In der hier vorliegenden Arbeit wurden mit Hilfe von Stimulationsparadigmen, welche Langzeitpotenzierung induzieren, sharp wave ripple Komplexe (SPW-R) induziert, denen die sukzessive Entladung hippokampaler Neurone zugrunde liegt. Diese SPW-R Komplexe ähneln in ihren Eigenschaften denen spontan auftretender SPW-R: sie entstehen primär in der Subregion CA3 und propagieren über das CA1 zum Subiculum. Dabei hängt ihre Aufrechterhaltung essentiell von AMPA/Kainat Rezeptoren ab, die Induktion hingegen steht in direktem Zusammenhang mit der Aktivierung der NMDA Rezeptoren. Einmal induziert führt eine Blockade der NMDA Rezeptoren nicht mehr zur Unterbrechung der SPW-R; der exzitatorische Anteil über AMPA/Kainat Rezeptoren reicht zur Aufrechterhaltung aus. Im Gegenzug zur hochfrequenten Stimulation kann eine niederfrequente Reizserie, welche Langzeitdepression induziert, zum Zusammenbruch der SPW-R Komplexe führen. Auf intrazellulärer Ebene zeichnet sich während der SPW-R Komplexe eine dynamische Reorganisation neuronaler Netzwerke ab, die gleichsam Zellen in ein Netzwerk rekrutieren als auch aus dem Zellverband ausschließen kann. Unter dem Einfluß des GABA A Rezeptor Antagonisten Bicucullin werden bereits vorher induzierte SPW-R Komplexe in prolongierte Entladungen mit deutlich höherer Amplitude transformiert. Der Frequenzinhalt der ripples entspricht hier etwa 300 Hz. Als Ausdruck fehlender Inhibition zeigen intrazelluläre Ableitungen von Pyramidenzellen eine wesentlich stärkere und verlängerte Depolarisation mit bis zu 40 Aktionspotentialen während eines SPW-R Komplexes. Insgesamt ähneln diese Eigenschaften denen recurrenter epileptiformer Entladungen (RED), wie sie schon im Menschen oder der Ratte nachgewiesen wurden. In weiteren Experimenten wurde der Einfluß von Serotonin, Dopamin und Norepinephrin auf sowohl tetanisch als auch durch Carbachol oder Kainat induzierte Gammaoszillation untersucht. Die pharmakologisch induzierten Gammaoszillationen wurden durch alle getesteten Monoamine dosisabhängig supprimiert und ihr Frequenzinhalt leicht erhöht. Dieser Effekt wird durch Isoproterenol bei tetanisch und pharmakologisch induzierten Gammaoszillation gleichermaßen simuliert und weist mechanistisch auf einen Zusammenhang mit intrazellulärem zyklischem Adenosinmonophosphat hin. Bei tetanischer Induktion hingegen werden die Oszillationen unter dem Einfluss der Monoamine augmentiert. Diese Ergebnisse deuten eine Beteiligung anderer Zelltypen bei tetanisch induzierter Gammaoszillation als bei pharmakologischer Induktion an.
In this present work stimulation paradigms, that are known to induce long term potentiation, were used to induce sharp wave ripple complexes (SPW-R), which are based on successive discharge of hippocampal neurons. Those SPW-R complexes are similar in their characteristics to spontaneously occurring SPW-R: they primarily originate in the subregion CA3 and propagate via CA1 to the subiculum. The induction is essentially dependant on the activation of NMDA receptors, while AMPA/kainate receptors are crucial for their maintenance. Once induced the blockage of NMDA receptors no longer lead to an interruption of SPW-R; the excitation through AMPA/kainate receptors is sufficient for their maintenance. In contrast to high-frequency stimulation a series of low-frequency stimulation leads to long-term depression and to the collapse of SPW-R complexes. On an intracellular level the SPW-R complexes are characterized by a dynamic reorganization of neuronal networks, which can recruit or exclude cells into the network. Under the influence of the GABA A receptor antagonist bicuculline previously induced SPW-R complexes are transformed into a prolonged discharge with considerably higher amplitude. The frequency content of the ripples corresponds to about 300 Hz. During a SPW-R complex intracellular recordings of pyramidal cells show a substantially stronger and prolonged depolarisation with up to 40 spikes as an expression of lack of inhibition. Overall those characteristics resemble of recurrent epiletiform discharges (RED), which have already been shown in humans and rats. In further experiments the influence of serotonin, dopamine and norepinephrine on tetanically as well as on carbachol or kainate induced gamma oscillation was examined. The pharmacological induced gamma oscillation was suppressed dose dependently in all tested monoamines and the frequency content was shifted to slightly higher frequencies. This effect is simulated by isoprotenerol in tetanically and pharmacologically induced gamma oscillation likewise, a finding that hints to a mechanistic correlation with intracellular cyclic adenosine monophosphate. However tetantically induced oscillations are augmented under the influence of monoamines. These results indicate the participation of other types of cells during tetanically induced gamma oscillation compared to pharmacological induction.
