EBAG9 ist als Negativregulator am Transport zytotoxischer Effektormoleküle zu den sekretorischen Lysosomen in CD8+-T-Zellen beteiligt. Eine Defizienz an EBAG9 führt zu einer verbesserten zytolytischen Funktion der CD8+-T-Lymphozyten gegen Suspensionszellen bzw. hämatopoetische Zellen in vitro und in vivo. Die IFN-γ Sekretion bleibt hingegen unbeeinflusst. Es wurde im E.G-7/OT-I Modell untersucht, ob durch EBAG9-Defizienz in T-Lymphozyten gegenüber Wt T-Zellen im Rahmen adoptiver T-Zelltherapie eine verbesserte Elimination solider Tumore erreicht werden kann. Nach subkutaner Applikation von E.G-7 Zellen kam es in C57BL/6 Wt und EBAG9-/- Empfängermäusen gleichermaßen zum Auswachsen solider Tumore. Wurden die Tiere zuvor gegen das dominante Peptidantigen der Tumorzellen, SIINFEKL, immunisiert, so kam es in beiden Gruppen zur erfolgreichen Abstoßung der Tumorzellen. EBAG9-/- Mäuse sind wie Wt Tiere in der Lage, ein funktionierendes immunologisches Gedächtnis auszubilden. Beim adoptiven Transfer von voraktivierten 1x106 Wt bzw. EBAG9-/- OT-I zytotoxischen T-Zellen in tumortragende C57BL/6 Tiere konnte bezüglich Tumorprogression und Überleben kein statistisch signifikanter Unterschied festgestellt werden. In beiden Gruppen kam es nach initial gutem Therapieansprechen zu Rezidiven und erneuter Tumorprogression. Dennoch ließ sich ein Trend zugunsten der EBAG9-deletierten OT-I T-Zellen nachweisen. Dieser wurde ersichtlich durch rascheres erreichen der Remissionen sowie eine stärkere Reduktion des mittleren Tumorvolumens nach Transfer verglichen mit Wt Tieren. Obwohl die direkte Granzym und Perforin vermittelte Lyse von Tumorzellen durch CTLs einen postulierten Mechanismus zur Kontrolle solider Tumore durch das Immunsystem darstellt, konnte trotz nachgewiesener stärkerer zytolytischer Kapazität gegen hämatopoetische Zielzellen kein statistisch messbarer relevanter Vorteil von EBAG9-/- CTLs gegenüber Wt CTLs im soliden E.G-7/OT-I Tumormodell festgestellt werden. Dies deutet darauf hin, dass andere Mechanismen, wie z.B. die Sekretion von IFN-γ und die dadurch vermittelte Hemmung der Angiogenese im Tumorgewebe, im Kontext solider Tumore eine wichtigere Rolle spielen könnten. Auch ein funktioneller Antagonismus von TGF-β gegenüber der Defizienz an EBAG9 kommt als Ursache in Frage. Zur Entwicklung eines technologischen Ansatzes mit dem Ziel, eine verminderte Expression von EBAG9 auch in adoptiv zu transferierenden T-Zellen ohne genetische Defizienz zu ermöglichen, wurden für EBAG9 retrovirale Transduktionen von shRNAs in primären Maus T-Lymphozyten durchgeführt. Anschließend erfolgte eine Anreicherung der transduzierten Zellen durch Zellsortingverfahren sowie eine Analyse mittels Westernblot. Dabei konnte durch die retrovirale Transduktion eine Herunterregulation von EBAG9 in primären T-Zellen um bis zu 42.4% erreicht werden. Dies demonstriert die prinzipielle Eignung dieses Verfahrens für die therapeutische Modulation der Effektorstärke bei adoptiven T-Zelltransfers.","CD8+ T-cells promote tumor rejection via direct cellular cytotoxicity and IFN-γ mediated effects on neoplastic and stromal cells upon antigen-specific activation. The deficiency of EBAG9, a molecule involved in intracellular protein trafficking and the biogenesis of the lytic granules, significantly enhances the cytotoxic capacity of CD8+ T-cells against single cell suspension in vitro and in vivo. Secretion of IFN-γ on the contrary remains unaffected. To investigate, whether a lack of EBAG9 translates into a desirable advantage against solid cancer compared to EBAG9+ CD8+ T-cells the E.G-7 tumor model was used together with the OT-I TCR for adoptive T-cell therapy. Subcutaneous inoculation with E.G-7 cells and subsequent monitoring of tumor-growth by caliper revealed comparable tumor incidence, progression and overall survival in naive C57BL/6 EBAG9+/+ and EBAG9-/- mice. When vaccinated against the dominant tumor antigen prior to tumor cell challenge, both groups showed a comparable capacity for rejecting the inoculated E.G-7 cells. This clearly demonstrated that loss of EBAG9 does not impede the formation of an efficient immunologic memory response. In the context of adoptively transferred 1x106 OT-I EBAG9+/+ and 1x106 OT-I EBAG9-/- T-cells into tumor-bearing miece, no significant benefit with respect to tumor progression or overall survival could be observed. However, careful analysis of the acquired data demonstrated a strong trend in favour of EBAG9-deficient OT-I CD8+ T-cells especially in early stages of adoptive transfer. The EBAG9-/- OT-I transfer group showed a faster onset of remissions and stronger reduction of the mean tumor volumes compared with the respective OT-I Wt transfer group. Even though EBAG9-/- cytotoxic T-cells posses enhanced cytolytic capacity and direct Granzyme/Perforin mediatet lysis of cancer cells is a postualted mechanism for tumor cell rejection, no significant advantage for EBAG9-deficient T-cells could be detected. This result could be explained by more recent discoveries, that IFN-γ secretion by CD8+ T-cells and its capacity to inhibit neoangiogenesis within solid tumors might play a more pivotal role for tumor rejection than direct cellular toxicity. Another plausible explanation could be a functional antagonism between tumor-associated TGF-β action, which is the reduction of protein expression of the cytotoxic gene molecules on the one hand, and the EBAG9 knock-out effect with increases intracellular protein- trafficking on the other. In order of achieving EBAG9 downregulation without genetic deficiency in T-cells designated for adoptive transfer, retroviral transductions with shRNAs for EBAG9 were performed in primary mouse T-cells. Enrichment of transduced cells via fluorescence assisted cell sorting and assessment of EBAG9 expression levels by Western Blot analysis revealed a decrease of EBAG9 protein levels up to 42.4%.
