Chlamydien zählen zu den am weitest verbreiteten pathogenen Bakterien. Mit den über Aerosole verbreiteten C. pneumoniae kommen bis zu 80% aller Menschen im Laufe ihres Lebens ein- oder mehrmals in Kontakt. Der Lebenszyklus der Chlamydien durchläuft zwei Phasen: Aus den infektiösen, Sporen-ähnlichen Elementarkörperchen entwickeln sich intrazellulär die metabolisch aktiven Retikularkörperchen, die sich teilen und schließlich wieder zu Elementarkörperchen differenzieren. Infektionen mit C. pneumoniae verlaufen meist asymptomatisch. Sie werden jedoch auch mit einer Vielzahl akuter und bei fortgesetzter Dauer vor allem chronischer Krankheiten assoziiert. Krankheiten wie Asthma, rheumatoide Arthritis, Artheriosklerose oder Altzheimer'sche Krankheit entwickeln sich über einen längeren Zeitraum und werden in Verbindung mit dem persistenten Entwicklungsstadium gebracht, in welchem die Bakterien jahrelang verharren können. In der vorliegenden Arbeit wurde der Beginn einer durch Eisenmangel ausgelösten persistenten Entwicklung auf Proteinebene untersucht. Dazu wurde die metabolische Markierung der Proteine der obligat intrazellulären Bakterien mit 35S etabliert und ein 2D-Referenzgel von C. pneumoniae erstellt. Während in den ersten 24 h nach Infektion unter Eisenmangel nur ein Proteinspot signifikant hochreguliert war, wurden im Zeitraum von 24-48 h 11 hoch- und 8 herunterregulierte Proteinspezies gefunden. Die Identifizierung des Chromosomen-Partitionierungsproteins (ParB) als ein in der Persistenz herunterreguliertes Protein steht in Einklang mit bekanntem Beobachtungen, nach denen die Zellteilung persistenter Chlamydien gestört ist. Mit den differentiell regulierten Enzymen Thioredoxin-Reduktase (TrxB) und der Adenosylmethionin-8-Amino-7-Oxononanoat Aminotransferase (BioA) wurden Hinweise auf metabolische Besonderheiten gefunden, die wie die hypothetischen Proteine mit unbekannter Funktion Cpn0011 und Cpn0623 Ziele weitergehender Arbeiten sein können. Proteine am Ende des aktiven Lebenszyklus wiesen auf Stoffwechsel- und Biosyntheseaktivität hin (Glukose-6-phosphat Dehydrogenase, Glycerinaldehyde-3-phosphat Dehydrogenase, Glukosamin-6-phosphat Isomerase/Deaminase, Thiol-spezifische Antioxidanz Peroxidase, EF-Ts und Hsp70), auf die Umorganisation der Bakterienmembran beim Übergang der Retikularkörperchen in die infektiösen und osmotisch stabilen Elementarkörperchen (Acyl-carrier Protein Synthase, Penizillin Toleranz Protein LytB und 10 kDa Chaperonin), sowie auf Synthese und Zusammensetzung des Typ III-Sekretionsapparates in den späten Retikularkörperchen (Yop Translokations-Protein S). Die beiden polymorphen Membranproteine PmpD und PmpG/I waren in den Retikularkörperchen in Fragmenten vorhanden. Eine Spaltung steht im Einklang mit bioinformatischen Analysen, die einen Typ V-Sekretionsmechanismus für die Proteine der Pmp-Familie vorhersagen. Prozessierung, Lokalisierung und Funktion des potentiellen Autotransporters PmpD wurden daher näher untersucht. Mit Antikörpern gegen den N-terminalen Teil von PmpD konnten durch Immunblots infizierter HEp-2 Zellen sowie isolierter Elementarkörperchen die sukzessive Prozessierung und das Verbleiben des abgespaltenen N-Terminus über eine feste Interaktion mit der äußeren Membran aufgeklärt werden. Immunfluoreszenz, -elektronenmikroskopie und limitierender Trypsinverdau verifizierten die Lokalisierung des N-Terminus an der Oberfläche sowohl der intrazellulären Retikular- als auch der extrazellulären Elementarkörperchen. Die Antikörper neutralisierten die Infektivität von C. pneumoniae in vitro, ein Indiz für eine Funktion von PmpD bei Adhäsion und/oder Invasion eukaryontischer Zellen. Postuliert worden war diese Rolle durch die für die Pmp charakteristischen Aminosäuremotive GGAI und FxxN, die auch Adhäsinen anderer Organismen zu eigen sind. Darüberhinaus stimulierte der rekombinante, N-terminale Teil von PmpD die metabolische Aktivität menschlicher Monozyten sowie deren IL-8 Sekretion konzentrationsabhängig in vitro. Dabei hatten Komplexe des rekombinanten Proteins mit dem Antikörper, hitzedenaturiertes oder mit Trypsin verdautes Protein den stärksten Effekt. PmpD könnte somit ein Pathogenitätsfaktor sein, der wie andere Moduline unabhängig von Endotoxinen zur Inflammation und den nachfolgenden Gewebeläsionen bei Infektionen mit C. pneumoniae beiträgt.
