Monozyten und Makrophagen gehören zu den zellulären Bestandteilen des
Immunsystems denen vielfältige Aufgaben bei der Eradikation eindringender
Pathogene und der Aufrechter-haltung der Homöostase zukommen. Ziel dieser
Studie war es, den Einfluss von IFNγ und ver-schiedener Toll-Like Rezeptor
(TLR)-Agonisten auf die undifferenzierte und differenzierte Monozytenzelllinie
THP-1 zu untersuchen. Des Weiteren wurde überprüft, ob und inwiefern
pflanzliche Naturstoffe Einfluss auf eine derartig ausgelöste Entzündung im
Kontext der TNFα-Sekretion und der mRNA-Expression spezifischer
Adhäsionsmoleküle haben. Die Ergebnisse dieser Arbeit verdeutlichten, dass die
Stimulation mit IFNγ und TLR-Agonisten alleine sowie in Kombination zu einer
starken Aktivierung spezifischer immunolo-gischer Prozesse führte. In
undifferenzierten THP-1 Zellen äußerte sich dies in einer frühen (3 h)
Steigerung der mRNA-Expression von TNFα und ICAM-1 und einer späten (24 h)
Aktivierung von PECAM-1, MAC-1, L-Selektin, ESL-1, PSGL-1 und CD86, sowie
einer starken interzellulären Adhäsion. Für die gesteigerte Sekretion von TNFα
und die beobachtete Hemmung der Lysozymaktivität bedurfte es jedoch der
Aktivierung der Proteinkinase C durch den Phorbolester PMA. Die oben
beschriebenen Effekte äußerten sich am stärksten bei der Kom-bination von IFNγ
mit den synthetischen TLR-Agonisten LPS und FSL-1, sowie bestimmten
bakteriellen Präparationen von Bacillus subtilis, Bacillus firmus und
Escherichia coli und de-monstrieren die Bedeutung von TLR-Agonisten bei der
Aktivierung des angeborenen Immun-systems. Traditionelle Heilpflanzen erfreuen
sich zunehmender Beliebtheit, doch sind molekularbi-ologische Untersuchungen
hinsichtlich der Wirkung einzelner Bestandteile selten. β-glykosidisch
gespaltenes Aucubin und Catalpol zeigten einen stimulatorischen Einfluss auf
differenzierte, IFNγ/LPS-unbehandelte und -behandelte THP-1 Zellen. Die
Iridoidglykoside Harpagosid und Harpagid sowie das Diterpen Aconitin führten
hingegen zu einer Reduktion der TNFα-Sekretion auf zirka 50 %. Im Gegensatz
dazu stimulierten sie die mRNA-Expression von TNFα, ICAM-1, L-Selektin, ESL-1,
PSGL-1 und CD86 deutlich und wirkten einem zeitlich bedingten Abfall der
Expression von TNFα und ICAM-1 entgegen, wodurch die Rolle pflanzli-cher
Naturstoffe als Immunmodulatoren, die immunkompetente Zellen zum Ort der
Entzün-dung dirigieren, dort jedoch anti-inflammatorische Prozesse induzieren,
verdeutlicht wurde.

Monocytes and Macrophages are part of the cellular immune system and play a
vital role in the eradication of foreign particles and the maintenance of
homeostasis. Aim of this study was the investigation of the influence of IFNγ
and different Toll-like receptor (TLR)-agonist on the undifferentiated and
differentiated monocytic cell line THP-1. Furthermore it was addressed if and
to what extend various natural products of plant origin had an effect on an
induced infection in the context of TNFα-secretion and mRNA-expression of
certain adhesion molecules. The results of this work showed that the
stimulation with IFNγ and TLR-agonists alone and in combination led to a
strong activation of specific immunologic processes. In undifferentiated THP-1
cells this stimulation resulted in an early (3 h) increase of TNFα and ICAM-1
mRNA-expression and a late (24 h) activation of PECAM-1, MAC-1, L-Selectin,
ESL-1, PSGL-1 and CD86, as well as a strong intercellular adhesion. Yet, for
the increase of TNFα-secretion and the observed inhibition of lysozyme
activity the stimulation of the protein kinase C with the phorbol ester PMA
was required. The above mentioned effects showed to be the strongest in the
combination of IFNγ with either the synthetic TLR-agonists LPS and FSL-1 as
well as certain bacterial preparations of Bacillus subtilis, Bacillus firmus
and Escherichia coli which demonstrates the importance of TLR-agonists on the
activation of the innate immune system. Traditional medicinal plants become
more and more popular yet molecular insights into underlying mechanisms are
scarce. β-glycosidically cleaved Aucubin and Catalpol exhibited a stimulatory
impact on differentiated, IFNγ/LPS untreated and treated THP-1 cells. The
iridoid glycosides Harpagoside and Harpagide as well as the diterpene Aconitin
led to a reduction of TNFα-secretion down to 50 %. On the other hand they
stimulated mRNA-expression of TNFα, ICAM-1, L-Selectin, ESL-1, PSGL-1 and CD86
and diminished the time related decrease of TNFα and ICAM-1 expression which
demonstrates the importance of natural products as immune modulators that not
only guide immune competent cells to sites of infection but also impair local
pro-inflammatory stimuli.
