In the case
where 0.01 % GA cross-linked Hb-MPs, which were also quenched with glycine and
reduced with sodium borohydride, the p50 was less than 2 mmHg, if the
particles were cross-linked under normal atmospheric oxygen partial pressure.
The p50 was 7.0 mmHg, if the particles were cross-linked under low-oxygen
condition. PEGylation of the particles could not reduce the aggregation after
preparation. However, it positively affected the immunological properties of
the particles. After sonication, the PEGylated particles hardly aggregated in
platelet rich plasma. The particles had a reduced zeta potential and increased
oxygen affinity with a p50 of 5.3 mmHg. The preparation of Hb mixture-
particles with hyaluronan (HA) or human serum albumin (HSA) or modification
with dextran (Dx) reduced the aggregation of the particles after preparation.
Compared to unmodified Hb-MPs, the zeta potential of particles was moderately
(by Hb-HSA-MPs) or strongly (by Hb-HA-MPs and Hb-Dx-MPs) increased. A slightly
increased phagocytosis of particles which were co-precipitated with HSA and
significantly increased phagocytosis of particles which were co-precipitated
with HA or modified with Dx were observed. The preparation of mixture-
particles of Hb and HSA or modification with Dx could not reduce the
aggregation of the particles in platelet rich plasma. After incubation of the
Hb-HA-MPs with platelet rich plasma, no aggregation was observed. After
modification with HA, HSA or Dx, the oxygen affinity was increased again; the
p50s from these particles were smaller than 4 mmHg. Due to their size in the
micron range and their high oxygen affinity, it is to expect that our
particles would not access to vascular space and thus would have a limited
scavenging of the vasodilating agent nitric oxide (NO), and they would not
release O2 prematurely in the precapillary arterioles. Therefore, the
particles would not cause vasoconstriction. The low p50 of the Hb-MPs would
allow their use as targeted oxygen delivery system to hypoxic tissues which
are observed in case of severe hypoxia and tumors.
