Ziel der Arbeit ist anhand des C57BL/6 Mausmodels für die Haarforschung ein
Modell für das Studium der Pathologie und pharmakologische Manipulation der
ZIA zu etablieren In der Studie wird die Wir- kung einer Einzeldosis von 150
mg/kg CYP(Endo- xan) auf den Haarwuchs und die Wiederherstelung der HF-
Morphologie studiert. Die CYP-bedingte ZIA wird makroskopisch anhand des
Haarschaftverlusts und der Änderung des Hautkolorits der Rückenhaut
untersucht. Weitere Aspekte werden histomorphomet-risch und
elektronenmikroskopisch analysiert. Die Daten zeigen,dass CYP-behandelten
Tiere sowohl makroskopisch als auch histologisch die gleichen Phänomene
aufweisen welche nach CYP-Behandlung bei Menschen vorkommen wie das anfangs
diffuse Effluvium und die ungleichmäßige Alopezie mit rapidem Eintritt und
Progression. Die Daten unter-stützen die Existenz von zwei Grundreaktionen von
Anagen VI-HF auf zytostatische Noxen: dystrophen Anagen VI- und den dystrophen
Katagen. Es gibt regionale Differenzen der HF-Reaktion auf CYP. Die HF-
Architektur wird vor dem Eintritt in die Katagenphase mittels Primärer
Erholung repariert. Nach folgender Transformation ins Telogen wird das
Haarkleid durch Initiation der Sekundären Erholung und de novo Generation von
HF wiederher- gestellt. Die Primären und Sekundären Erholungs- muster der
Morphologie, Repigmentation und der Wiederherstellung des Haarkleides
entsprechen den Mustern nach einer ZIA bei Menschen. Die ZIA in dem
vorgestellten Mausmodell konnte durch eine dreimalige Applikation des IPL CsA
in einer Dosis von 250 mg/kg abgeschwächt werden. Die Daten zeigen, dass die
systemische Gabe von CsA eine protektive Wirkung auf die HF hat. Der Eintritt
der CYP-bedingten ZIA konnte verzögert, jedoch nicht verhindert werden. Die
CsA Gabe favorisiert den Eintritt in das dystrophe Anagen VI-Stadium und
fördert die schnelle Primäre Erholung der geschädigten HF. Im Gegensatz dazu
bedingt 150 mg/kg CYP einen massiveren Übertritt ins dystro- phe Katagen,
sodass die Reparatur des Haarkleides infolge Sekundärer Erholung zustande
kommt.

We introduce cyclophosphamid-induced alopecia (CYP-IA) in C 57 BL-6 mice as a
clinically relevant model for studying the biology of chemotherapy-induced
alopecia and for developing anti-alopecia drugs. One i.p. injection of 150
mg/kg CYP on day 9 p.d. to mice with all back skin follicles in anagen VI
induces severe alopecia that strikingly reproduces the follicle response,
recovery, and histopathology seen in human CYP-IA. CYP dose-dependently
induces abnormal follicular melanogenesis and dystrophic anagen or, in more
severely, damaged follicles, dystrophic catagen. Both dystrophy forms are
followed by an extremely shortened telogen phase, but differ in the associated
hair loss and in recovery patterns, which determines hair regrowth. This
follicular response to CYP can be manipulated pharmacologically: 250 mg/kg
systemic cyclosporine A on day 7,9 and 11 p.d shifts it toward a mild form of
dystrophic anagen, thus retarding CYP-IA and prolonging ''primary recovery''
fallowed by the regrowth of normally pigmented hair (secondary recovery).