Chlamydiae are among the most widely distributed pathogenic bacteria. About 80% of the population gets infected at least once during their lifetime with the airborne species C. pneumoniae. The chlamydial life cycle can be divided into two phases: Infectious, sporelike elementary bodies differentiate inside the cell into the metabolically active reticular bodies. The reticular bodies divide by binary fission and develope back into elementary bodies at the end of the life cycle. Most of the primary infections with C. pneumoniae are subclinical or asymptomatic but in the long term associated with a variety of acute and chronic diseases. Examples of these chronic diseases are asthma, rheumatoic arthritis, atherosclerosis and altzheimer disease. Bacteria that cause chronic diseases are considered to be in a persistent state that can last for years. Here, the initial events leading into the persistent state induced by iron deficiency was analyzed on protein level. The metabolic labelling of the obligate intracellular bacteria using 35S was established and a reference gel from the C. pneumoniae proteome was established. During the first 24 h post infection only one protein spot was significantly upregulated under iron deficiency. From 24-48 h, 11 protein species were found to be up- and 8 were found to be downregulated. Among the downregulated proteins the Chromosom Partitioning protein (ParB) could be identified, which is in agreement with the observation of an altered or interrupted cell division in persistent Chlamydiae. The two differentially regulated enzymes Thioredoxine Reductase (TrxB) and Adenosylmethionine-8-Amino-7-Oxononanoate Aminotransferase (BioA) point into the direction of metabolic alterations that can be the focus of further research, together with targets like the identified, hypothetical proteins Cpn0011 and Cpn0623 with unknown functions. Proteins present at the end of the active life cycle are responsible for metabolic and biosynthetic processes (Glucose-6-phosphate Dehydrogenase, Glyceraldehyde-3-phosphate Dehydrogenase, Glucoamine-6-phosphate Isomerase/Desaminase, Thiol-specific Antioxidant Peroxidase, EF-Ts and Hsp70), for reorganisation of the bacterial membrane from reticular bodies into the infectious and osmotically stabile elemetary bodies (Acyl-carrier protein Synthase, Penicillin Tolerance Protein LytB and 10 kDa Chaperonine), as well as for synthesis and assembly of the type III-secretion apparatus in the late reticular bodies (Yop Translocation protein S). The two polymorphic membrane proteins PmpD and PmpG/I were found to be present as fragments in the reticular bodies. Cleavage of these proteins was in agreement to bioinformatic analyses that predicted a type V-secretion mechanism for proteins of the Pmp- family. Processing, localization and function of the putative autotransporter PmpD was therefore studied in more detail. The successive processing steps and the fate of the cleaved N-terminus attached to the outer membrane via a strong interaction could be demonstrated with immunoblots of infected HEp-2 cells and isolated elementary bodies using antibodies raised against the N-terminal part of PmpD. Localization of the N-terminus on the surface of both, the intracellular reticular bodies and the extracellular elementary bodies could further be shown by immunofluorescence- and immunoelectron microscopy. The antobodies neutralized infectivity of C. pneumoniae in vitro indicating a function of PmpD in adhesion and/or invasion of eukaryotic cells. This functional role for Pmp proteins was postulated before due to their characteristic GGAI and FxxN tetraaminoacid motifs reminiscent of adhesins in other organisms. Moreover, the recombinant N-terminal part of PmpD stimulated mebabolic activity of human monocytes and IL-8 release in a concentration- dependent manner in vitro. Interestingly, complexes of the recombinant protein with antibodies, heat-denatured protein or tryptic peptides had the strongest activity. Therefore, PmpD can be considered to be a pathogenicity factor possibly contributing to inflammation and tissue damaging in the course of natural infections with C. pneumoniae independently of endotoxins in analogy to other members of the so called modulines.
