Die Infektion von Zellen mit einem Virus führt zu einer
Effektor-T-Zellantwort, welche die Viren meist rasch und effektiv eliminiert.
Diese T-Zellantwort unterliegt der Kontrolle durch regulatorische T-Zellen,
die in einer Vielzahl von Szenarien wie chronischen Infektionen,
Autoimmunerkrankungen, Immunreaktionen auf Tumoren und Transplantate eine
Rolle spielen und die in den vergangenen Jahren besonderes Interesse auf sich
zogen. Ziel dieser Arbeit war es zu klären, ob es im Rahmen der Immunantwort
gegen das Influenza-Virus, gegen CMV und EBV neben der Aktivierung virus-
spezifischer Effektor-T-Zellen auch zur Induktion virus-spezifischer IL-10
produzierender regulatorischer CD8+T-Zellen kommt. T-Zellen aus dem peripheren
Blut gesunder HLA-A2.1 positiver Spender wurde zunächst ex vivo mittels
durchflusszytometrischer intrazellulärer Zytokinfärbung auf virus-spezifische
T-Zellantworten untersucht. Zusätzlich wurde getestet, ob über 3, 7 und 10
Tage eine Expansion und damit einhergehende Sensitivierung des
Nachweisverfahrens möglich ist. Ex vivo ließen sich virus-spezifische IL-10
produzierende CD8+ T-Zellen nach Stimulation auf alle drei Viruspeptide
nachweisen. 5 von 11 Spendern reagierten positiv auf das Influenza-Virus und
je 3 auf das CMV und das EBV, wobei die Frequenzen virus-spezifischer T-Zellen
zwischen 0,04 % und 0,09 % (Flu), 0,06 % und 0,09 % und 0,03 % und 0,07 %
(EBV) der CD3+ CD8+ T-Zellen lagen. Nach Expansion der Zellen stieg die
Frequenz IL-10 produzierender T-Zellen auf Maximalwerte von 1,91 % an Tag 7
an. Auch virus-spezifische Effektor-T-Zellen ließen sich über ihre Produktion
von IFNγ reproduzierbar nachweisen. Ihre Zahlen lagen im Mittel stets höher
als die der IL-10 produzierenden Zellen und stiegen auch noch an Tag 10 auf
Werte von bis zu 34,45 % der CD3+ CD8+ T-Zellen. Es fand sich keine
Korrelation der Frequenzen virus-spezifischer IL-10 und IFNγ produzierender
T-Zellen. Die Reaktionen auf die drei Viren, das Influenza-Virus als
Repräsentant einer akuten Virus Infektion und CMV und EBV als Stellvertreter
für chronisch verlaufende Infektionen, unterschieden sich entgegen den
Vermutungen nicht wesentlich. Insgesamt zeigen die Ergebnisse dieser Arbeit,
dass sich im peripheren Blut gesunder Spender virus-spezifische CD8+ T-Zellen
nachweisen lassen. Dabei kann durch die Bestimmung des Zytokinprofils
reaktiver Zellen zwischen Effektorzellen und vermutlich regulatorischen IL-10
produzierenden Zellen unterschieden werden. Auch eine Expansion beider
Populationen für einen sensitiveren Nachweis ist gelungen.

dc.description.abstract
Viral infection causes the activation of effector T cells, leading to a quick
and effective elimination of the virus. This reaction is controlled by
regulatory T cells that have been shown to be of critical importance in
chronic infection, immune mediated disease, the immunological environment of
malignant tumors and transplantation. In this study, we aimed for the
detection of virus-specific IL-10 producing CD8+ T cells activated by peptides
of the influenza virus (Flu), the cytomegalovirus (CMV) and the Epstein-Barr
virus (EBV). We analysed T cells from peripheral blood of healthy HLA-A2.1
positive donors by intracellular cytokine staining and flow cytometry after
stimulation with the viral peptides. Additionally, we expanded these cells
over 3, 7 and 10 days. We succeeded to detect IL-10 producing CD8+ T cells ex
vivo specific for each virus. In 5 out of 11 donors, we found IL-10 producing
cells as an reaction to the influenza-peptide and 3 donors reacted to each CMV
and EBV. The frequencies of virus-specific cells ranged from 0.04 % to 0.09 %
(Flu), 0.06 % to 0.09 % (CMV) and 0.03 % to 0.07 % (EBV) of all CD3+ CD8+ T
cells. After expansion, the frequencies of IL-10 producing T cells increased
to a maximum of 1.91 % on day 7. Concomitantly, we found a reliable effector T
cell reaction as measured by the production of IFNγ. Their population reached
higher numbers compared to the IL-10 positive cells and increased until day 10
to 34.45 % of the total CD3+ CD8+ T cell population. We found no significant
correlation between the frequencies of virus-specific IL-10 producing and IFNγ
producing T cells. There was also no detectable difference between the
reaction to the influenza peptide, representing an acute infection, and the
reaction to CMV and EBV, representing infections with a chronic or persistent
course of disease. Overall, the results show that virus-specific CD8+ T cells
do exist in peripheral blood of healthy donors. They can successfully be
activated and expanded in vitro and the analysis of their cytokine profile
allows differentiating between effector T cells and presumable regulatory
IL-10 producing cells.
