Die Endozytierung synaptischer Vesikel-Proteine von der Plasmamembran zurück in synaptische Vesikel ist ein streng regulierter Prozess. Bisher wurde davon ausgegangen, dass sich der Sortierungsprozess für alle synaptischen Vesikel- Proteine ähnelt. Allerdings existieren Hinweise, die auf eine spezielle Sortierung von vesikulären Glutamattransportern (VGluts) hindeuten. Diese Sortierung ist jedoch weitestgehend unverstanden. Es scheint, dass VGlut1 und 2 nicht nur während der Endozytose an die Plasmamembran sortieren, sondern (tageszeitabhängig) gezielt dorthin transportiert werden. Fraglich ist, ob sie an der Plasmamembran funktionell (z.B. als Natrium-Phosphat-Transporter) ebenfalls relevant sind und ob ein Funktionswechsel eine Rolle in der neuronalen Transmission spielt. In dieser Arbeit wurde der Sortierungsprozess von VGlut1 und 2 in verschiedenen zellulären und subzellulären Systemen untersucht und die Bedeutung von Interaktionspartnern für die Regulation von VGlut1 und 2 betrachtet. Hierfür wurden unter anderem GST-Bindungsstudien mit Mäusegehirnfraktionen, Neurotransmitter-aufnahmeversuche und konfokale Mikroskopie eingesetzt. Die Untersuchung der Translokation von VGlut zwischen Plasma- und synaptischer Vesikelmembran wurde über zwei Wege bearbeitet. Erstens zeigten GST-Bindungsstudien mit GST-Fusionsprotein, bestehend aus GST und der SH3-Domäne von Endophilin bzw. dem C-Terminus von VGlut1 und 2, dass nur VGlut1 an Endophilin bindet, nicht jedoch VGlut2. VGlut2 hingegen bindet direkt an den Adapterproteinkomplex AP2, was auf individuelle Sortierungsprozesse in den jeweiligen VGlut-Neuronen hinweist. Neueste Studien zeigten eine Kolokalisation von VGlut1 und 2 in ein und demselben Neuron. Neurone, die sowohl VGlut1 als auch VGlut2 tragen, könnten über die individuelle Sortierung nicht nur unterschiedliche VGlut1 und 2-Vesikelpools generieren. Ebenfalls ist denkbar, dass eine spezifische Sortierung eines der Transporter z.B. an die Plasmamembran möglich ist, während der andere Transporter z.B. auf dem Vesikel „gelagert“ wird. VGlut1 (und VGlut2) zeigt ein zirkadianes Sortierungsverhalten und lokalisiert in Abhängigkeit der Tageszeit zwischen Plasma- und Vesikelmembran. Um zu prüfen, ob der zirkadianen Oszillation der vesikulären VGlut1-Mengen eine ebenfalls oszillierende Interaktion von VGlut1 mit Endophilin zugrunde liegt, fanden zweitens in dieser Arbeit Untersuchungen an Mäusen statt, die in einem 12h Licht/12h Dunkel- oder 12h Dunkel/12h Dunkelzyklus gehalten wurden. Mit Hilfe von GST-Fusionsprotein, bestehend aus GST und der SH3-Domäne von Endophilin, konnte in WT-Mäusen eine oszillierende Bindung von VGlut1/Dynamin in Abhängigkeit der Tageszeit an Endophilin festgestellt werden. Die Deletion des Uhrengens Period 2 in Per2BRDM1-Mäusen veränderte die Rhythmizität der Mäuse, was sich in einer gleichbleibenden Bindefähigkeit von VGlut1 (bezogen auf Dynamin) an GST-SH3-Endophilin zeigte. Damit wird deutlich, dass VGlut1 anders als andere synaptische Vesikel-Proteine individuell sortiert wird und diese Sortierung nicht nur im Zuge der Endo- bzw. Exozytose eine Rolle spielt. Als Modell für die VGlut2-Translokation wurde eine VGlut2-exprimierende Phäochromozytomzelllinie der Ratte (PC12) charakterisiert. In diesen Zellen kommt der Transporter vornehmlich an der Plasmamembran vor. Nur VGlut2- und nicht pcDNA3-transfizierte PC12-Zellen zeigen in Gegenwart von Phosphat eine unmittelbare Natriumaufnahme über die Plasmamembran. VGlut2 wirkt in diesen Zellen also eher als Natrium-Phosphat-Co-transporter und weniger als vesikulärer Glutamattransporter, was auf eine funktionelle Relevanz in Abhängigkeit der Sortierung deutet. Zusammenfassend kann festgestellt werden: 1) Während VGlut1 mit Endophilin interagiert, bindet VGlut2 direkt den Adapterproteinkomplex AP2. 2) Die Bindung von VGlut1 an Endophilin unterliegt einer tageszeitabhängigen Regulierung. 3) VGlut2 besitzt in transfizierten PC12-Zellen die Funktion eines Natrium-Phosphat-Transporters.
