HINTERGRUND: Die Atherosklerose ist eine inflammatorische Erkrankung des arteriellen Gefäßsystems, die in verschiedenen Stadien über Jahre bis Jahrzehnte fortschreitet und sich in Form von koronaren Infarkten, Apoplex, pAVK oder Mesenterialinfarkt klinisch manifestiert. Sie ist häufigste Ursache für Tod und Behinderung in den westlichen Industriestaaten. Neben einer Reihe gesicherter Risikofaktoren konnte in den letzten Jahren die Bedeutung inflammatorischer Prozesse als Ursache sklerotischer Gefäßveränderungen nachgewiesen werden. Ziel dieser Arbeit ist die Untersuchung, wie HDL und sein assoziiertes Sphingosin-1-Phosphat antiinflammatorische Signalwege im Rahmen der Atherosklerose beeinflussen. MATERIAL UND METHODEN: Es wurde die Expression der im Rahmen der Atherosklerose beteiligten proinflammatorischen Gene iNOS, MMP9 und sPLA2 überprüft. Untersuchungsmaterial waren glatte Gefäßmuskelzellen (VSMCs), die aus der thorakalen Aorta von Wistar-Kyoto- Ratten gewonnen wurden. Nach Inkubation mit pro- und antiinflammatorischen Testsubstanzen (nach etablierten Arbeitsschritten) erfolgten RNA-Isolierung, reverse Transkription und Polymerase-Kettenreaktion (PCR) zur Expressionsbestimmung der untersuchten Gene. ERGEBNISSE: HDL und das assoziierte Sphingosin-1-Phosphat waren in der Lage, die Expression der proinflammatorischen Gene iNOS, sPLA2 und MMP9 signifikant zu hemmen. Die Blockierung des S1P3-Rezeptors durch das Einschleusen von siRNA hob die antiinflammatorischen Effekte des S1P und der HDL auf. Bei Inkubation mit verschiedenen Antagonisten und Agonisten der S1P-Rezeptorsystems zeigte sich, dass die Beeinflussung der S1P-Rezeptoren1 und -2 die antiinflammatorischen Wirkungen von HDL und S1P nicht verändern. Die Antagonisierung des S1P3-Rezeptors hingegen hob die antiinflammatorische Wirkung von HDL und S1P auf, die Stimulierung des S1P3-Rezeptors mit dem selektiven Agonisten FTY 720 vermittelte eine signifikante Hemmung der Expression proinflammatorischer Gene. SCHLUSSFOLGERUNG: Es konnte gezeigt werden, dass HDL und seine Komponente S1P die Induktion der proinflammatorischen Enzyme iNOS, MMP9 und sPLA2 über S1P-Rezeptoren hemmen, wobei der Subtyp S1P3-Rezeptor eine entscheidende Rolle spielt. Somit sind HDL auch in späten Stadien der Atherosklerose skleroprotektiv wirksam. Weitere Untersuchungen müssen nun klären, inwieweit die untersuchten Mechanismen auch in vivo wirksam sind. In ersten Untersuchungen an Mäusen deutet sich jedoch bereits an, dass der S1P3-Rezeptoragonist FTY 720 die Atheroskleroseneigung deutlich reduziert
BACKROUND: Atherosclerosis is known as an inflammatory disease of the arterial vessel system which develops in several stadiums for years. The clinical manifestation is possible as infarction of the heart, of the brain, of the gastrointestinal system or of extremities. Today atherosclerosis is the main reason for death and disability in the western world. There are a lot of well known risk factors but in the last years chronic inflammatory processes of the arterial wall were identified to play a key role in the development of atherosclerosis. With this dissertation we will show how HDL and it´s associated Sphingosine-1-Phosphate influence anti-inflammatory pathways in atherosclerosis. MATERIALS AND METHODS: We examined the expression of pro- inflammatory genes, such as iNOS, MMP9 and sPLA2. We used vascular smooth muscle cells (VSMCs) from the thoracic aorta of Wistar-Kyoto-rats. After incubation with test solutions RNA was isolated, then cDNA was synthesized by reverse Transcription and amplified by Polymerase chain reaction (PCR) to identify the expression of the examined genes. RESULTS: HDL and the associated Sphingosine-1-Phosphate were able to decrease the expression of the pro- inflammatory genes of iNOS, MMP9 and sPLA2 significant. The inhibiting of the S1P3-Receptor by using siRNA blocked the anti-inflammatory effects of HDL and the associated S1P. Using agonists and antagonists of the S1P1- and S1P2-receptors had no effect on the anti-inflammatory effects of HDL and S1P but an antagonized S1P3-receptor inhibited the anti-inflammatory effects. Stimulation by the S1P3-selective Agonist FTY 720 decreased the expression of pro-inflammatory genes significant. CONCLUSIONS: It was possible to prove, that HDL and the associated Sphingosine-1-Phosphate inhibited the induction of pro-inflammatory genes such as iNOS, MMP9 and sPLA2 via S1P3-receptor. That suggests that HDL has a protective role even in later stadiums of atherosclerosis. Further examinations now have to prove these results in vivo. First tests showed a decreased rate of atherosclerosis in FTY 720 – treated mice.
