Die starke Zunahme an DNA-basierten Nachweismethoden seit Erfindung der PCR
und der Entwicklung der Real-Time PCR setzt sich auch vermehrt in der
Lebensmittelüberwachung durch. Die vorliegende Arbeit umfasst den
molekularbiologischen Nachweis mehrerer pathogener Keime aus Lebensmitteln.
Veröffentlichte Methoden wurden evaluiert und darauf aufbauend optimierte
Schnellverfahren entwickelt. Der Nachweis von Salmonella spp. mittels PCR
wurde für den Einsatz in der amtlichen Überwachung etabliert. Zusätzlich wurde
er als Real-Time PCR weiterentwickelt und umfassend validiert. Diese zeigte
eine hohe Spezifität und Sensitivität. Pro Ansatz konnten weniger als 5 Keime
pro 25 g Lebensmittel nach Anreicherung detektiert werden. Diese Real-Time PCR
wurde mit einer neu entwickelten internen Amplifikationskontrolle als Duplex-
Real-Time PCR etabliert. In weiterführenden Arbeiten wurde sie in einem
Ringversuch validiert, und stellt als Teil der Methodensammlung nach §64 LFGB
eine offizielle Untersuchungsmethode dar. Zur genauen Typisierung von
Salmonella-Serovaren wurde zudem die PFGE getestet. Die Ergebnisse zeigten,
dass sie das passende Werkzeug für eine differenzierte epidemiologische
Untersuchung darstellt. PCR und Real-Time PCR können für die amtliche
Überwachung zum raschen Screening eingesetzt werden, an das sich eine
molekularbiologische Serotypisierung mittels PFGE anschließen kann. Der
Nachweis von thermophilen Campylobactern wurde mittels PCR etabliert und zudem
eine Real-Time PCR neu entwickelt. Diese Methode zeigte eine Exklusivität von
100% und eine Inklusivität von 98,1% und damit statistisch eine fast
vollständige Übereinstimmung. Die Nachweisgrenze für die Real-Time PCR lag bei
ca. 5 DNA-Kopien pro Ansatz. Im Vergleich mit der kulturellen Methode konnten
ebenfalls weniger als 5 Keime pro 25 g Lebensmittel sicher nachgewiesen
werden. Für die beiden Real-Time PCRs wurde zur Vermeidung falsch-negativer
Ergebnisse eine Amplifikationskontrolle neu entwickelt, kloniert und
etabliert. Für beide Methoden wurden mit interner und externer
Inhibitionskontrollen unterschiedliche Strategien verfolgt. Des weiteren wurde
der molekularbiologische Erregernachweis für Listeria monocytogenes etabliert
und ein Microarray zum Nachweis von drei Keimen, Salmonella spp., C.
jejuni/coli sowie L. monocytogenes ausgetestet. Zusammenfassend zeigten die
PCRs und Real-Time PCRs eine hohe Spezifität und Sensitivität. Gerade die
Real-Time PCR bietet für die amtliche Überwachung eine schnelle und
verlässliche Detektion pathogener Keime. Dabei ermöglicht die interne
Amplifikationskontrolle falsch-negative Ergebnisse auszuschließen. Im Rahmen
der Arbeit wurden mehrere solcher Methoden entwickelt, bzw. weiterentwickelt,
validiert und in die Überwachung eingeführt. Eine rasche und zuverlässige
Erkennung von Infektionserregern ermöglicht es, Lebensmittel, von denen eine
gesundheitliche Gefährdung ausgeht, schnellstmöglich aus dem Verkehr zu
ziehen.

dc.description.abstract
Since the invention of the PCR and Real-Time PCR methods, their use in food
control and surveillance has increased tremendously. The present study details
the detection of several pathogenic microorganisms in food by PCR and Real-
time PCR. Published methods were evaluated and modified and new methods
developed. Their sensitivity, specificity and practicability were tested. For
the detection of Salmonella spp. a PCR was established and developed further
as a Real-Time PCR. This method was comprehensively compared to the cultural
method using artificially and naturally contaminated samples. It showed a high
specificity and sensitivity and generally yielded the same results as the
cultural method. This Real-Time PCR was then combined and validated as a
duplex PCR with a newly developed internal amplification control. In further
work, this duplex-Real-Time PCR has been validated in a collaborative study in
Germany and now represents an official diagnostic method according to §64
LFGB. Furthermore PFGE was used for a thorough typing of Salmonella serovars.
It showed itself a precise tool to gain detailed information for
epidemiological conclusions. PCR and Real-Time PCR can be used for fast
screening in governmental food control and can be followed by exact
identification and molecular biological typing with PFGE. Additionally the
detection of Campylobacter jejuni and coli with PCR was established and a new
Real-Time PCR was developed. This method was similarly validated as the above.
The comparison of the Real-Time PCR with the cultural method resulted in an
exclusivity of 100% and an inclusivity of 98,1%, which means almost complete
concordance. The limit of detection for the Real-Time PCR was found to be
about 5 copies DNA per sample. Compared with the cultural method, both methods
were able to reliably detect less than 5 cfu per 25 g food. To prevent false-
negative results, an amplification control was developed, cloned and
established for the Real-Time PCR for Salmonella spp. as well as for
Campylobacter coli/jejuni. For the different Real-Time PCR methods, either an
internal or an external inhibition control were used. Furthermore the
detection of Listeria monocytogenes using nucleic acid based methods was
tested, comparing the PCR and Real-Time PCR with a microarray. All three
methods showed similar results. In summary both the PCRs and the Real-Time PCR
showed a high specificity as well as sensitivity. The Real-Time PCR in
particular offers very fast and reliable detection of bacterial pathogens. In
addition the new internal amplification control reliably excludes false-
negative samples. Within the scope of this work several methods to detect
pathogenic microorganisms in food were developed, refined and implemented in
food surveillance. Fast detection of bacterial pathogens ensures that food
products that present a health risk or hazard to consumers can quickly be
withdrawn from public consumption.