The Endocytosis of synaptic vesicle proteins from the plasma membrane back into synaptic vesicles is a highly regulated process. Until now research assumed the trafficking process of synaptic vesicle proteins to be similar. Data exists, however, indicating that the trafficking of vesicular glutamate transporters (VGluts) differs even though it is still poorly understood. It seems that VGlut1 and 2 sort to the plasma membrane not only during exocytosis, but are also transported there in a specific day time dependent manner. Questions remain, such as whether VGlut1 and 2 are functionally relevant at the plasma membrane, e.g. as a sodium phosphate transporter, and whether this change in function is important for synaptic transmission. In this thesis, the trafficking of VGlut1 and 2 in different cellular and subcellular compartments was investigated as well as the impact of interaction partners for the regulation of VGlut1 and 2 using GST pulldown assays with mouse brain extract, neurotransmitter uptake assays and confocal microscopy. This study approaches the translocation of VGlut to the plasma membrane in two different ways. Firstly, GST pulldown assays using GST fusion proteins with GST and the SH3 domain of Endophilin or, accordingly, the C-Terminus of VGlut1 and 2, showed an interaction of Endophilin and VGlut1 but not for VGlut2. VGlut2 interacts directly with the adapter protein complex AP2 indicating individual trafficking routes for VGluts. The latest data reveals the coexistence of VGlut1 and 2 in the same neuron. Neurons carrying VGlut1 and 2 might not only generate different vesicle pools; it is further possible that one transporter is sorted to the plasma membrane leaving the other on the synaptic vesicle. VGlut1 (and VGlut2) harbours a circadian trafficking pattern and cycles in a day time dependent manner between plasma and vesicle membrane. Secondly, in order to investigate whether an oscillating binding of VGlut1 to Endophilin forms the basis of the circadian oscillation in the amount of vesicular VGlut1, brain extracts of mice entrained in a 12h light/12h dark- cycle or 12h dark/12h dark-cycle were analysed by GST pulldown assays. The analysis of wild type mice using GST fusion protein containing GST and the SH3 domain of Endophilin, showed an oscillating binding pattern of VGlut1 (in ratio to Dynamin) to Endophilin – depending on the time of the day. The deletion of the period gene Period 2 in Per2BRDM1 mice resulted in the loss of this oscillating interaction between VGlut1/Dynamin and Endophilin. This effect emphasizes the individual trafficking process of VGlut1 in comparison to other synaptic vesicle proteins, and its importance not only during endo- but also exocytosis. In order to study the trafficking of VGlut2, a Rat adrenal pheochromocytoma cell line (PC12) carrying VGlut2 was characterised. In these cells, VGlut2 presumably translocates to the plasma membrane. Only VGlut2- and not pcDNA3 transfected PC12 cells exhibit an immediate sodium uptake across the plasma membrane in the presence of phosphate. VGlut2 functions in these cells as a sodium-phosphate-transporter and not as a vesicular glutamate transporter, indicating a functional relevance subject to the trafficking of the transporter. In summary, it was shown that: 1) whereas VGlut1 binds to Endophilin, VGlut2 directly interacts with the adaptor complex AP2. 2) The interaction of VGlut1 and Endophilin is regulated in a manner that depends on the time of the day. 3) In transfected PC12 cells, VGlut2 functions more as a sodium-phosphate transporter within the plasma membrane than as a vesicular glutamate transporter.
