In der vorliegenden Arbeit wurde die Isolierung der mitochondrialen Cytochrom c Reduktasen aus Kartoffel und Weizen mit Hilfe von Cytochrom c-Affinitätschromatographie und Gelfiltrationschromatographie beschrieben. Die verwendete Präparationsmethode führt zu einem hochgradig aufgereinigten und aktiven Enzymkomplex in monodisperser Form. Die Cytochrom c Reduktase aus Kartoffel wurde physiologisch und strukturell eingehend untersucht. Der Enzymkomplex liegt als Dimer vor und katalysiert nach Rekonstitution in Phospholipidmembranen eine Antimycin- und Myxothiazol-sensitive Reduktion von Cytochrom c mit einem Umsatz von 50 s-1. Daneben konnte eine Ubiquinol- Ubiquinon Transhydrogenase-Aktivität nachgewiesen werden. Die Cytochrom c Reduktase setzt sich aus 10 Untereinheiten zusammen, die sich auf der Basis von immunologischen und strukturellen Daten den Komponenten der entsprechenden Proteinkomplexe aus Pilzen und Säugern zuordnen lassen. Demnach kommen neben den respiratorischen Untereinheiten Cytochrom b, Cytochrom c1 und einem Eisen- Schwefel-Protein noch vier Komponenten mit niedrigem Molekulargewicht vor, die als "Q-binding"-, "Hinge"-, "Core-linked"- und "Cytochrome c1-linked"-Proteine bezeichnet werden, sowie drei Untereinheiten im 50 kd-Bereich, die zu der Gruppe der sogenannten "Core"-Proteine gehören. Für Cytochrom b der Kartoffel konnte erstmals die Abspaltung des Initiator Methionins eines mitochondrial kodierten Proteins gezeigt werden. Damit muß für pflanzliche Mitochondrien das Vorhandensein von Methioninaminopeptidasen angenommen werden. Für Cytochrom c1 der Kartoffel erfolgte eine Isolierung und Sequenzierung von cDNA-Klonen. Die abgeleiteten Aminosäuresequenzen erlauben im Vergleich zu der N-terminalen Sequenz des reifen Proteins die Definition der Leitsequenz. Auch pflanzliche Komponenten des mitochondrialen Intermembranraums zeichnen sich demnach analog zu den Proteinen dieses Subkompartiments aus Pilzen und Säugern durch eine "Targeting"-Sequenz mit einem charakteristischen zweigeteilten Polaritätsprofil aus. In der Cytochrom c Reduktase aus Kartoffel kommt im Gegensatz zu dem Enzymkomplex aus anderen Organismen eine dritte Core- Untereinheit vor. Die drei Core-Proteine sind immunologisch unterscheidbar und stellen integrale Bestandteile des respiratorischen Proteinkomplexes dar. Sie kreuzreagieren mit Antikörpern, die gegen die beiden Komponenten der mitochondrialen Prozessierungsprotease - MPP und PEP - aus Pilzen gerichtet sind. Ausgedehnte Aminosäuresequenzierungen bestätigen einen Bezug dieser Untereinheiten zu MPP und PEP. Auch physiologisch erweisen sich die isolierten Cytochrom c Reduktasen aus Kartoffel und Weizen als spezifische Prozessierungsproteasen. Somit ist der Komplex III der Atmungskette höherer Pflanzen bifunktional und wird als "Cytochrom c Reduktase / Processing Peptidase-Komplex" bezeichnet.
The mitochondrial cytochrome c reductase complexes from potato and wheat were isolated by cytochrome c-affinity and gel filtration chromatography. Enzymes were obtained in monodisperse form at high purity. Cytochrome c reductase from potato was physiologically and structurally characterized. The enzyme complex exists as a dimer. Upon reconstitution into phospholipid membranes it catalyzes Antimycin- and Myxothiazol-sensitive reduction of cytochrome c at a rate of 50 per second. Besides, an ubiquinone-ubiquinol oxidoreductase activity was monitored. Cytochrome c reductase of potato consists of 10 subunits which structurally resemble the subunits of the homologous protein complexes from fungi and mammals. Besides the three respiratory subunits cytochrome b, cytochrome c1 and the Iron-sulfur subunit it includes four small subunits (the "Q-binding"-, "Hinge"-, "Core-linked"- and "Cytochrome c1-linked"-proteins) as well as three large subunits in the 50 kDa range (which all resemble the so-called “Core” subunits). It has been demonstrated for the first time that mitochondrial cytochrome b lacks the initiator methionine. This implies presence of a methionine aminopeptidase within the mitochondria of plants. Copy-DNA clones encoding cytochrome c1 from potato were isolated and sequenced. Comparison of the deduced amino acid sequences and the directly determined N-terminal amino acid sequence of the isolated protein allowed defining the N-terminal pre-sequence for mitochondrial targeting. Similar to the situation in fungi and mammals the pre-sequence of cytochrome c1 from potato also comprises a bipartite profile. In contrast to cytochrome c reductase complexes from other groups of organisms the complex from potato includes a third „Core“ subunit. All three Core subunits are distinguishable by immunoblotting procedures and represent integral parts of the enzyme complex. Furthermore, they immunologically cross-react with antibodies directed against the two subunits of the mitochondrial processing peptidase (termed MPP and PEP). This result was confirmed by extensive direct amino acid sequence determination. Indeed, isolated cytochrome c reductase complexes from potato and wheat exhibit processing peptidase activity. To describe this bifunctionality the enzyme complex is designated the “cytochrome c reductase / processing peptidase complex”.
